Abstract Weaning and transport represent a high stress time for calves. Preconditioning (PC) by weaning before the transport separate these stressors. The stressors generate oxidative stress, which can be reduced by mineral and vitamin supplementation (MVS) with an antioxidant capacity. Our objective was to evaluate the effect of PC and MVS on performance of steers. The experiment used a 2 × 2 factorial arrangement design, considering a 26-d PC treatment from weaning to transport to the feedlot (d 0); and injectable MVS on d -45, -26, and 0. The MVS consisted of Cu, Zn, Mn, Se, vitamin E (0.2, 0.8, 0.2, 0.1, and 1 mg/kg body weight (BW), respectively), and vitamin A (1190 IU/kg). Sixty Angus-crossbred steers (186.4 ± 27.6 kg) were randomly assigned to the four treatments (MVS+PC; N+PC; MVS+N; N+N; n = 15/treatment). Body weight (BW) was recorded on d -45, -26, 0, 8, 15, and 29. On d 0, an additional BW was taken 30 minutes after the 5-hour transportation (d 0.5). Between d 0 to 29, dry matter intake (DMI) and average daily gain (ADG) to DMI ratio (G:F) were measured. Between d -26 to 29 plasma concentrations of glucose, free fatty acids (NEFA), cortisol, insulin, total antioxidant status (TAS), and thiobarbituric acid-reactive substances (TBARS) were evaluated. Data were analyzed using the MIXED procedure of SAS with repeated measures, using treatment, time, and treatment × time as fixed effects, and steer as a random effect. Between d -26 to 0, there was an interaction of MVS × PC (P ˂ 0.01) for ADG. From d -26 to 0, N+N and N+PC had the greatest and lesser ADG, respectively. On d 0.5, no-PC steers tended to lose BW while the PC steers tended to gain BW (P = 0.09). In the period d 0 to 8, there were no differences (P ≥ 0.27) in DMI, but the PC steers had greater G:F and ADG (P < 0.01) compared with no-PC steers. Plasma NEFA concentration on d 0 was affected by MVS×PC (P < 0.01), because MVS decreased plasma NEFA concentration in no-PC steers, but it increased in the PC steers. Plasma concentrations of glucose, insulin, and cortisol did not differ among treatments (P ≥ 0.23). There was a MVS×PC interaction (P = 0.09) for TAS on d 0; N+N had the greatest and MVS+N had the lowest TAS concentrations. In conclusion, a 26-d PC decreased steers BW compared with no-PC steers. The BW loss during preconditioning was not recovered 29-d after feedlot entry. Despite this BW loss, MVS treatment decreased BW loss in the steers allocated to PC treatment on the day of transport.

Abstract The objective was to evaluate the influence of melatonin supplementation on bovine testis biometrics, thermoregulation, and blood flow in two age groups. Mature and peri-pubertal bulls received four subdermal 24 mg melatonin implants (MEL; n = 5 and n = 6, respectively) or no implants (CON; n = 5 and n = 6, respectively) every 30 ± 4 d for 120 ± 5 d. Body weight, scrotal circumference (SC), testes temperature (average temperature of left and right testes as quantified with thermal imaging), and total testicular artery blood flow (TBF; total blood flow to left and right testes as measured with Doppler ultrasonography) were evaluated on d 60, 90, and 120 ± 4. On d 120, peri-pubertal bulls were castrated and total testes weight (TTW) determined. Data were analyzed using MIXED procedures of SAS specific for repeated measures with treatment, time, and the interaction as fixed effects and sire (peri-pubertal only) as a random effect. On d 120, peri-pubertal bull data were analyzed using MIXED procedures of SAS with treatment as a fixed effect and sire as a random effect, and relationships of TBF, SC, TTW, testes temperature, and ambient temperature assessed using CORR procedures of SAS. Body weight, testes temperature, TBF, and TBF as a proportion of SC changed over time in peri-pubertal (P < 0.05) but not mature (P > 0.05) bulls. On d 120, TBF was increased (P = 0.05) and TBF as a proportion of TTW tended (P = 0.10) to be increased in MEL (95.48 ± 14.63 mL/min and 0.29 ± 0.03 mL/min*g, respectively) compared to CON (65.06 ± 15.10 mL/min and 0.21 ± 0.03 mL/min*g, respectively) peri-pubertal bulls. TBF was positively correlated with SC (r = 0.59; P = 0.04) and TTW (r = 0.66; P = 0.02). Ambient temperature was positively correlated with testes temperature (r = 0.78; P < 0.01). No other differences were identified (P > 0.05). Limited alterations in testis physiology were observed in melatonin-supplemented bulls.

