Phytophthora root rot of citrus in Florida is caused by Phytophthora nicotianae and P. palmivora. A naturally occurring isolate of P. nicotianae (Pn117) was characterized as hypovirulent on citrus roots. Pn117 infected and colonized fibrous roots, but caused significantly less disease than the virulent isolates P. nicotianae Pn198 and P. palmivora Pp99. Coincident inoculation of rootstock seedlings of Cleopatra mandarin (Citrus reticulata) or Swingle citrumelo (C. paradisi × Poncirus trifoliata) with the hypovirulent Pn117 and the virulent isolates Pn198 and Pp99 did not reduce the severity of disease caused by the virulent Phytophthora spp. When either rootstock was inoculated with the hypovirulent Pn117 for 3 days prior to inoculation with virulent isolates, preinoculated seedlings had significantly less disease and greater root weight compared with seedlings inoculated with the virulent isolates alone. Recovery of the different colony types of Phytophthora spp. from roots of sweet orange (C. sinensis) or Swingle citrumelo was evaluated on semiselective medium after sequential inoculations with the hypovirulent Pn117 and virulent Pp99. Pn117 was isolated from roots at the same level as the Pp99 at 3 days post inoculation. Preinoculation of Pn117 for 3 days followed by inoculation with Pp99 resulted in greater recovery of the hypovirulent isolate and lower recovery of the virulent compared with coincident inoculation.

Phytophthora nicotianae is a devastating oomycete plant pathogen with a wide host range. On tobacco, it causes black shank, a disease that can result in severe economic losses. Deployment of host resistance is one of the most effective means of controlling tobacco black shank, but adaptation to complete and partial resistance by P. nicotianae can limit the long-term effectiveness of the resistance. The molecular basis of adaptation to partial resistance is largely unknown. RNAseq was performed on two isolates of P. nicotianae (adapted to either the susceptible tobacco genotype Hicks or the partially resistant genotype K 326 Wz/Wz) to identify differentially expressed genes (DEGs) during their pathogenic interactions with K 326 Wz/Wz and Hicks. Approximately 69% of the up-regulated DEGs were associated with pathogenicity in the K 326 Wz/Wz-adapted isolate when sampled following infection of its adapted host K 326 Wz/Wz. Thirty-one percent of the up-regulated DEGs were associated with pathogenicity in the Hicks-adapted isolate on K 326 Wz/Wz. A broad spectrum of over-represented gene ontology (GO) terms were assigned to down-regulated genes in the Hicks-adapted isolate. In the host, a series of GO terms involved in nuclear biosynthesis processes were assigned to the down-regulated genes in K 326 Wz/Wz inoculated with K 326 Wz/Wz-adapted isolate. This study enhances our understanding of the molecular mechanisms of P. nicotianae adaptation to partial resistance in tobacco by elucidating how the pathogen recruits pathogenicity-associated genes that impact host biological activities.

DISEASE: Black shank of tobacco. GEOGRAPHICAL DISTRIBUTION: Africa (Malawi, Mauritius, Uganda); Asia (Ceylon, China, Formosa, India, Indonesia, Japan, Java, Malaya, Philippines, Sumatra); Central America & West Indies (Cuba, Jamaica, Puerto Rico, Santo Domingo, Trinidad); Europe (Bulgaria, Germany, Greece, Italy, Poland, Romania, U.S.S.R.); North America (U.S.A.); South America (Brazil, Colombia, Venezuela). TRANSMISSION: Soil-borne, persisting in soil for at least 4 years between tobacco crops, and not eliminated by a 3 year rotation or a 4 year fallow (41: 409; 39: 126). Tobacco leaves have been used to indicate disease potential in infested soil after serial dilution with sterile soil (42: 408). Spread in contaminated pond water used in overhead irrigation also suspected (43, 1413). Wind-borne spread up to 800 ft. has been recorded (39: 500).

