When the abaxial epidermis was peeled from 5 to 6 day old oat primary leaves, and 3 cm segments were floated on radioactive phenylalanine or cinnamic acid solutions, more than 90 per cent of the radioactivity was incorporated within 3 to 7 h depending on the developmental stage of the leaf. C-glycosylflavones were labelled within 15 min and radioactivity in these compounds increased for several hours. Pulse labelling and pulse chase experiments with either phenylalanine or cinnamic acid, unequivocally demonstrate that oat flavones are stable end products of metabolism. However, this procedure does not distinguish between sequential biosynthesis of various flavones and their interconversion. Cinnamic acid was more efficiently (ca. 20 x) converted into oat leaf flavones than was phenylalanine, when the precursor was fed to leaf pieces, and flavones recovered from mesophyll protoplasts. Different labelling patterns were obtained with whole leaf segments and protoplasts which apparently reflect differences in tissue specific flavone biosynthesis of mesophyll and epidermis. Isolated mesophyll protoplasts incubated with [14C]cinnamic acid synthesize 14C-labelled flavones characteristic of the mesophyll, as well as several unidentified phenylpropanoid derivatives not found in the intact tissue. Data suggest that photosynthetically active mesophyll cells are a main site of tissue specific flavone biosynthesis

Abstract: Diploon is a monospecific genus represented by Diploon cuspidatum, an arboreal species that has morphological characteristics distinct from those of other Sapotaceae species. In this study, Diploon cuspidatum leaves were characterized morphoanatomically in order to reveal additional diagnostic characters of their external morphology of the genus. The Diploon petiole presents shape and arrangement of the vascular system flat-convex, occasionally with one or two accessory bundles, many laticifers, and many prismatic crystals. The midrib is biconvex with a U-shaped cuticle on the abaxial side, and laticifers are associated with the vascular tissues. Mesophyll is dorsiventral, palisade parenchyma has two cell layers, T- and Y-shaped malpighiaceous trichomes are on the abaxial epidermis with a small stalk cell and long arm. The venation pattern is brochidodromous. Intersecondary veins run parallel to the secondary veins, and quaternary veins branch freely. Higher order veins are not present. Morphoanatomical analysis revealed important characteristics that reveal a set of structures common to Sapotaceae, in addition to characters that are important for the recognition and identification of D. cuspidatum.

Low K+ in Paphiopedilum leeanum leaf epidermis: implications for stomatal functioning

Abaxial epidermal strips from leaves of Paphiopedilum leeanum were analyzed via sodium cobaltinitrite staining and atomic absorption spectrophotometry for the presence and location of potassium. On a dry weight basis K content of the abaxial epidermis was found to be 103 times less than has been reported in other species, and unlike other species no localization of K+ in guard cells of open stomata could be detected via the sodium cobaltinitrite stain for potassium.Flame photometric analysis of the mesophyll indicated that it contained normal amounts of K+ (about 1.87% on a dry weight basis). Analysis showed that the K+ content of the abaxial epidermis (0.032%) was considerably less than that of the mesophyll, a situation unlike previous reports for other species in which the epidermal concentration was found to be greater than the mesophyll. A process for exclusion of K+ from the abaxial epidermis is suggested, as is the lack of involvement of K+ as the major osmoticum in the stomatal mechanism of this species.