Abstract Four hundred and eighty (PIC 337 X 1050, PIC Genus, Hendersonville, TN) pigs were used to evaluate a novel threonine source (ThrPro, CJ America Bio, Fort Dodge, IA) for nursery pigs from approximately 7 to 20 kg body weight (BW). After weaning, pigs were sorted by sex and fed a common diet for 1 wk. Upon completion of the first week, pigs were sorted into randomized complete blocks, equalized by weight, within 16 replications. Pigs were allocated to one of three dietary treatments: positive control (POS)—standard ileal digestible threonine-to-lysine ratio (SID; Thr:Lys) 0.60, negative control (NEG)—SID Thr:Lys ≤0.46, and alternative Thr source (TEST)—SID Thr:Lys 0.60. The alternative Thr source included fermentative biomass and was assumed to contain 75% Thr and a digestibility coefficient of 100% based on the manufacturer’s specifications. All other nutrients met or exceeded the NRC recommendations. Growth and intake data were analyzed as repeated measures with a compound symmetry covariance structure using the MIXED procedure in SAS 9.4 (SAS Institute Inc., Cary, NC) with pen as the experimental unit. Treatment, phase, the interaction between treatment and phase, and block were included as fixed effects in the model. Differences in total removals were tested using Fisher’s Exact Test of PROC FREQ. Results were considered significant at P ≤ 0.05 and considered a trend at P > 0.05 and P ≤ 0.10. During the first 14 d, pigs fed TEST had decreased gain-to-feed ratio (G:F; 0.77 vs. 0.80, P = 0.022) compared to POS and increased G:F (0.77 vs. 0.73, P < 0.001) compared to NEG. Over days 14–28, pigs fed TEST had similar G:F (0.71 vs. 0.70, P = 0.112) compared to POS and increased G:F (0.71 vs. 0.63, P < 0.001) compared to NEG. Overall (days 0–28), pigs fed TEST had similar average daily gain (ADG; 0.47 vs. 0.47 kg/d, P = 0.982) and G:F (0.76 vs. 0.74, P = 0.395) compared to POS and increased ADG (0.47 vs. 0.43 kg/d, P < 0.001) and G:F (0.76 vs. 0.67, P < 0.001) compared to NEG. The average daily feed intake was not significantly different across treatments for the entirety of the study. In conclusion, the replacement of crystalline L-Thr with a novel Thr source resulted in similar growth performance in nursery pigs from approximately 7 to 20 kg.

Abstract Ninety-one early-weaned (65 ± 11 d) Angus steers (92 ± 4 kg) were blocked by age to a 2 × 2 factorial examining effects of injectable vitamin C (VC) at weaning and/or prior to transport to the feedlot on antibody titers and growth performance. Injections (20 mL/steer) of VC (250 mg sodium ascorbate/mL) or saline (SAL) were given at time of weaning on d 0 (WEAN) and/or prior to a 6 hr trucking event to a feedlot on d 49 (TRANS). Steers were given booster vaccinations on d 0. Steers were weighed on d 0, 1, 14, 48, 49, 64, 106, and 107. Blood was collected (12 steers/treatment) on d 0, 1, 2, 14, 49 (pre- and post-transit), 50, and 51. Data were analyzed via Proc-Mixed of SAS (experimental unit = steer; n = 22–23/treatment) with fixed effects of block, WEAN, TRANS, and WEAN × TRANS. Plasma ascorbate concentrations for weaning (d 0, 1, and 2) and transit (d 49-pre-trucking, 49-post-trucking, 50, and 51) were analyzed as repeated measures (repeated effect = day). Plasma ascorbate concentrations were greater on d 1 and 2 for steers that received VC at weaning (VC = 19.6, SAL = 8.8 ± 1.26 µM; WEAN × day P < 0.01). Similarly, ascorbate concentrations were greater on d 49 post-trucking, 50, and 51 for steers that received VC pre-transit (TRANS × day P = 0.01). Treatments did not affect bodyweight or average daily gain throughout the trial (P > 0.32). There were no effects of treatment on serum Bovine Viral Diarrhea Virus type 2 antibody titers on d 14 or 51 (P > 0.33). An injection of VC administered to early weaned beef steers at weaning or pre-transit increases plasma ascorbate concentrations but does not improve growth performance or antibody response to vaccination booster.

