Abstract Temperature is one of the most important abiotic factors regulating development and biological processes in ectotherms. By 2050, climate change may result in temperature increases of 2.1–3.4°C in Manitoba, Canada. Lake sturgeon, Acipenser fulvescens, from both northern and southern populations in Manitoba were acclimated to 16, 20 and 24°C for 30 days, after which critical thermal maximum (CTmax) trials were conducted to investigate their thermal plasticity. We also examined the effects of temperature on morphological and physiological indices. Acclimation temperature significantly influenced the CTmax, body mass, hepatosomatic index, metabolic rate and the mRNA expression of transcripts involved in the cellular response to heat shock and hypoxia (HSP70, HSP90a, HSP90b, HIF-1α) in the gill of lake sturgeon. Population significantly affected the above phenotypes, as well as the mRNA expression of Na+/K+ ATPase-α1 and the hepatic glutathione peroxidase enzyme activity. The southern population had an average CTmax that was 0.71 and 0.45°C higher than the northern population at 20 and 24°C, respectively. Immediately following CTmax trials, mRNA expression of HSP90a and HIF-1α was positively correlated with individual CTmax of lake sturgeon across acclimation treatments and populations (r = 0.7, r = 0.62, respectively; P < 0.0001). Lake sturgeon acclimated to 20 and 24°C had decreased hepatosomatic indices (93 and 244% reduction, respectively; P < 0.0001) and metabolic suppression (27.7 and 42.1% reduction, respectively; P < 0.05) when compared to sturgeon acclimated to 16°C, regardless of population. Glutathione peroxidase activity and mRNA expression Na+/K+ ATPase-α1 were elevated in the northern relative to the southern population. Acclimation to 24°C also induced mortality in both populations when compared to sturgeon acclimated to 16 and 20°C. Thus, increased temperatures have wide-ranging population-specific physiological consequences for lake sturgeon across biological levels of organization.

The impact of early rearing environment on phenotypic development in teleosts has been reasonably well-documented, but only recently has attention been paid to sturgeon raised for conservation purposes. In the present study, we hypothesized that rearing environment will result in the development of distinct metabolic phenotypes in age-0 lake sturgeon (Acipenser fulvescens) and that these phenotypes will drive differential survival rates during a simulated overwintering event. Lake sturgeon gametes were fertilized and raised in one of three different environments: 16 °C + 100% dissolved oxygen (DO), 14 °C + 100% DO, and 16 °C + 80% DO, each with or without substrate. We measured standard metabolic rate, forced maximum metabolic rate, metabolic scope, energy density, hepatosomatic index, Fulton’s condition factor, and enzyme activities associated with ATP production. Our results suggest that subtle changes in abiotic environments during early life history result in the development of distinct metabolic phenotypes during the first year of life. These have important implications for survival of age-0 lake sturgeon when stocked in the fall of their first year.

Understanding biotic and abiotic factors that influence spatial distribution patterns, condition factor, and growth of lotic fish species within river impoundments is essential for the development of effective management and conservation strategies. This study aimed to compare relative abundance, condition factor, and growth rate of juvenile lake sturgeon (Acipenser fulvescens Rafinesque, 1817) among eight sections of a 41 km long impoundment of the Winnipeg River, Manitoba, Canada. Relative abundance of juvenile lake sturgeon, as measured by catch per unit effort (CPUE), was 3–6 times greater in the two farthest upstream sections when compared with the five farthest downstream sections. Growth in length was slowest for individuals captured in the two farthest upstream sections, moderate in the third section, and highest in the fourth section, with individuals from the fourth section attaining lengths approximately double those from the two farthest upstream sections by age 6. Condition factor varied among sections of the impoundment in a pattern similar to that observed for growth. Given similarities in many environmental factors such as water temperature and water chemistry among sections of this study area, our results provide important insight into how abiotic and biotic factors, combined with behavioural characteristics of this species, may influence distribution patterns and growth of juvenile lake sturgeon within river impoundments.

In fishes, environmental ion availability can have substantial effects on growth and development. This study examined the development of Lake Sturgeon in response to the varying environmental ion availability that they experience as part of a conservation stocking program. We reared sturgeon in natural water from the Coosa River, which had higher concentrations of Mg2+, Na+, and Zn2+ than standard hatchery conditions, while [Ca2+] at the Warm Springs National Fish Hatchery was 2× higher than in the Coosa River. Eggs were hatched in each water type and the larvae were sampled at time points before and after yolk absorption during the first 8 weeks of development. Total length and weight in WSNFH larvae were significantly higher than larvae in Coosa River water starting at 8 dph, indicating that growth was dependent on the different environmental ion levels. Concentrations of the ions of interest were also determined for whole-body acid digests of the exposed Lake Sturgeon. We found that Lake Sturgeon reared in Coosa River water had significantly higher magnesium and zinc than Lake Sturgeon reared in WSNFH water (p < 0.05), while calcium was significantly higher in WSNFH than Coosa River water. This difference shows that different environmental ion concentrations also impact the overall development of larval Lake Sturgeon.

