Abstract Background Grapevine reproductive development has direct implications on yield. It also impacts on berry and wine quality by affecting traits like seedlessness, berry and bunch size, cluster compactness and berry skin to pulp ratio. Seasonal fluctuations in yield, fruit composition and wine attributes, which are largely driven by climatic factors, are major challenges for worldwide table grape and wine industry. Accordingly, a better understanding of reproductive processes such as gamete development, fertilization, seed and fruit set is of paramount relevance for managing yield and quality. With the aim of providing new insights into this field, we searched for clones with contrasting seed content in two germplasm collections. Results We identified eight variant pairs that seemingly differ only in seed-related characteristics while showing identical genotype when tested with the GrapeReSeq_Illumina_20K_SNP_chip and several microsatellites. We performed multi-year observations on seed and fruit set deriving from different pollination treatments, with special emphasis on the pair composed by Sangiovese and its seedless variant locally named Corinto Nero. The pollen of Corinto Nero failed to germinate in vitro and gave poor berry set when used to pollinate other varieties. Most berries from both open- and cross-pollinated Corinto Nero inflorescences did not contain seeds. The genetic analysis of seedlings derived from occasional Corinto Nero normal seeds revealed that the few Corinto Nero functional gametes are mostly unreduced. Moreover, three genotypes, including Sangiovese and Corinto Nero, were unexpectedly found to develop fruits without pollen contribution and occasionally showed normal-like seeds. Five missense single nucleotide polymorphisms were identified between Corinto Nero and Sangiovese from transcriptomic data. Conclusions Our observations allowed us to attribute a seedlessness type to some variants for which it was not documented in the literature. Interestingly, the VvAGL11 mutation responsible for Sultanina stenospermocarpy was also discovered in a seedless mutant of Gouais Blanc. We suggest that Corinto Nero parthenocarpy is driven by pollen and/or embryo sac defects, and both events likely arise from meiotic anomalies. The single nucleotide polymorphisms identified between Sangiovese and Corinto Nero are suitable for testing as traceability markers for propagated material and as functional candidates for the seedless phenotype.

AbstractBryozoa is a phylum of aquatic, colonial suspension-feeders within the Lophotrochozoa. In the Phylactolaemata embryonic development occurs in an internal brood sac on the body wall accompanied by extraembryonic nutrition. Owing to previous contradictive descriptions, many aspects of their sexual reproduction require restudy. Consequently, this study analyses embryogenesis of the freshwater bryozoan Plumatella casmiana by serial sections, 3D reconstruction and transmission electron microscopy. Early embryos cleave and soon develop into blastulae with a small central cavity. The mesoderm forms by delamination starting from the distal side towards the proximal end. In later embryos two polypides form on the posterior side that ultimately will be covered by a ciliated mantle in the larva. Embryos increase in size during development and form temporary cell contacts to the embryo sac. Mesodermal cells of the embryo sac show signs of transcellular transport indicating that embryos are nourished by transferring nutrients from the maternal coelom towards the brood cavity. This study clarifies several details such as mesoderm formation and the onset of bud development. Embryos are connected to their respective embryo sacs by a variety of temporary cytoplasmic processes formed by both tissues during embryogenesis, including a ‘placental’ ring zone. Although ultrastructural data of these cell contacts are not entirely conclusive about their function, we suggest that embryos absorb nutrients via the entire surface. The close opposition of embryos to the embryo sac implies placentation as matrotrophic mode in phylactolaemate bryozoans, with embryo sacs acting as placental analogues.

Angiosperm reproduction relies on the precise growth of the pollen tube through different pistil tissues carrying two sperm cells into the ovules’ embryo sac, where they fuse with the egg and the central cell to accomplish double fertilization and ultimately initiate seed development. A network of intrinsic and tightly regulated communication and signaling cascades, which mediate continuous interactions between the pollen tube and the sporophytic and gametophytic female tissues, ensures the fast and meticulous growth of pollen tubes along the pistil, until it reaches the ovule embryo sac. Most of the pollen tube growth occurs in a specialized tissue—the transmitting tract—connecting the stigma, the style, and the ovary. This tissue is composed of highly secretory cells responsible for producing an extensive extracellular matrix. This multifaceted matrix is proposed to support and provide nutrition and adhesion for pollen tube growth and guidance. Insights pertaining to the mechanisms that underlie these processes remain sparse due to the difficulty of accessing and manipulating the female sporophytic tissues enclosed in the pistil. Here, we summarize the current knowledge on this key step of reproduction in flowering plants with special emphasis on the female transmitting tract tissue.