To establish the effects of hydroelectric generation on the health of lake sturgeon (Acipenser fulvescens), seasonal variations in plasma nonesterified fatty acids (NEFAs) upstream and downstream from hydroelectric stations were measured over a 2-yr period. Plasma NEFA profiles were also compared up- and downstream of the stations for differences in utilization of individual NEFA species as substrates for lipid oxidation. Significantly higher levels of total plasma NEFA were found in lake sturgeon upstream (2355 ± 395.9 nmol/mL) compared with those downstream (798 ± 133.5 nmol/mL) of the generating stations during the spring. The NEFA profiles for several key fatty acid species differed significantly among seasons up- and downstream of the facilities. In particular, during spring and summer, the levels of oleic acid (18:1n9) were highest upstream of the stations and levels of a polyunsaturated fatty acid, docosahexaenoic acid (22:6n3), were higher below rather than above the stations. The differences in plasma NEFA concentration may be attributed to altered nutritional status due to the varying flow regime located downstream of the hydroelectric stations.

Abstract The purpose of this study was to evaluate the effect that intranasal oxytocin administered at abrupt weaning (day 0) had on weaning stress behaviors such as walking distance and time devoted to walking, calf body weight, and plasma non-esterified fatty acids (NEFA), β-hydroxybutyrate (β-HB), and cortisol. Twenty Simmental × Angus heifer calves were randomly assigned to one of two treatments, intranasal oxytocin (OXT; n = 10) or intranasal saline (CON; n = 10). All calves were given the respective intranasal treatment on the day of weaning (day 0), and then placed on pasture together. Calves were weighed and a blood sample was obtained on days 0, 1, 7, and 14 post-weaning. Blood samples were subsequently used to quantify plasma NEFA, β-HB, and cortisol concentrations. All calves in both treatment groups were fitted with an individual global positioning system (GPS) that recorded calf location every ten seconds for a 16 hour period on days 0, 7, and 14 to quantify and evaluate walking behaviors. There was no treatment × day effect for distance walked (P = 0.82), walking time (P = 0.80), non-walking time (P = 0.88), area utilization index (P = 0.84), calf body weight (P = 0.82), average daily gain (P = 0.54), NEFA (P = 0.22), or cortisol concentrations (P = 0.32). There was a tendency for a treatment × day effect observed for average walking speed (P = 0.09) and β-HB (P = 0.10), such that calves in the CON treatment tended to have lesser average walking speeds on day 14 and tended to have greater β-HB concentration after weaning. There was a treatment effect (P = 0.02) for NEFA concentrations, with the CON calves having a greater plasma NEFA concentration throughout the study compared with OXT calves. These data imply that OXT calves may have had differing metabolic responses immediately after weaning that could have altered the mobilization of NEFA, but this change was not substantial enough to impact body weights or walking behaviors.