Abstract Background Glioblastoma multiforme (GBM) is the leading cause of death among adult brain cancer patients. Glutathione peroxidase 2 (GPX2), as a factor in oxidative stress, plays an important role in carcinogenesis. However, its role in GBM has not been well established. The study aimed to investigate the clinical significance of GPX2 with GBM prognosis. Methods Data of GBM and healthy individuals were retrospectively collected from oncomine, cancer cell line encyclopedia (CCLE), gene expression profiling interactive analysis (GEPIA), UALCAN, and Human Protein Atlas. GPX2 mRNA expression was first assessed across various cancer types in oncomine and cancer cell lines from CCLE. The mRNA expression of GPX2 was compared between normal and GBM tissues using GEPIA (normal = 207; GBM = 163) and UALCAN (normal = 5; GBM = 156). The GPX2 methylation was analyzed using data from UALCAN (normal = 2; GBM = 140). The prognostic value of GPX2 in GBM was explored in GEPIA and UALCAN using Kaplan–Meier method. STRING database was used to construct protein–protein interaction (PPI) network and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway. Statistical significance was set as <0.05. Results The current study revealed no significant differences in GPX2 expression between normal and GBM from GEPIA data (P > 0.05) and UALCAN (P = 0.257). Patients with higher GPX2 intended to have a poorer prognosis (P = 0.0089). The KEGG pathways found that chemokine-signaling pathway were the more preferred. Conclusions The findings demonstrated that GPX2 might be a potential diagnosis and prognostic indicator for GBM. Chemokine-signaling pathway may be involved in GPX2 function.

To establish the effects of hydroelectric generation on the health of lake sturgeon (Acipenser fulvescens), seasonal variations in plasma nonesterified fatty acids (NEFAs) upstream and downstream from hydroelectric stations were measured over a 2-yr period. Plasma NEFA profiles were also compared up- and downstream of the stations for differences in utilization of individual NEFA species as substrates for lipid oxidation. Significantly higher levels of total plasma NEFA were found in lake sturgeon upstream (2355 ± 395.9 nmol/mL) compared with those downstream (798 ± 133.5 nmol/mL) of the generating stations during the spring. The NEFA profiles for several key fatty acid species differed significantly among seasons up- and downstream of the facilities. In particular, during spring and summer, the levels of oleic acid (18:1n9) were highest upstream of the stations and levels of a polyunsaturated fatty acid, docosahexaenoic acid (22:6n3), were higher below rather than above the stations. The differences in plasma NEFA concentration may be attributed to altered nutritional status due to the varying flow regime located downstream of the hydroelectric stations.

Creatine monohydrate (CrM) supplementation has been shown to increase fat-free mass and muscle power output possibly via cell swelling. Little is known about the cellular response to CrM. We investigated the effect of short-term CrM supplementation on global and targeted mRNA expression and protein content in human skeletal muscle. In a randomized, placebo-controlled, crossover, double-blind design, 12 young, healthy, nonobese men were supplemented with either a placebo (PL) or CrM (loading phase, 20 g/day × 3 days; maintenance phase, 5 g/day × 7 days) for 10 days. Following a 28-day washout period, subjects were put on the alternate supplementation for 10 days. Muscle biopsies of the vastus lateralis were obtained and were assessed for mRNA expression (cDNA microarrays + real-time PCR) and protein content (Kinetworks KPKS 1.0 Protein Kinase screen). CrM supplementation significantly increased fat-free mass, total body water, and body weight of the participants ( P < 0.05). Also, CrM supplementation significantly upregulated (1.3- to 5.0-fold) the mRNA content of genes and protein content of kinases involved in osmosensing and signal transduction, cytoskeleton remodeling, protein and glycogen synthesis regulation, satellite cell proliferation and differentiation, DNA replication and repair, RNA transcription control, and cell survival. We are the first to report this large-scale gene expression in the skeletal muscle with short-term CrM supplementation, a response that suggests changes in cellular osmolarity.

Historically, many population genetics studies have utilized microsatellite markers sampled at random from the genome and presumed to be selectively neutral. Recent studies, however, have shown that microsatellites can occur in transcribed regions, where they are more likely to be under selection. In this study, we mined microsatellites from transcriptomes generated by 454-pyrosequencing for three vertebrate species: lake sturgeon (Acipenser fulvescens), tiger salamander (Ambystoma tigrinum), and kangaroo rat (Dipodomys spectabilis). We evaluated (i) the occurrence of microsatellites across species; (ii) whether particular gene ontology terms were over-represented in genes that contained microsatellites; (iii) whether repeat motifs were located in untranslated regions or coding sequences of genes; and (iv) in silico polymorphism. Microsatellites were less common in tiger salamanders than in either lake sturgeon or kangaroo rats. Across libraries, trinucleotides were found more frequently than any other motif type, presumably because they do not cause frameshift mutations. By evaluating variation across reads assembled to a given contig, we were able to identify repeat motifs likely to be polymorphic. Our study represents one of the first comparative data sets on the distribution of vertebrate microsatellites within expressed genes. Our results reinforce the idea that microsatellites do not always occur in noncoding DNA, but commonly occur in expressed genes.
