An experiment was conducted to study the efficacy of two tomato pastes and aronia nectar (fruit juice + pulp from the black chokeberry, Aronia melanocarpa Elliot) as inhibitors of nitrosamine production in cancer prophylaxis programmes. White male rats of the Wistar strain were employed in an acute trial. Aminopyrin + sodium nitrite (APSN) were used as precursors for generation of endogenous nitrosamine. The animals were allocated to different dietary groups and fed by intubation with APSN or APSN + food products. Introduction of tomato paste (TP), high-β-carotene tomato paste (HCTP) and aronia nectar (AN) as inhibitors of n−nitrosamine formation exerted a positive effect on blood and liver variables which was demonstrated by decreased concentrations of glutamic-oxaloacetic transaminase (EC 2.6.1.1), glutamic-pyruvic transaminase (EC 2.6.1.2) and uric acid in serum and lipid content in hepatocytes. Animals treated with APSN developed dystrophic changes in liver such as centrolobular necrosis, intense exangia, and enlarged cells with two, often large, pyknotic nuclei, while the structure of livers of rats fed with TP, HCTP or AN was well protected and almost normal. TP had a particularly beneficial effect on serum total protein and albumin concentrations as had AN on the urea value. The inhibitory effect of th food products used is explained by their chemical nature including pH, ascorbic index (ascorbate:nitrate), lycopene and β-carotene contents.

Abstract: Epidemiological study in Manado indicated that the prevalence of Diabetes Melitus (DM) was 6.1%. It is necessary to do scientific assessment of the various species of plants which are thought containing medicinal substances (phytopharmaca). Based on the information from the people living in the subdistrict Pineleng Minahasa red gedi leaves (Abelmoschus manihot L.Medik ) can be used as herbal treatment for lowering blood sugar level. This study aimed to evaluate the effect of red gedi leaves extract in lowering blood sugar level on white male rat induced by aloksan. This study used 15 white male rats of Wistar strain with diabetic induced by 200 mg/kg BW aloksan intraperitoneal. The rats were randomly divided into 5 groups; each group were given aquadest 2,5 ml/ 200 g BW, insulin Novomix 0,9 U/Kg BW, red gedi leaf extract 1.25 mg/200 g BW, 2.5 mg/200 g BW, and 3.75 mg/200 g BW, once daily for 24 hours. Blood sugar levels were measured at 6th, 12th, 18th, and 24 th hours after treatment by using glucometer. Untuk itu perlu dilakukan pengkajian ilmiah terhadap berbagai spesies tumbuhan yang diduga mengandung zat berkhasiat obat (fitofarmaka). Berdasarkan informasi masyarakat di daerah kecamatan Pineleng kabupaten Minahasa daun Gedi Merah (Abelmoschus manihot L.Medik) dapat dimanfaatkan sebagai penanganan herbal untuk menurunkan kadar gula darah. Penelitian ini bertujuan menguji efektivitas ekstrak daun Gedi Merah (Abelmoschus manihot L. Medik) dalam menurunkan kadar gula darah tikus putih jantan galur Wistar (Rattus norvegicus) yang diinduksi aloksan. Tikus galur Wistar sejumlah 15 dinduksi dengan aloksan intraperitoneal 200 mg/kg BB dan dibagi secara acak dalam 5 kelompok penelitian yaitu kelompok perlakuan dengan aquadest 2,5 ml/200 g BB, insulin Novomix 0,9 U / Kg BB, ekstrak daun gedi merah 1,25 mg/ 200 g BB, 2,5 mg/ 200 g BB dan 3,75 mg/ 200 g BB. Perlakuan diberikan satu kali sehari selama 6 hari. Pengukuran kadar glukosa darah dilakukan pada hari ke 2, 4 dan 6 setelah perlakuan menggunakan glukometer. Hasil penelitian menunjukkan bahwa ekstrak daun gedi merah dapat menurunkan kadar glukosa darah tikus putih jantan diabetes yang diinduksi dengan aloksan.Kata kunci: ekstrak daun gedi merah, glukosa darah, aloksan, diabetes

