The present paper is the first of a series of reports on the relative reaction of living tissues as determined by vital staining with indicators. It is possible to bring about a localized and a general coloration of living rats and mice with litmus. The animals remain in good health and the coloration of some of the tissues persists for months. Much of the dye is stored in cell granules, especially in those of the reticulo-endothelial elements, but a diffuse staining of certain tissues occurs, notably of bone, epidermis, cartilage, and connective tissue everywhere. In the intensity and localization of the bony coloration litmus has resemblances to madder. Diffuse staining with it renders blue most, if not all, of the tissues affected, while a granular staining causes others to become notably pink, owing to the fact that the indicator, though introduced into the organism in the blue form and circulating as such in the body fluids, is ordinarily red when stored in cells. The polymorphonuclear elements and macrophages of a peritoneal exudate, may become so laden with material colored red by litmus that the blue color of the fluid constituent is masked and the exudate appears a deep, turbid red. The phenomenon is but one manifestation of a notable acidity within cell granules throughout the organism. Like many another in the stained animals it would appear to be of physiological import. Some of the questions suggested by the work will be dealt with in the paper immediately following.

Trypan blue injected intravenously is bound almost at once by the intercellular connective tissue elements all over the body,—by collagen, reticulin, and elastic fibers. This union of dye and tissue elements is the factor responsible for the early macroscopic blue color and is antecedent to cellular colloidopexic action. Different examples of connective tissue differ among themselves in their ability to hold the dye. Diffuse staining of elastic fibers noted by previous observers is merely a special case of the general affinity of connective tissue for the dye. The evidence suggests that the histiocytes are cells specialized to segregate noxae that become diffusely bound to the intercellular connective tissue matrix.

The acidity of the macrophage granules in which litmus comes to be stored during life is considerable. It has proved possible to stain these granules in vivo with some of the phthalein indicators and the results, had they been obtained under controlled conditions, would indicate a pH of 3.0 or less. The amount of acid material which may accumulate within the cells of animals stained with litmus is great, sufficient in the case of the elements of a peritoneal exudate for the acid reaction to prevail when they are gathered together and crushed. The material is derived, not from the dye, but from living elements responding characteristically to a stimulus far from unique. Such responses may well play a rôle in normal physiological activities and in the cellular defense against microorganisms. Vital staining with litmus demonstrates anew that the intracellular reaction during life is independent of that of the body fluids. By means of color changes in the stored indicator one can distinguish sick as well as dead cells of certain sorts and follow their distribution and fate within the organism. There are data to suggest that with the aid of the indicator the normal period of survival of certain elements at least can be determined. By the indicator method, of which the foregoing observations afford a crude illustration, much should be learnt in the future about body processes. The present paper is the second of a series upon the theme.


Attempts to achieve “internal disinfection,” that is to say, to kill organisms in the body of infected animals by means of drugs, have hitherto afforded little promise of success in the case of the common pathogenic bacteria, the only exception being the action of ethyl-hydrocuprein in pneumococcus infections, discovered by Morgenroth with his co-workers, Levy and others. The reasons for the failure are probably to be attributed mainly to two facts; in the first place, antiseptics in general enter into combination with proteins of the tissues and body fluids, either by a process of physical adsorption or by the formation of chemical compounds. In either case, the usual result is that the bactericidal action of a substance, as determined in a watery medium, is greatly reduced by protein-solutions, e. g. , serum. Secondly, the majority of chemical antiseptics are general protoplasm poisons, and exert on mammalian tissues a degree of toxic action equal to, or greater than, that which they exhibit towards micro-organisms; hence these substances prove lethal for animals in doses which are insufficient to confer bactericidal properties on the body fluids. The classical example of such failures was afforded by mercuric chloride, which Koch employed in the hope of treating effectively anthrax septicæmia in animals. Ehrlich and Bechhold investigated compounds which were more potent antiseptics than any hitherto known, among the most active being tetrabrom- (and chlor-) ortho-biphenol; they found, however, that the bactericidal properties of these substances were also greatly reduced when the organisms were suspended in a serum medium. Thus, tetrachlor-orthobiphenol in a concentration of 1 : 320,000 prevented the growth of diphtheria bacilli in bouillon, whereas in serum growth occurred in the presence of 1 : 10,000 of this reagent. Similarly we have found that the dose of perchloride of mercury which is required to inhibit completely the growth of Staphylococcus aureus or B. coli in serum is 100 times greater than that which produces this effect in watery medium containing a small amount of nutrient peptone (0·7 per cent.).