Abstract Two treatments were evaluated in heifers to determine the effects of a yeast supplement on immune and metabolic responses to a combined (tandem viral-bacterial) respiratory disease challenge. Thirty-two beef heifers (325 ± 20.1 kg BW) were selected from a larger population previously assigned to one of two treatments: Control (CON), receiving no yeast supplement in the diet, or yeast (YST), CON diet plus a combination live yeast (2.5 g·heifer−1·d−1) and yeast cell wall (2.5 g·heifer−1·d−1) supplement (Phileo Lesaffre Animal Care, Milwaukee, WI). Heifers were maintained on treatments for 31 d prior to the challenge. On day −3 all heifers were fitted with an indwelling vaginal temperature recording device, received an intranasal challenge with 2 × 108 plaque-forming units of bovine herpesvirus-1 (BHV-1), and placed in outdoor pens. On day 0, all heifers were fitted with an indwelling jugular catheter, challenged intratracheally with an average dose of 3.0 × 107 colony-forming units (cfu) of Mannheimia haemolytica in 100 mL media, and were transferred into individual stanchions in an enclosed, environmentally controlled barn. Whole blood samples were collected at −72 h and at 0, 2, 4, 6, 8, 12, 24, 36, 48, 60, and 72 h (hematology) and at 0, 1, 2, 3, 4, 5, 6, 7, 8, 12, 24, 36, 48, 60, and 72 h (serum isolation) relative to M. haemolytica challenge (0 h). Data were analyzed using the MIXED procedure of SAS specific for repeated measures with fixed effects of treatment, time, and the treatment × time interaction. Vaginal temperature and cortisol concentrations were similar between treatments (P ≥ 0.39). Although total leukocyte count following BHV-1 challenge was similar between treatments (P = 0.21), there was a tendency (P = 0.07) for CON heifers to have greater neutrophil counts than YST heifers. Serum haptoglobin concentration was similar between treatments (P = 0.13). Heifers in the YST treatment had similar serum glucose concentrations (P = 0.25) and decreased serum concentrations of urea nitrogen compared to CON (P = 0.03). Dietary treatment did not affect serum nonesterified fatty acid concentrations (P = 0.37). Nasal lesion score severity (0–4) tended (P = 0.07) to be decreased in YST compared to CON (2.5 vs. 3.19 ± 0.26), while water intake tended to be increased in YST (P = 0.06). Feeding a yeast supplement had little effect on the acute-phase response but improved metabolic outcomes in heifers during a respiratory disease challenge.

Abstract Castration is a routine procedure performed on beef and dairy operations in the U.S. All methods of castration cause behavioral, physiologic, and neuroendocrine changes associated with pain. The American Veterinary Medical Association and the American Association of Bovine Practitioners recommend that anesthesia and analgesia be administered at castration. The objective of this study was to evaluate the effectiveness of bupivacaine liposome suspension a novel, long-acting, local anesthetic administered as a nerve block at castration. The authors chose to investigate this novel formulation as an alternative to current industry standards using lidocaine nerve blocks alone, or in combination with meloxicam. Thirty male Holstein calves, 16-20 weeks of age, were enrolled and randomly assigned to 1 of 4 treatment groups prior to surgical castration: 1) bupivacaine liposome suspension block + oral placebo (BUP); 2) lidocaine block + oral placebo (LID); 3) lidocaine block + oral meloxicam (1 mg/kg) (LID + MEL); and 4) saline block + oral placebo (CON). Biomarkers were collected at -24 h and from 0 to 120 hours post-castration and included infrared thermography, pressure mat gait analysis, chute defense and behavior scoring (pain and activity), and blood sampling for serum cortisol and prostaglandin E2 metabolites (PGEM). Responses were analyzed using repeated measures, with calf nested in treatment as a random effect, and treatment, time, and their interaction designated as fixed effects. Results from pressure mat gait analysis show the CON had a shorter front limb stance time from baseline (-8.73%; 95% CI: -24.84 to 7.37%) compared to BUP and LID + MEL (> 5.70%; 95% CI: -22.91 to 23.79%) (P < 0.03). The CON tended to have an increase in front limb force from baseline (6.31%; 95% CI: -1.79 to 14.41%) compared to BUP, LID, and LID + MEL (< -5.06%; 95% CI: -14.22 to 0.95%) (P < 0.04). The CON displayed higher counts of hunched standing (2.00; 95% CI: 1.68 to 2.32) compared to LID + MEL (1.43; 95% CI: 1.13 to 1.72) (P = 0.05). The CON had higher cortisol concentrations at 24 h (7.70 ng/mL; 95% CI: 1.52 to 13.87 ng/mL) relative to BUP (3.11 ng/mL; 95% CI: -2.56 to 8.79 ng/mL) (P = 0.002). At 4 and 24 h, LID + MEL had lower PGEM concentrations from baseline (-32.42% and -47.84%; 95% CI: -78.45 to -1.80%) compared with CON (27.86% and 47.63%; 95% CI: 7.49 to 82.98%) (P < 0.02). Administration of bupivacaine liposome suspension as a local anesthetic block at the time of castration was as effective at controlling pain as a multi-modal approach of lidocaine and meloxicam.

