Simultaneous Determination of Prostaglandin and Hormones in Excreta of Trogopterus xanthipes

Abstract The excreta of Trogopterus xanthipes (also called Wulingzhi in Chinese, WLZ) is a well-known traditional Chinese medicine used for the treatment of irregular menstruation in clinic. Few reports are available on the chemical profiling of WLZ. In this work, qualitative and quantitative analyses of endogenous prostaglandin and hormones in WLZ were performed using UHPLC-orbitrap-MSn. In total, 48 compounds were identified in urine of T. xanthipes. Furthermore, the contents of four target compounds were simultaneously quantitated in 20 batches of samples by UPLC-MS/MS. The quantitative method showed a good linear correlation (R > 0.995) in a wide range for each compound. The method had a high sensitivity with LOD (0.5–1.0 ng/mL) and LOQ (1.0–2.5 ng/mL). The intra- and inter-day precisions were < 9.17 (RSD %), and repeatability and stability were < 6.14 (RSD %). The recovery of the analytes varied between 85.8% and 97.3% at three different concentrations. The present integrated qualitative and quantitative assessment of WLZ provides an evaluation strategy to assess the constituent in traditional Chinese medicine.

Eclipta prostrataL. is one of the Chinese medicinal tonics which are usually used for treating loose teeth, dizziness, tinnitus, hemoptysis, hematuria, and uterine bleeding. However, quality control of this herbal medicine has been not satisfactory. This study reported its qualitative and quantitative analyses based on LC/MS method. UHPLC-DAD-Q-TOF-MS fingerprinting and MS fragmentation cleavage pathway were investigated for qualitative analysis. Furthermore, a method for simultaneous quantitative determination of nine compounds, luteolin 7-O-β-D-glucopyranoside, ecliptasaponin C, luteolin, eclalbasaponin IV, apigenin, ecliptasaponin A, echinocystic acid 28-O-β-D-glucopyranoside, echinocystic acid, and 3-oxo-16α-hydroxy-olean-12-en-28-oic acid inE. prostrata, was established. The method was validated for samples ofE. prostratafrom different habitats. The results showed good linear correlation, precision, accuracy, and repeatability that could be used for contents determination of the nine compounds inE. prostratafrom different habitats.

In the present review, the literature data on the chemical constituents and biological investigations of the genus Pedicularis are summarized. Some species of Pedicularis have been widely applied in traditional Chinese medicine. A wide range of chemical components including iridoid glycosides, phenylpropanoid glycosides (PhGs), lignans glycosides, flavonoids, alkaloids and other compounds have been isolated and identified from the genus Pedicularis. In vitro and in vivo studies indicated some monomer compounds and extracts from the genus Pedicularis have been found to possess antitumor, hepatoprotective, anti-oxidative, antihaemolysis, antibacterial activity, fatigue relief of skeletal muscle, nootropic effect and other activities.

CuxO-NiO nanocomposite film for the non-enzymatic determination of glucose was prepared by the novel modifying method. At first, anodized Cu electrode was kept in a mixture solution of CuSO4, NiSO4 and H2SO4 for 15 minutes. Then, a cathodization process with a step potential of -6 V in a mixture solution of CuSO4 and NiSO4 was initiated, generating formation of porous Cu-Ni film on the bare Cu electrode by electrodeposition assisted by the release of hydrogen bubbles acting as soft templates. Optimized conditions were determined by the experimental design software for electrodeposition process. Afterward, Cu-Ni modified electrode was scanned by cyclic voltammetry (CV) method in NaOH solution to convert Cu and Ni nanoparticles to the nano-scaled CuxO-NiO film. The electrocatalytic behavior of the novel CuxO-NiO film toward glucose oxidation was studied by CV and chronoamperometry (CHA) techniques. The calibration curve of glucose was found linear in a wide range of 0.04–5.76 mM, with a low limit of detection (LOD) of 7.3 µM (S/N = 3) and high sensitivity (1.38 mA mM-1 cm-2). The sensor showed high selectivity against some usual interfering species and high stability (loss of only 6.3 % of its performance over one month). The prepared CuxO-NiO nanofilm based sensor was successfully applied for monitoring glucose in human blood serum and urine samples.

The X-ray fluorescence spectral qualitative and quantitative analyses were performed for the elemental composition of 4 marks of bullets for pneumatic weapon of home and foreign production, and also for imitators of common clothing materials camouflage fabric and cotton knitwear. It is determined that lead bullets for pneumatic weapons are capable to introduce some elements of their composition, which are inherent in one or another mark of bullets, to the area of damage on certain clothing materials, in particular, camouflage fabric and cotton knitwear. This expands the possibilities of forensic medical examination when dealing with a problem of determining the type and mark of a bullet or of a small group of bullets similar in their elemental composition, due to which the pneumatical shot injuries of examined objects took place.