CARACTERES EPIDÉRMICOS FOLIARES DE PLANTAS ORNAMENTALES, TÓXICAS PARA ANIMALES DOMÉSTICOS. LEAF EPIDERMAL CHARACTERS OF ORNAMENTAL PLANTS, TOXIC FOR DOMESTIC ANIMALS. Patricia Nasca de Zamora DOI: https://doi.org/10.33017/RevECIPeru2009.0003/ RESUMEN El tejido epidérmico de los vegetales presenta un diseño característico para cada especie, de manera que muchas plantas pueden reconocerse a través de su estudio microscópico. Además, este tejido resiste el pasaje por el tracto digestivo de los animales, permaneciendo prácticamente inalterable. Basados en este hecho, los investigadores Baungartder y Martin (1939) idearon la técnica microhistológica, que consiste en estudiar la composición botánica de la dieta de animales herbívoros a través del análisis microscópico de fragmentos de epidermis vegetales encontrados en la materia fecal de los mismos. A lo largo de los años, el uso de esta técnica se extendió ampliamente, adaptándose a diferentes objetivos, tanto ecológicos como económicos. El objetivo del presente trabajo, que se llevó a cabo en la provincia de Tucumán, República Argentina, es describir las epidermis foliares de cuatro especies ornamentales tóxicas, a fin de aportar datos para la confección de una clave dicotómica de identificación de las mismas, para ser usada como elemento de diagnóstico en medicina veterinaria. Se extrajeron en laboratorio - con diversas técnicas - epidermis foliares de Evonymus japonicus, Hedera helix, Ficus benjamina y Vinca difformis; especies ornamentales citadas como tóxicas en la bibliografía veterinaria y cultivadas habitualmente en jardines particulares y espacios públicos de la Provincia. Se confeccionaron preparados microscópicos permanentes, se tomaron registros fotográficos y se describieron las características epidérmicas de cada especie, teniendo en cuenta la forma de las células, la presencia o ausencia de estomas, presencia de tricomas, tipos de estomas, tipos de tricomas, etc. Hedera helix y Vinca difformis presentan células epidérmicas de contorno lobulado, mientras que las células epidérmicas de Evonymus japonicus. y Ficus benjamina son isodiamétricas y poligonales. Las cuatro especies descriptas presentan estomas sólo en la zona internerval de la cara abaxial de sus hojas. Se encontraron tricomas simples solamente en la zona nerval de la epidermis adaxial de Vinca difformis, las demás epidermis son completamente glabras. Los caracteres encontrados en las especies estudiadas y volcados en las descripciones, constituyen un aporte de utilidad para la confección de una clave dicotómica de indentificación de estas plantas tóxicas a través de sus epidermis. ABSTRACT The epidermal tissue of the vegetables presents a typical design for every species so that many plants can be recognized across its microscopic study. In addition, it resists the passage for the digestive tract of the animals, remaining practically inalterable. Based on this fact, Baungartder and Martin (1939) designed the microhistological technique, which consists of studying botanical composition of diet of herbivorous animals across the microscopic analysis of plant´s epidermis fragments found in the fecal matter. Throughout the years, the use of this technique spread widely, adapting to different aims, both ecological and economic. The aim of the present work, realized in Tucumán's province, Argentina, is to characterize epidermis of ornamental, toxic plants for domestic animals, in order to be used as element of diagnosis in veterinary medicine. Epidermis of Evonymus japonicus, Hedera helix, Ficus benjamina and Vinca difformis were extracted in laboratory. All of them are ornamental species, mentioned like toxic in the bibliography veterinary and cultivated habitually in particular gardens and public spaces of the Province. They were made prepared microscopic permanent, photographic records took and there were described the epidermal characteristics of every species, bearing in mind the form of the cells, the presence or absence of stomata and trichomes, and types of stomata and trichomes. Hedera helix and Vinca difformis present lobulated epidermal cells, whereas Evonymus japonicus and Ficus benjamina ones are isodiametrics and polygonal. Four species present stomata only in the internerval zone of abaxial epidermis. They were trichomes only in the nerval zone of adaxial epidermis of Vinca difformis. The epidermal characters found in the studied species, constitute an usefull contribution for the confection of a dichotomous indentification key of these toxic plants. The understanding of the behavior of each cultivar under adverse climatic conditions is important in the choice of plants that best fit the region to be inserted. Due to the large number of cultivars available on the market it makes it difficult for producers to choose which material to plant. In view of the above, this study aimed to know the morphophysiological characteristics of coffee. The experimental design was in randomized blocks with 10 treatments, that is, arabic coffee varieties: Catuai IAC62; Catuai IAC99; Ouro IAC4397; Tupi RN IAC1669-13; Obatã IAC1669-20; Mundo Novo IAC379-24; Mundo Novo IAC 388-17-2; Mundo Novo SH3 Faz São José; Bourbon IACJ15 and Icatu IAC 2944-11 and with four replications totaling 40 plots, where each plot was composed of seven plants. The Mundo Novo IAC 388-17-2 coffee variety shows higher yield in the seventh year of cultivation. Variety of Bourbon IACJ15 coffee presented water use efficiency (EUW) which did not reflect in higher productivity. The Catuai V IAC99 arabica coffee variety stood out in the internal morphology of the leaves. The thickness of the adaxial and abaxial epidermis (TADE and TABE) and the CO2 assimilation rate (A) showed negative correlations with the productivity of processed coffee bags. The basis of differentiation between sun and shade leaves is related to different light intensities. In order to understand the adaptability of the leaves of Rhizophora mangle L., associated with different lighting conditions, leaves were collected from the upper peripheral six individuals (sun leaves) and the lower region of the same internal (shade leaves). The variables analyzed leaf thickness, palisade parenchyma, adaxial and abaxial epidermis, adaxial and abaxial cuticle, stomatal density and index. Measurements were made ??on microscope equipped with ocular micrometer. Sun leaves were lower and with more xeromorphic characteristics, such as increased thickness of the cuticle and the adaxial and abaxial epidermis. The palisade parenchyma and limbus showed up thicker than shade leaves, with no significant difference between the cuticle of the abaxial surface. It was also observed a higher frequency of stomata per mm², an average of 70/mm², while shade leaves showed 47/mm², with no differences between length and width. Differences between the sun leaves and shade leaves indicate adaptive capacity of this species to remain active at different light conditions.