Abstract This study examined the effects of injectable vitamin C (VC) before transport and duration of transit on feedlot performance, inflammation, and muscle fatigue in cattle. One hundred thirty-one, Angus-cross steers (409 ± 4 kg) were stratified by bodyweight (BW) to a 2 × 2 factorial of intramuscular injection (INJ; 20 mL/steer): VC (250 mg sodium ascorbate/mL) or saline (SAL) and road transit duration (DUR): 18 (18; 1,770 km) or 8 h (8; 727 km). On d 0, steers were weighed and received INJ of SAL or VC immediately before transport. Upon return (d 1), BW and blood were collected before steers returned to pens with GrowSafe bunks. Steers were weighed on d 0, 1, 7, 15, 30, 31, 54, and 55. Data were analyzed via ProcMixed of SAS (experimental unit = steer; 32–34 steers/treatment) with fixed effects of INJ, DUR, and the interaction. Blood was collected on d -5, 1, 2, and 3 (9 steers/treatment); blood parameters were analyzed as repeated measures. Average daily gain (ADG) and BW were greater on d 7 and 15 for SAL-18 compared to all other treatments (INJ × DUR, P < 0.01). Final BW, overall ADG, and gain:feed were greater for 18 than 8 (P < 0.01). Injection did not affect BW (P > 0.13) but VC decreased overall dry matter intake compared to SAL (P = 0.03). Steers transported for 18 h had greater serum lactate, haptoglobin, and non-esterified fatty acid concentrations on d 1 compared to steers transported for 8 h (DUR × DAY, P < 0.01). Day 1 plasma ascorbate concentrations were greater for VC and returned to baseline concentrations by d 2 (INJ × DAY, P < 0.01). In contrast to previous work, VC did not improve post-transit performance; however, longer transit duration increased indicators of muscle fatigue and inflammation.

Urinary catecholamine excretion, blood glucose, and plasma nonesterified fatty acids (NEFA) concentration were studied in five full-term, seven premature, and 22 small-for-date (SFD) babies (16 at term and six prematurely born) during the first 8 to 10 days of life. Episodes of hypoglycemia (arbitrarily designed as < 40 mg/100 ml for full term and < 30 mg/100 ml for premature infants) were observed between the second and fifth day of age in 12 SFD babies. During their low blood sugar period, these infants exhibited a significantly higher catecholamine excretion (three- to fourfold for norepinephrine and five- and sixfold for epinephrine) when compared to full term, premature, and "nonhypoglycemic" SFD babies. All SFD babies during the first 2 days of life showed a significantly higher plasma NEFA concentration than the other groups of neonates; among the former, the hypoglycemic ones had a tendency to maintain for a longer period a higher NEFA concentration than the nonhypoglycemic ones. These findings show that SFD babies are capable of reacting to an hypoglycemic stress by mobilizing their fat stores and by releasing catecholamine; therefore, adrenal medullary unresponsiveness cannot be considered as a cause of their transient neonatal hypoglycemia.

Abstract This experiment was designed to study the effect of days on feed and an aggressive implant strategy on Jersey steer growth performance. Steers (n = 30; start of trial body weight (BW) 183 ± 43 kg) were randomly assigned to one of two treatments; negative control (CON) or implanted with Revalor 200 every 70 d (REV; d 0, d 70, d 140, d 210, d 280, d 350) for a total of 6 implants. Steers were weighed every 35 d and daily feed delivery was recorded daily. Data for dry matter intake (DMI) and gain to feed ratio (G:F) were analyzed via a mixed model; the fixed effects were day and treatment. Live growth BW and average daily gain (ADG) used day as the repeated measure and animal as the subject in a repeated measures analysis. Change in BW increased linearly (P < 0.01); treatments differed in BW from d 280 through d 350 (P < 0.01). Average daily gain decreased (P < 0.01) linearly and differed (P < 0.01) at d 70, d 140, d 280, and d 350, but not (P ≥ 0.10) at d 210 or d 420. Dry matter intake was greater (P < 0.05) for REV steers (7.6 kg/d) than CON (6.8 kg/d). Gain to feed ratio did not differ (P ≥ 0.78) between CON steers (0.13 kg/kg dry matter (DM)) and REV (0.14 kg/kg DM) steers, yet G:F differed (P < 0.01) amongst periods (d 70 = 0.21 kg/kg, d 140 = 0.16 kg/kg, d 210 = 0.15 kg/kg, d 280 = 0.13 kg/kg, d 350 = 0.08 kg/kg, d 420 = 0.08 kg/kg). Aggressively implanting Jersey steers improved growth performance by 9.6%, 13.2%, 11.3%, and 7.7% for BW, ADG, DMI, and G:F, respectively.
