The effect of lactose and inulin on intestinal morphology, selected microbial populations and volatile fatty acid concentrations in the gastro-intestinal tract of the weanling pig

AbstractTwenty piglets (21 days, 7·8 kg live weight (LW)) were used in a 2×2 factorial to investigate interactions between lactose and inulin on intestinal morphology, microbiology and volatile fatty acid (VFA) production of the weanling pig. The piglets were offered the following diets for 6 days and then sacrificed: (T1) 150 g/kg lactose, (T2) 150 g/kg lactose +15 g/kg inulin, ( T3) 330 g/kg lactose, and ( T4) 330 g/kg lactose +15 g/kg inulin. Tissue samples were taken from the duodenum, jejunum and ileum for morphological measurements. Digesta samples were taken from the ileum, caecum and colon. There was an interaction ( P<0·05) between lactose and inulin in villous height in the jejunum. The inclusion of inulin at 150 g/kg lactose increased villous height compared with 150 g/kg lactose without inulin. However, inulin had no effect on villous height at 330 g/kg lactose inclusion. There was a linear relationship between food intake and villous height in the duodenum (P<0·001, R2=0·45) and the jejunum (P< 0·01, R2=0·25). The inclusion of 330 g/kg lactose increased ( P<0·05) total VFA compared with 150 g/kg lactose in the caecum and the population of lactobacilli in the caecum and colon (P<0·1). There was an interaction (P<0·05) between lactose and inulin for total VFA concentration in the colon. The pigs receiving 330 g/kg lactose had a higher total VFA concentration compared with pigs on 150 g/kg lactose. However, there was no difference between 150 g/kg and 330 g/kg lactose when the diets were supplemented with inulin. In conclusion, the inclusion of high dietary concentrations of lactose resulted in increased lactobacilli and short-chain fatty acid concentrations. The inclusion of inulin with low dietary concentrations of lactose resulted in improved intestinal health through a reduction of intestinal pH and increases in villous height.

1. Twelve steers fitted with rumen cannulas were used in three separate experiments to investigate the effects of the presence or absence of rumen ciliate protozoa on methane production. The diet consisted of 850 g barley and 150 g protein supplement/kg, and was given in three feeds daily at a restricted level of 61 g/kg live weight0.75. Animals were defaunated initially by allowingad lib. consumption of this diet and were then maintained ciliate-free by isolation or were faunated by inoculation with a mixed ciliate suspension. Samples of rumen fluid were taken routinely for the assessment of microbial populations and for volatile fatty acid (VFA) analysis and energy and nitrogen balances and digestibility measurements were made at intervals while animals were confined in respiration chambers.2. In each experiment the rumen VFA proportions changed from a high-propionate pattern under ciliate-free conditions to a low-propionate, high-butyrate pattern in the presence of ciliates: differences between treatments were highly significant (P < 0.001). There were also marked differences between treatments in CH, production but a reliable comparison was possible only in Expt 3, in which CH4was significantly higher (P < 0.001) in the presence of a rumen ciliate population. In Expt 3 the increased loss of energy as CH4in the faunated animals amounted to 44 MJ/100 MJ energy intake.3. Stoichiometric estimates of CH4production derived from the observed VFA proportions showed good agreement with CH4production as measured in respiration chambers. On average, the stoichiometric CH4values overestimated CH4production by a factor of 1.08.4. Highly significant linear relationships (P < 0.001) were observed between the molar proportion of each major VFA and the quantity of CH4produced: the proportion of propionic acid was inversely related to CH4and showed the lowest residual standard deviation of all the relationships examined.5. The losses of energy in faeces and urine did not differ between treatments hence the increased loss of energy as CH4in the faunated animals resulted in a significant reduction in the metabolizability of the diet from 0.73 to 0.69 (P < 0.05). No significant differences were detected between treatments in heat production, apparent digestibility coefficients or N balance.6. It is suggested that the rumen ciliates, by modifying the rumen VFA proportions, are directly responsible for the increased CH4production in faunated animals.