We have devised methods for the separation and isolation of the important indicator constituents of litmus, azolitmin, and erythrolitmin, with a view to employing them as vital stains. Analysis of the color intensities of these dyes shows slight differences in them, azolitmin being the weaker pigment, weight for weight. Study of a third coloring matter, erythrolein, which exists in litmus has shown it to be an unsatisfactory indicator, and toxic for animals. Analyses with the spectrophotometer of the absorption of light by erythrolitmin and azolitmin, prepared by our methods, and tested over a wide acid-alkali range, show them to be pure substances, comparable in this respect with synthetic indicators. The errors in the interpretation of the indicator phenomena on vital staining, which are incident to changes in the concentration of the dyes, are so slight as to be negligible. The salt and protein errors on the other hand are large. The factors responsible for the Donnan equilibrium fail to influence the distribution of the indicators between fluid and gelatin. Erythrolein was found useless when employed for vital staining, and azolitmin proved unsatisfactory since it colors poorly and is toxic. But erythrolitmin can be used to great advantage. It is readily absorbed, and in non-toxic doses stains intensely. The range of pH at which it changes from red to blue fits it for the demonstration of changes in the reaction of living tissues. By reason, however, of the salt and protein errors to which it is liable, the pH cannot be accurately ascertained. Intravital staining with erythrolitmin yields results similar to those following injection of purified "whole litmus."

In this globalization era, health is the most important thing to be able to run various activities. Without good health, this will hinder many activities. Diabetes mellitus is one of the diseases caused by unhealty lifestyle.There are many treatments that can be done to prevent the occurrence of diabetes. The treatments are giving the insulin and also checking the glucose rate to the patients.Checking the glucose rate needs the tools which is safety to the body. This research want to develop non invasive tool which is safety and do not injure the patient. The purpose of this research is also finding the best model which derived from Linear, Quadratic, and Cubic Spline Regression. Some respondents were taking to get the glucose measuring by invasive and non invasive tools. It could be seen clearly that Spline Linear Regression was the best model than Quadratic and Cubic Spline Regression. It had 70% and 33.939 for R2 and RMSEP respectively.

Abstract: The phenomenon of the people who forcibly took covid's corpse 19 from the hospital to be taken care of by Fardhu Kifayah by his family and the community, became a conclusion that there was community doubt about the management of Tajhiz Mayat conducted by the hospital. Coupled with the circulation of the video of the Ruku movement 'in the corpse prayer conducted by unscrupulous parties at the Hospital, became added doubts from the public against the hospital. To solve this problem, this research uses a Descriptive Analysis approach, namely by formulating a question, namely How to arrange Covid 19's body in Banda Aceh and this question will be answered with several theories and data sets from the field. So it was concluded in a conclusion that answered the formulation of the problems mentioned. Theoretically the spread of covid 19 is very fast, the size of the virus is only 0.1 micrometer and is in body fluids, especially nasopharyngeal fluid and oropharyngeal fluids of infected people, fluids in the body of covid 19 bodies can get out through every gap of the body such as mouth, nose, eye and rectum, because it requires special techniques in its management. Fardhu kifayah to covid 19 bodies should be carried out by trained Ustad and trained health workers, so that the spread stopped. The results of this study concluded that the management of the Moslem bodies died at Zainal Abidin Hospital in Banda Aceh was in accordance with the Fatwa of the Aceh Ulama Council (MPU) and the bodies were handled by trained Ustad and health workers.