Uma alternativa de manejo das doenças causadas por Phytophthora spp. é o uso de matéria orgânica. No presente trabalho foi avaliada a potencialidade do lodo de esgoto na indução de supressividade in vitro a P. nicotianae. O efeito do lodo de esgoto incorporado ao solo na sobrevivência de P. nicotianae foi avaliado mediante um experimento fatorial com dois fatores: doses de lodo de esgoto (0, 10, 20 e 40% p/p) e concentrações de inóculo [0, 10 ou 20 g de grãos de trigo (Triticum aestivum) colonizados kg-1]. Aos 21 dias, quando aumentaram as doses de lodo de esgoto, a sobrevivência de P. nicotianae e os pHs das misturas diminuíram, e as condutividades elétricas (CE) aumentaram. As correlações entre a CE e a sobrevivência do patógeno foram negativas e significativas (P>0,05). Para estudar o efeito dos compostos químicos envolvidos na supressividade, foram obtidos extratos em água, H2SO4 2N e KOH 0,4N de misturas de areia – lodo de esgoto (20% p/p), e foram acrescentados ao meio de cultura e seu efeito avaliado no crescimento das colônias de P. nicotianae. O extrato ácido (H2SO4 2N) do tratamento com 20% de lodo de esgoto inibiu significativamente (P>0,05) o crescimento da colônia do patógeno. O efeito biológico foi estudado mediante isolamento de microrganismos em meio de cultura e seleção por antagonismo. No bioensaio com plântulas de alfafa (Medicago sativa) destacaram-se os isolados F9.1 (Aspergillus sp.) e A12.1 (actinomiceto, não identificado); e no teste de culturas pareadas destacou-se um Trichoderma sp. e dois actinomicetos por antibiose, e um Trichoderma sp. e três Aspergillus sp. por hiperparasitismo.

Este trabalho avaliou a resistência à infecção de tronco e de raízes por Phytophthora nicotianae em híbridos somáticos de citros com potencial para serem utilizados como porta-enxertos. Os híbridos somáticos avaliados foram laranja 'Hamlin' (Citrus sinensis) + toranja 'Indian Red' (Citrus grandis) (plantas 1 e 2) e laranja 'Hamlin' (C. sinensis) + toranja 'Singapura' (C. grandis). Plantas de limão 'Cravo' (Citrus limonia), laranja 'Caipira' (C. sinensis), laranja-azeda (C. aurantium) e Poncirus trifoliata 'Davis A' (Poncirus trifoliata) foram utilizadas como plantas-controle devido à reação conhecida à infecção pelo patógeno. Avaliações realizadas entre 30 e 60 dias após as inoculações com o patógeno incluíram o comprimento das lesões no tronco e a massa seca do sistema radicular nas plantas avaliadas. O híbrido somático laranja 'Hamlin' + toranja 'Indian Red' (planta 1) mostrou-se tolerante a P. nicotianae, indicando potencial para continuidade nas suas avaliações como porta-enxerto para citros.

This work aimed to evaluate the potassium phosphite-based commercial product, Phytogard®, as a complementary tool for the management of gummosis in citrus. Seedlings of tangerine ‘Sunki’ were sprayed at concentrations 0; 0.5; 2 and 5 mL L-1 of Phytogard® and subsequently inoculated with zoospores of Phytophthora nicotianae. The disease incidence was reduced by 84% in plants sprayed at the concentration 0.5 mL L-1 and the plants sprayed at concentrations of 2 and 5 mL L-1 showed zero disease incidence. There was increased an production of fresh matter of shoot and roots in plants sprayed and inoculated with the pathogen compared to unsprayed plants. Plants sprayed with Phytogard® and inoculated with the pathogen showed lower values for total phenols, enzyme activity for phenylalanine ammonia-lyase and peroxidase and for total protein content in root tissues compared to non-inoculated plants. There was higher activity of the enzyme β 1.3-glucanase in root tissues of plants inoculated with the pathogen that received the product at the concentration of 2 mL L-1. The results of this study showed that the potassium phosphite-based product Phytogard® has potential for the control of Phytophthora nicotianae in seedlings of tangerine ‘Sunki’. However, it is not possible to conclude that this control occurs through resistance induction.
