The areas in which Stromal Vascular Fraction cells (SVFs) have been used include radiotherapy based tissue damage after mastectomy, breast augmentation, calvarial defects, Crohn’s fistulas, and damaged skeletal muscle. Currently, the authors present their experience using regenerative cell therapy in breast reconstruction. The goal of this study was to evaluate the safety and efficacy of the use of Engineered Fat Graft Enhanced with Adipose-derived Stromal Vascular Fraction cells (EF-e-A) in breast reconstruction. 121 patients that were affected by the outcomes of breast oncoplastic surgery were treated with EF-e-A, comparing the results with the control group (n = 50) treated with not enhanced fat graft (EF-ne-A). The preoperative evaluation included a complete clinical examination, a photographic assessment, biopsy, magnetic resonance (MRI) of the soft tissue, and ultrasound (US). Postoperative follow-up took place at two, seven, 15, 21, 36 weeks, and then annually. In 72.8% (n = 88) of breast reconstruction treated with EF-e-A, we observed a restoration of the breast contour and an increase of 12.8 mm in the three-dimensional volume after 12 weeks, which was only observed in 27.3% (n = 33) of patients in the control group that was treated with EF-ne-A. Transplanted fat tissue reabsorption was analyzed with instrumental MRI and US. Volumetric persistence in the study group was higher (70.8%) than that in the control group (41.4%) (p < 0.0001 vs. control group). The use of EF-e-A was safe and effective in this series of treated cases.

The Impact

Recent experiments with droplets impacting macro-textured superhydrophobic surfaces revealed new regimes of bouncing with a remarkable reduction of the contact time. Here we present a comprehensive numerical study that reveals the physics behind these new bouncing regimes and quantifies the roles played by various external and internal forces. For the first time, accurate three-dimensional simulations involving realistic macro-textured surfaces are performed. After demonstrating that simulations reproduce experiments in a quantitative manner, the study is focused on analysing the flow situations beyond current experiments. We show that the experimentally observed reduction of contact time extends to higher Weber numbers, and analyse the role played by the texture density. Moreover, we report a nonlinear behaviour of the contact time with the increase of the Weber number for imperfectly coated textures, and study the impact on tilted surfaces in a wide range of Weber numbers. Finally, we present novel energy analysis techniques that elaborate and quantify the interplay between the kinetic and surface energy, and the role played by the dissipation for various Weber numbers.

Engineered skin that can facilitate tissue repair has been a great advance in the field of wound healing. A well-designed dressing material together with active biological cues such as cells or growth factors can overcome the limitation of using auto-grafts from patients. Recently, many studies showed that human adipose-derived stem cells (hASCs) can be used to promote wound healing and skin tissue engineering. hASCs have already been widely applied for clinical trials. hASCs can be harvested abundantly because they can be easily isolated from fat tissue known as the stromal vascular fraction (SVF). On the other hand, increasing studies have proven that cells from spheroids can better simulate the biological microenvironment and can enhance the expression of stemness markers. However, a three-dimensional (3D) scaffold that can harbor implanted cells and can serve as a skin-repaired substitute still suffers from deficiency. In this study, we applied a gelatin/microbial transglutaminase (mTG) hydrogel to encapsulate hASC spheroids to evaluate the performance of 3D cells on skin wound healing. The results showed that the hydrogel is not toxic to the wound and that cell spheroids have significantly improved wound healing compared to cell suspension encapsulated in the hydrogel. Additionally, a hydrogel with cell spheroids was much more effective than other groups in angiogenesis since the cell spheroid has the possibility of cell–cell signaling to promote vascular generation.

AbstractCoalescing drops spontaneously jump out of plane on a variety of biological and synthetic superhydrophobic surfaces, with potential applications ranging from self-cleaning materials to self-sustained condensers. To investigate the mechanism of self-propelled jumping, we report three-dimensional phase-field simulations of two identical spherical drops coalescing on a flat surface with a contact angle of 180°. The numerical simulations capture the spontaneous jumping process, which follows the capillary–inertial scaling. The out-of-plane directionality is shown to result from the counter-action of the substrate to the impingement of the liquid bridge between the coalescing drops. A viscous cutoff to the capillary–inertial velocity scaling is identified when the Ohnesorge number of the initial drops is around 0.1, but the corresponding viscous cutoff radius is too small to be tested experimentally. Compared to experiments on both superhydrophobic and Leidenfrost surfaces, our simulations accurately predict the nearly constant jumping velocity of around 0.2 when scaled by the capillary–inertial velocity. By comparing the simulated drop coalescence processes with and without the substrate, we attribute this low non-dimensional velocity to the substrate intercepting only a small fraction of the expanding liquid bridge.