ABSTRACT Pain is an unpleasant sensory and emotional experience due to tissue damage both actual and potential. Analgesics are a drug used to relieve pain without losing consciousness. Keji beling leaves (Strobilanthes crispus L) contain glycosidic ester compounds from caffeic acid, which have analgesic effects. This study aims to determine whether there is an analgesic effect on the ethanol extract of keji beling leaves with different doses, namely dose 0.0023g / 200gBW, 0.0047g / 200gBW, and 0.0094g / 200gBW in test animals of wistar strain male rats. This study uses a laboratory experimental research design using wistar strain male white rat as a test animal. The analgesic effect test was carried out by the heat stimulation method using a 53 °C water bath. The rat response was observed in the form of movements of licking the hind legs or jumping from before giving test material and after giving test material at 30, 60, 90 and 120 minutes, respectively. From the results of statistical data analysis with One Way Anova and LSD showed that the extracts keji beling leaves has an analgesic effect. Where among those three doses the best dose was 0.0023g / 200gBW. Keywords: Analgesic, anova, keji beling leaves, white male wistar rats. ABSTRAK Rasa nyeri merupakan pengalaman sensorik dan emosional yang tidak menyenangkan akibat kerusakan jaringan baik aktual maupun potensial. Analgesik ialah suatu obat yang digunakan untuk menghilangkan rasa sakit tanpa menghilangkan kesadaran. Daun keji beling (Strobilanthes crispus  L) mengandung senyawa glikosidik ester dari asam caffeic yang memiliki efek analgesik. Penelitian ini bertujuan untuk mengetahui ada tidaknya efek analgesik pada ekstrak etanol daun keji beling dengan dosis berbeda-beda yaitu dosis 0,0023g/200gBB, 0,0047g/200gBB, dan 0,0094g/200gBB pada hewan uji tikus putih jantan galur wistar. Penelitian ini menggunakan Rancangan penelitian eksperimental laboratorium dengan menggunakan hewan uji tikus putih jantan galur wistar. Pengujian efek analgesik dilakukan dengan metode rangsangan panas menggunakan waterbath suhu 53°C. Respon tikus diamati berupa gerakan menjilat kaki belakang atau melompat dari sebelum pemberian bahan uji dan sesudah pemberian bahan uji  berturut-turut pada menit ke 30, 60, 90 dan 120. Dari hasil analisis data secara statistika dengan One Way Anova dan LSD menunjukkan bahwa ekstrak daun keji beling memiliki efek analgesik. Dimana antara ketiga dosis tersebut yang paling terbaik ialah dosis 0,0023 g/200gBB. Kata kunci : Analgesik, Anova, Daun Keji Beling, tikus putih jantan galur wistar

Objective: To compare the number of apoptosis, necrosis and viability of germinal epithelial cells in the contralateral testicle (CT) and plasma testosterone (PT) levels in white male rats which administered Nifedipine after unilateral testicular torsion (TT) compared with control group. Material & Methods: This research was an experimental study using male white rats (Rattus Norvegicus, Wistar strain) aged 10-12 weeks and body weight 150-200 gram. A total of 30 rats were then randomly divided into 5 groups (n=6) which were negative control group (KN), positive control group (KP1 and KP2) and Nifedipine-administered group (N1 and N2). Each group performed unilateral left side torsion of testicular of 1080o anticlockwise except the KN group. There was 4-hour ischemic duration in the KP1 and N1 groups while 10 hours in the KP2 and N2 groups. Administration of Nifedipine 30 minutes before detorsion by intraperitoneal injection dosed 100 μg/kgBW. All groups performed right orchidectomy and plasma blood sampling. Measurement of apoptosis, necrosis and viability of germinal epithelial cells in the CT using flowcytometry. Measurement of PT levels using Enzyme-Linked Immunosorbent Assay (ELISA). Results: The number of apoptosis, necrosis and viability of contralateral testicular germinal epithelial cells and PT levels in the KN group compared with KP1 and KP2 groups were significantly different (p<0.05). There was no significant discrepancy in apoptosis (p>0.05) in KP1 group compared with N1 group, as well as in KP2 group compared with N2 groups. The number of necrosis, viability of germinal epithelial cells in the CT and PT level in KP1 group in compared with N1 group, as well as in KP2 group in compared with N2 group were significantly different (p<0.05). Conclusion: Nifedipine administration prior to testicular detorsion can maintain cell viability and decrease the amount of necrotic germinal epithelial cells in the CT and prevent the decrease in PT levels after unilateral TT.