AbstractAn investigation was conducted to test the hypothesis that dietary oil inclusion increases ammonia nitrogen (NH3-N) emission from the manure, due to a negative effect of either unsaturated or saturated oil on microbial activity in the intestine. Dietary oil was included at 45 g/kg, as either crude palm oil (PO), soya oil (SO) or a 50: 50 SO: PO blend (OB), to a basal barley-soya-wheat diet (control) at the expense of dextrose and maize starch. Diets were formulated to contain 13·2 MJ digestible energy per kg and 11·0 g/kg lysine. Four boars were assigned to each dietary treatment, and were transferred to metabolism crates (mean live weight, 74·0 (± 2·89) kg) following 14 days dietary acclimatization. Urine and faeces were collected separately over a 5-day digestibility/nitrogen (N) balance period and a 2-day manure collection period. Inclusion of dietary oil increased apparent digestibility of oil (P < 0·01) and decreased the apparent digestibility of dry matter (P < 0·01) and gross energy (P < 0·05). Dietary oil did not affect the apparent digestibility of nitrogen, acid-detergent fibre, neutral-detergent fibre or hemicellulose. Apparent digestibility of oil was lower when oil was included as PO compared with SO and OB (P < 0·001). The concentration of faecal volatile fatty acid (VFA) was similar in all treatments, although the acetic: propionic acid ratio was lower when oil was included as PO and OB compared with SO (P < 0·05). N balance, manure N concentration and NH3-N emission was not affected by oil inclusion or oil source. In conclusion, dietary oil inclusion reduced apparent dry matter and energy digestibilities, although no effect on fermentation was indicated by digestibility of fibre or faecal VFA content. Consequently, ammonia emission was not affected by dietary oil included at 45 g/kg.

AbstractTwelve Suffolk-Finn Dorset lambs were reared from 25 to 40 or 25 to 55 kg body weight on either pelleted dried grass or a ration of pelleted grass plus barley (ratio 1:1) in a comparative slaughter experiment designed to determine the amounts of total nitrogen and individual amino acids accreted in different body components during growth. Nitrogen (N) balance measurements were determined frequently during this growth phase and accumulated N retentions were compared with the total N accretion determined by comparative slaughter. Total N and individual amino acids accumulated in carcass, wool, skin, offal and blood, head and feet, gastro-intestinal tract and liver were linearly related to body weight in all cases other than for cysteine in carcass. At 25 kg live weight, proportionately 0·52 of total body N was in carcass components, 0·115 in wool, 0·08 in skin, 0·10 in offal and blood, 0·095 in head and feet, 0·06 in the gastro-intestinal tract and 0·02 in liver. However as the animals grew from 25 to 55 kg, 0·256 of the total N accretion was in wool, which was rich in cysteine (98 g/kg total amino acid). Carcass accretion represented only 0·449 of total body N accretion. The N balance technique overestimated net protein accretion by 0·24 (s.e. 0·036).

The current study was conducted to investigate the effect of dietary tributyrin (TB) administration on the intestinal and growth performances in Arbor Acres (AA) broilers under an isocaloric feeding regime. A total of 540 day-old healthy AA broilers were randomly assigned to five treatments with 12 replicates (pens) per treatment and nine birds per pen for 42 days. The dietary treatments were basal diet (control) and basal diet with TB at doses of 0.23 g/kg (TB1), 0.46 g/kg (TB2), 0.92 g/kg (TB3), and 1.84 g/kg (TB4). Particularly, to achieve the isocaloric and cost-saving experimental diets, soybean oil was replaced by the TB product (Eucalorie®) with equivalent metabolic energy contents, and the formulas were rebalanced with zeolite to get the sum of all the feed ingredients to 100%. On days 21 and 42, after weighing, the birds (one bird per replicate) whose body weight was close to the replicate average were euthanized to investigate the effect of dietary TB on intestinal morphology, intestinal bacterial population, and short-chain fatty acid contents. The results revealed that dietary TB administration increased the average daily gain, gain/feed ratio, and European broiler index (P &lt; 0.05) and improved the intestinal morphology (P &lt; 0.05) as indicated by higher villus height and the ratios of villus height/crypt depth in broilers. The incremental levels of TB increased the ileal Lactobacillus content (P = 0.05) and cecal Bacillus content (P = 0.02), respectively. Moreover, dietary TB administration also increased the contents of most of the selected short-chain fatty acids in ileal and cecal digesta (P &lt; 0.05). Collectively, dietary TB administration quadratically improved the growth performance, intestinal morphology, beneficial bacterial population, and short-chain fatty acid levels under the isocaloric feeding regime, indicating better profit return potential in practical poultry operation.