Abstract The switch from the vegetative to the reproductive pathway of development in flowering plants requires the commitment of the subepidermal cells of the ovules and anthers to enter the meiotic pathway. These cells, the hypodermal cells, either directly or indirectly form the archesporial cells that, in turn, differentiate into the megasporocytes and microsporocytes. We have isolated a recessive pleiotropic mutation that we have termed multiple archesporial cells1 (macl) and located it to the short arm of chromosome 10. Its cytological phenotype suggests that this locus plays an important role in the switch of the hypodermal cells from the vegetative to the meiotic (sporogenous) pathway in maize ovules. During normal ovule development in maize, only a single hypodermal cell develops into an archesporial cell and this differentiates into the single megasporocyte. In macl mutant ovules several hypodermal cells develop into archesporial cells, and the resulting megasporocytes undergo a normal meiosis. More than one megaspore survives in the tetrad and more than one embryo sac is formed in each ovule. Ears on mutant plants show partial sterility resulting from abnormalities in megaspore differentiation and embryo sac formation. The sporophytic expression of this gene is therefore also important for normal female gametophyte development.

Pollen is the male gametophyte of higher plants. Its major function is to deliver sperm cells to the ovule to ensure successful fertilization. During this process, many interactions occur among pollen tubes and pistil cells and tissues, and calcium ion (Ca2+) dynamics mediate these interactions among cells to ensure that pollen reaches the embryo sac. Although the precise functions of Ca2+ dynamics in the cells are unknown, we can speculate about its roles on the basis of its spatial and temporal characteristics during these interactions. The results of many studies indicate that calcium is a critical element that is strongly related to pollen germination and pollen tube growth.

In Darwinia the floral parts are differentiated in a "calyx-orolla-gynoeciumandroecium" sequence. In individual buds stages of microsporogenesis markedly precede corresponding stages of megasporogenesis. The anther is tetrasporangiate with all sporangia lying in one plane. The secretory tapetum is one- to three-layered within the same microsporangium and a large number of Ubisch bodies are formed. The anthers dehisce by minute lateral pores and an ingenious mechanism helps disperse the twocelled pollen grains. A basal placenta in the single loculus of the ovary bears four ovules in D. micropetala and two in D. fascicularis. In both species, however, only one ovule is functional after fertilization. The fully grown ovules are anatropous, crassinucellar, and bitegmic; the inner integument forms the micropyle. The parietal tissue is most massive at the completion of megasporogenesis but is progressively destroyed later. The embryo sac follows the Polygonum type of developnlent and when mature is five-nucleate, the three antipodals being ephemeral. Following fertilization, the primary endosperm nucleus divides before the zygote. Subsequent nuclear divisions in the endosperm mother cell are synchronous and lead to a free-nuclear endosperm which becomes secondarily cellular, starting from the micropylar end at the time the globular embryo assumes an elongated shape. Embryogeny is irregular and the mature embryo is straight with a massive radicle and a hypocotyl which terminates in two barely recognizable cotyledons. Sometimes the minute cotyledons are borne on a narrow neck-like extension of the hypocotyl. A suspensor is absent. Both integuments are represented in the seed coat and only the outer layer of the outer and the inner layer of the inner integuments, with their thick-walled tanniniferous cells, remain in the fully grown seed. The ovary wall is demarcated into an outer zone containing oil glands surrounded by cells containing a tannin-like substance and an inner zone of spongy parenchyma. In the fruit this spongy zone breaks down completely but the outer zone is retained. The two species of Darwinia, while closely resembling each other in their embryology, differ significantly from other Myrtaceae. However, no taxonomic conclusions are drawn at this stage, pending enquiry into the life history of other members of the tribe Chamaelaucieae.

Cotula australis has a discoid heterogamous capitulum in which the outermost three whorls of florets are female and naked. The bisexual disk florets are fully fertile and have a four-lobed corolla with four shortly epipetalous stamens. The anthers contain only two microsporangia. Wall formation and microsporogenesis are described and the pollen grains are shed at the three-celled condition. The ovule is teguinucellate and the hypodermal archesporial cell develops directly as the megaspore mother cell. Megasporogenesis is normal and the monosporio embryo sac develops from the chalazal megaspore. Breakdown of the nucellar epidermis takes place when the embryo sac is binucleate and its subsequent development follows the Polygonum type. The synergids extend deeply into the micropyle and one persists until late in embryogeny as a haustorium. The development of the embryo is of the Asterad type, and the endosperm is cellular. C. coronopifolia agrees with C. australis in the presence of only two microsporangia in each anther and the development of a synergid haustorium.