Hypertriglyceridemia is one of the risk factors for cardiovascular disease and acute pancreatitis (Pejic and Lee, 2006; Brahm and Hegele, 2013; Afari et al., 2015). One way to treat it by using the apple vinegar (Malus sylvestris Mill) of manalagi varieties which contained high antioxidants and acetic acid as well as more durable than the fresh fruit. The study aimed to know the effect of the Apple vinegar of manalagi varieties (Malus sylvestris Mill) to blood triglyceride levels of white male rats (Rattus norvegicus) wistar strain with high-fat diet and induced by alloxan. The study used the post test only control group design, with 24 rats samples which were divided into three groups. The negative control group (K0) only received standard diet for 24 days, the positive control group (K1) received a high-fat diet for 7 days and induced by alloxan with doses of 90 mg/kgBW of rats, and treatment group (K2) was given a high-fat diet for 7 days, induced by alloxan and received the apple vinegar (Malus sylvestris Mill) of manalagi varieties with a dose of 0,27 ml/100 g BW of rats for 14 days. The results by using one way ANOVA showed the significance value of the data p = 0,974 (p>0,05) so that there is no significant difference of the mean of blood triglycerides levels between K0, K1 and K2. This study concluded that apple vinegar (Malus sylvestris Mill) of manalagi varieties can not significantly lower the blood triglyceride levels of white male rats (Rattus norvegicus) wistar strain with high-fat diet and induced by alloxan. Abstract: Cholesterol is an important element in the body, but in high quantities, it can cause atherosclerosis that will eventually possess an impact on coronary heart disease. One of the foods that can reduce cholesterol is a food contains flavonoids which is found in melon seeds (Cucumis melo l.). It contains high fiber that can reduce either of excessive insulin or blood fat levels. The objective study was to determine the effect of Melon seeds flour Toward Total Cholesterol Level in White Male Wistar Strain Rats. This research was carried out with pre-expriment research design, used One group pretest-postest with 5 male wistar strain rats. Data collected in the form of data examination results of total cholesterol level before and after introducing Melon seeds flour. The mean result of total cholesterol test in white male rats before giving Melon seeds flour was 261 mg / dL, whereas the result of total cholesterol test after giving Melon seeds flour was 190.2 mg/dL. Cholesterol levels decreased by 70.8 mg/dl. In short, presenting Melon seeds flour (Cucumis melo l.) for 14 days can decrease total cholesterol level in white male wistar rats. Keywords: Melon Seeds, wistar strain, total cholesterol.

Abstract. It has been suggested that treatment of intact male rats with oestradiol benzoate (OeB) causes an interference with testosterone (T) production by the testes by a direct inhibitory effect on steroidogenesis. To test this hypothesis, different doses (5, 10 or 25 IU) of hCG were administered concomitantly with 50 μg of OeB to adult intact or hypophysectomized male rats. The testicular and plasma testosterone, and serum hCG levels were determined. The sex accessory weights were recorded. In the intact OeB-treated group of animals, hCG stimulated both the secondary sex organs and plasma testosterone levels above the intact control group. However, in hypophysectomized animals, although plasma testosterone levels increased above that of intact controls, their secondary sex organ weights did not. Moreover, inspite of high circulating hCG levels, the testicular testosterone content and concentration remained suppressed in OeB-treated animals. The reason for such dichotomy of hCG action on OeB-treated animals is not clear at present.