AbstractThe ex-vivo biochemical changes of different body fluids also referred as aging of fluids are potential marker for the estimation of Time since deposition. Infrared spectroscopy has great potential to reveal the biochemical changes in these fluids as previously reported by several researchers. The present study is focused to analyze the spectral changes in the ATR-FTIR spectra of three body fluids, commonly encountered in violent crimes i.e., semen, saliva, and urine as they dry out. The whole analytical timeline is divided into relatively slow phase I due to the major contribution of water and faster Phase II due to significant evaporation of water. Two spectral regions i.e., 3200–3400 cm−1 and 1600–1000 cm−1 are the major contributors to the spectra of these fluids. Several peaks in the spectral region between 1600 and 1000 cm−1 showed highly significant regression equation with a higher coefficient of determination values in Phase II in contrary to the slow passing Phase I. Principal component and Partial Least Square Regression analysis are the two chemometric tool used to estimate the time since deposition of the aforesaid fluids as they dry out. Additionally, this study potentially estimates the time since deposition of an offense from the aging of the body fluids at the early stages after its occurrence as well as works as the precursor for further studies on an extended timeframe.

Whether the constellation of various symptoms reported in various case-study reports on some patients who have had augmentation mammoplasty with silicone implants reflects a distinct, novel “silicone syndrome”or disease is important to settingproper endpoints for the epidemiological study of this patient population. To date, epidemiology studies on breast implant patients have focused on end-points which are typical of connective tissue disease, rheumatoid disease, and/ or autoimmune disorders. The consensus at this time, as was recently stated in a paper authored by Food and Drug Administration (FDA) personnel, is that the weight of the evidence from existing epidemiology studies is that silicone breast implants do not appreciably, if at all, increase the risk of these types of diseases. Critics of the epidemiology database have countered that had the analysis of association in these studies been done for a “silicone syndrome,” as opposed to the disease types which were analyzed, an association between silicone breast implantation and increased risk of “silicone syndrome” would have been observed. In the present analysis, this question is approached from two directions: First, the available single or multi-patient case reports available in the open literature were evaluated. The objective was to define those symptoms/ complaints that were reported in all studies or in at least 50% of the patients reported and to assign frequency distributions to individual symptoms or complaints reported in breast implant patients presenting for various complaints. By definition, if a “silicone syndrome” exists, then it can only be characterized by those symptoms or complaints which appear with regular frequency in patients so afflicted. Second, the symptoms or complaints which were used as criteria in the existing epidemiology studies were correlated with their frequency of occurrence among single or multi-patient case-reported breast implant patients. The working hypothesis in this present study is that if the number of “silicone syndrome” symptoms or complaints that also are symptoms of the existing epidemiology endpoints is large, then a distinct “silicone syndrome” is not likely to exist, and it can be concluded that existing epidemiology studies have adequately addressed the relevant issues. Also, to the extent that the frequency of symptom occurrence in “silicone syndrome” is similar to the distribution seen for known connective tissue, rheumatoid, and/ or autoimmune diseases, this will then add to the weight of evidence that no distinct “silicone syndrome” needs be postulated. Conversely, if a different set of symptoms or complaints occurs in silicone breast-implanted patients than is seen in patients with connective tissue diseases, this will argue that a distinct syndrome may exist. In the present study, the more recent suggestion that silicone may be broken down to silica in the body, and evidence for and against this suggestion are also discussed. The present analysis does not support the contention that a distinct “silicone syndrome” exists, but does support the contention that the disease endpoints used in existing epidemiology studies are adequate for examining the patient population. Also, consideration of the chemistry of silicone and its potential hydrolysis or oxidative cleavage indicates that if such reactions occur in the body at any significant rate, the product will be silicic acid, a normal and necessary constituent of the body, and not silica (i.e., silicon dioxide).
