Background and Aim: Hydrolyzable tannins are an important group of secondary plant metabolites, which are known for antimicrobial activity. This study aimed to assess the efficiency with which a hydrolyzable tannin extract from sweet chestnut wood (Castanea sativa Mill.) could inhibit mastitis-causing bacteria in vitro. Materials and Methods: The negative control used was sterile water, and the positive controls were penicillin and gentamicin. The treatments included five concentrations of hydrolyzable tannins (63, 190, 313, 630, and 940 mg/mL). In cows with subclinical mastitis, the bacteria causing the disease were isolated and identified. Then, the antibacterial activity of the hydrolyzable tannin extract was assessed by the disk diffusion method, by determining the minimum inhibitory concentration (MIC) and by determining the minimum bactericidal concentration (MBC). Results: Penicillin inhibited (p<0.01) the growth of Staphylococcus aureus, Streptococcus uberis, and Pseudomonas aeruginosa but could not inhibit (p>0.05) the growth of Streptococcus agalactiae, Escherichia coli, and Klebsiella pneumoniae. However, gentamicin and hydrolyzable tannins could inhibit (p<0.01) all isolated bacteria. Increasing the concentration of hydrolyzable tannin extract resulted in a quadratic increase in the inhibition zone diameter of S. aureus and S. agalactiae and a linear increase in the inhibition zone diameter of E. coli, K. pneumoniae, and P. aeruginosa. In addition, 630 and 940 mg/mL of hydrolyzable tannin extract showed the highest antibacterial activity against S. agalactiae and E. coli (p<0.01), while 940 mg/mL concentration had the highest antibacterial activity against K. pneumoniae (p<0.01). The MIC and MBC of the extract were 27.3-190 mg/mL and 58.8-235 mg/mL, respectively, with the MBC: MIC ratio being 2:1. Conclusion: The antimicrobial activity of the hydrolyzable tannin extract against subclinical mastitis bacteria was comparable to the antibiotics (positive controls) at concentrations over 630 mg/mL. Although these in vitro findings are promising, further research is needed to determine whether hydrolyzable tannins could be used to control or prevent subclinical mastitis in dairy cows.

Objective: To evaluate various explants and growth regulators in order to improve invitro propagation of Stevia rebaudiana through organogenesis.Design/Methodology/Approach: Explants and growth regulators in two differentconcentrations were evaluated. The explants were nodal segment, axillary bud, andapical meristem; while the growth regulators were benzylaminopurine (BAP) at 1.125 mgL -1 and 3.0 mg L -1 , naphthaleneacetic acid (NAA) at 1.5 mg L -1 and 3.0 mg L -1 , andCIDEF-4 brassinosteroids (BRs) at 1.0 mg L -1 and 1.5 mg L -1 . In total 18 treatments withseven repetitions. Contamination, oxidation, and survival were recorded duringinduction; while leaf number, regrowth height, and root presence were recorded duringmultiplication.Results: At the induction stage there was a differential response between explantsaccording to their ontogenetic age; during multiplication, the morphological componentsshowed differences between concentrations of growth regulators and explants, withhigher effectiveness when adding BAP to apical meristems.Study Limitations/Implications: Both the origin and the age of explants can inducedifferential growth while interacting with growth regulators. Findings/Conclusions: Apical meristem explants showed better advantages for in vitroreproduction of S. rebaudiana since they present less contamination and higher survivalat the induction stage, even when exhibiting the highest oxidation among explants,which did not influence the decrease in their survival. At the multiplication stage withapical meristem, height, leaf number, and root presence were increased. Values werehigh when interacting with BAP.

This paper presents the five-year long research results (2007-2011) of phenological observations (beginning, full flowering and the end of flowering), morphometric analysis (fruit weight, length, width and thickness of fruit, as well as length and width of the hilum), the ripening time and the average yield of selected genotypes of sweet chestnut (Castanea sativa Mill.). The results show that in terms of time of flowering chestnut trees examined in the group are very early flowering. The ripening of studied chestnut trees shows that the earliest harvest tree was Ostros II, VII and Kostajnica V (09.11.), and Kostajnica III (14.11.) were the latest. The average fruit weight of the selected sweet chestnut trees was (6.9 g). The highest average fruit weight (10.6 g), had the examined Ostros I tree, and the lowest (4.8 g), Kostajnica VI. The yield of the tested chestnut trees was on average (76.3 kg / tree). The selected trees Kostajnica II (66.0 kg / tree), Kostajnica I (69.0 kg / tree) had the smallest yield and Ostros VI (94.0 kg / tree) and Ostros V (87.0 kg / tree) had the highest. Obviously it can be concluded that population genetic variability is very high and gene expression is highly affected in tested samples on both locations. For further research it should be given attention to investigate genotypes in controlled conditions, the best in vitro in tissue culture.