The aim of the paper is to study thymus variability in white rats, which were exposed to formaldehyde, and to compare these data with the indicators in control animals. Materials and Methods. The trial enrolled 72 white male rats, initial body weight 40–50 g. The animals were divided into 2 groups (36 rats in each). The first group consisted of control rats. Animals of the second group were exposed to formaldehyde inhalation, 2.766 mg/m3. To characterize the variability of the organ size, centroids were determined. The superposition of landmark configurations was performed using the generalized Procrustes analysis method, MorphoJ 1.06d program. The principal component analysis and canonical analysis of the obtained data were carried out. Results. One-Way ANOVA revealed a high level of intergroup differences in Procrust distance (F=1.34; p<0.0001). The significant effect of the duration of formaldehyde exposure on centroid size was established. The Kruskal-Wallis criterion was 19.778 (p=0.0014). The analysis of the principal components indicated that each of the first 10 components stands for more than 1 % of Procrustes coordinate variance. In this case, the first 7 components compatibly explain 91.398 % of thymus variability. The proportion of the first main component to the total variance of the Procrustes coordinates is 40.236 %. PC1 (-) shows changes in the thymus shape, mostly affecting the tops of its lobes, the middle part of the right boundary and the entire left thymus boundary. The scattering ellipses of the thymus ordinates in rats exposed to formaldehyde, in the first two canonical variables are located higher than those in the control animals. Conclusion. Formaldehyde inhalation leads to thymus changes in white rat. The most significant differences with control data are determined along the second canonical variable. Изучение изменчивости формы тимуса белых крыс, находившихся в условиях влияния формальдегида, и сравнение этих данных с показателями, полученными у контрольных животных. Материалы и методы. Работа выполнена на 72 белых крысах-самцах с начальной массой тела 40–50 г. Животные были разделены на 2 серии (по 36 крыс). Первую серию составляли контрольные крысы. Животные второй серии подвергались ингаляционному воздействию формальдегида (ФА) в концентрации 2,766 мг/м3. Для характеристики изменчивости размеров органов определяли размер их центроидов. Процедуру суперимпозиции конфигураций ландмарок выполняли методом генерализованного прокрустова анализа с использованием программы MorphoJ 1.06d. Проводили анализ главных компонент и канонический анализ полученных данных. Результаты. Однофакторный дисперсионный анализ выявил высокий уровень межгрупповых различий по показателю прокрустовых расстояний (F=1,34; р<0,0001). Установлено значительное влияние продолжительности нахождения животных в условиях воздействия ФА на размер центроида. Критерий Краскела–Уоллиса составил 19,778 (р=0,0014). Анализ главных компонент указывал на то, что каждая из первых 10 компонент объясняет более 1 % дисперсии прокрустовых координат. При этом первые 7 компонент совместно объясняют 91,398 % изменчивости формы тимуса. Вклад первой главной компоненты в общую дисперсию прокрустовых координат составляет 40,236 %. РС1 (–) показывает изменения формы тимуса, в большей степени затрагивающие верхушки его долей, среднюю часть правого контура и весь левый контур тимуса. Эллипсы рассеивания ординат тимусов, принадлежащих крысам, подвергавшимся влиянию ФА, в пространстве первых двух канонических переменных расположены выше по отношению к таковым контрольных животных. Заключение. Ингаляционное воздействие формальдегида приводит к изменению формы тимуса белых крыс. Наибольшие различия с контрольными данными определяются вдоль второй канонической переменной. The investigation aimed to study the in vitro and in silico antioxidant properties of Melissa officinalis subsp. officinalis essential oil (MOEO). The chemical composition of MOEO was determined using GC–MS analysis. Among 36 compounds identified in MOEO, the main were beta-cubebene (27.66%), beta-caryophyllene (27.41%), alpha-cadinene (4.72%), caryophyllene oxide (4.09%), and alpha-cadinol (4.07%), respectively. In vitro antioxidant properties of MOEO have been studied in 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free-radical scavenging, and inhibition of β-carotene bleaching assays. The half-maximal inhibitory concentration (IC50) for the radical scavenging abilities of ABTS and DPPH were 1.225 ± 0.011 μg/mL and 14.015 ± 0.027 μg/mL, respectively, demonstrating good antioxidant activity. Moreover, MOEO exhibited a strong inhibitory effect (94.031 ± 0.082%) in the β-carotene bleaching assay by neutralizing hydroperoxides, responsible for the oxidation of highly unsaturated β-carotene. Furthermore, molecular docking showed that the MOEO components could exert an in vitro antioxidant activity through xanthine oxidoreductase inhibition. The most active structures are minor MOEO components (approximately 6%), among which the highest affinity for the target protein belongs to carvacrol.

The process of fruit ripening involves many chemical changes occurring not only in the mesocarp but also in the epicarp, including changes in the triterpenoid content of fruit cuticular waxes that can modify the susceptibility to pathogens and mechanical properties of the fruit surface. The aim of the study was the determination of the ripening-related changes in the triterpenoid content of fruit cuticular waxes of three plant species from the Rosaceae family, including rugosa rose (Rosa rugosa), black chokeberry (Aronia melanocarpa var. “Galicjanka”) and apple (Malus domestica var. “Antonovka”). The triterpenoid and steroid content in chloroform-soluble cuticular waxes was determined by a GC-MS/FID method at four different phenological stages. The profile of identified compounds was rather similar in selected fruit samples with triterpenoids with ursane-, oleanane- and lupane-type carbon skeletons, prevalence of ursolic acid and the composition of steroids. Increasing accumulation of triterpenoids and steroids, as well as the progressive enrichment of the composition of these compounds in cuticular wax during fruit development, was observed. The changes in triterpenoid content resulted from modifications of metabolic pathways, particularly hydroxylation and esterification, that can alter interactions with complementary functional groups of aliphatic constituents and lead to important changes in fruit surface quality.
