Oxidative stress has been suggested to negatively affect oocyte and embryo quality and developmental competence, resulting in failure to reach full term. In this study, we investigated the effect of N-acetyl-L-cysteine (NAC), a cell-permeating antioxidant, on developmental competence and the quality of oocytes and embryos upon supplementation (0.1–10 mM) in maturation and culture medium in vitro using slaughterhouse-derived oocytes and embryos. The results show that treating oocytes with 1.0 mM NAC for 8 h during in vitro maturation attenuated the intracellular reactive oxygen species (ROS) (p < 0.05) and upregulated intracellular glutathione levels (p < 0.01) in oocytes. Interestingly, we found that NAC affects early embryonic development, not only in a dose-dependent, but also in a stage-specific, manner. Significantly (p < 0.05) decreased cleavage rates (90.25% vs. 81.46%) were observed during the early stage (days 0–2), while significantly (p < 0.05) increased developmental rates (38.20% vs. 44.46%) were observed during the later stage (from day 3) of embryonic development. In particular, NAC supplementation decreased the proportion of apoptotic blastomeres significantly (p < 0.05), resulting in enhanced hatching capability and developmental rates during the in vitro culture of embryos. Taken together, our results suggest that NAC supplementation has beneficial effects on bovine oocytes and embryos through the prevention of apoptosis and the elimination of oxygen free radicals during maturation and culture in vitro.

Progeny of northern spring field cricket adults lay non-diapause eggs, undergo nymphal diapause, and overwinter as nymphs. Progeny of northern fall adults lay diapause eggs, do not undergo nymphal diapause, and overwinter as eggs. The two populations cannot interbreed freely in the field owing to a temporal difference in breeding seasons; they did not interbreed in the laboratory. Rearing experiments show that the developmental differences are genetically based rather than environmentally conditioned, and it is, therefore, unlikely that hybrids would be viable even if they were produced in the field. Consequently these two populations behave as good species. Field crickets from Virginia developed much more rapidly than did spring crickets from Quebec. Quebec spring males and Virginia females produced hybrids with developmental rates intermediate between those of their parents. More female than male hybrids were produced, and the females developed more rapidly than did male hybrids. Offspring were produced by hybrid females and Quebec spring males, but not by hybrid females and Virginia males. Partial, but incomplete reproductive isolation exists between Quebec and Virginia field crickets. A possible mechanism of sympatric speciation in insects is discussed.

Abstract Climate change has resulted in increasing temperature and acidification in marine systems. Rising temperature and acidification act as stressors that negatively affect host barriers to infection, thus enhancing disease processes and influencing the emergence of pathogens in ecologically and commercially important species. Given that crustaceans are ectotherms, changes in temperature dominate their physiological and immunological responses to microbial pathogens and parasites. Because of this, the thermal ranges of several crustacean hosts and their pathogens can be used to project the outcomes of infections. Host factors such as molting, maturation, respiration, and immune function are strongly influenced by temperature, which in turn alter the host’s susceptibility to pathogens, further amplifying morbidity and mortality. Microbial pathogens are also strongly influenced by temperature, arguably more so than their crustacean hosts. Microbial pathogens, with higher thermal optima than their hosts, grow rapidly and overcome host immune defenses, which have been weakened by increased temperatures. Pathogen factors such as metabolic rates, growth rates, virulence factors, and developmental rates are often enhanced by rising temperature, which translates into increased transmission, dispersal, and proliferation at the population level, and ultimately emergence of outbreaks in host populations. Less well known are the effects of acidification and salinity intrusion on host-pathogen processes, but they operate alongside temperature, as multiple stressors, that impose significant metabolic and physiological demands on host homeostasis.
