High levels of variability in immune response using antigens from two reproductive proteins in brushtail possums

Immune-based fertility control is being considered as an effective long-term approach for controlling the pest brushtail possum (Trichosurus vulpecula) population in New Zealand. This relies heavily on the immune response of each immunised possum. A strong and lasting immune response in the majority of individuals in a population is essential. In this study, possums and the model macropod species, the tammar wallaby (Macropus eugenii) were immunised with either a luteinising hormone or androgen receptor synthetic peptide coupled to the carrier molecule KLH (keyhole limpet haemocyanin). The antibody response of wallabies to the antigens was relatively uniform. In contrast, the possum immunoglobulin response to both synthetic peptides and KLH was variable. The apparent high level of variation in the immune response of possums raises questions about the feasibility of using these two antigens to control possum numbers in New Zealand.

Objective: To assess the uses of Community Treatment Orders (CommTOs) in New Zealand. Method: A retrospective study of patients' records held by the regional administrator of mental health legislation and a survey of psychiatrists attending a conference in Dunedin. Results: Males under Community Treatment Orders (CommTOs) outnumbered females 6:4; a high proportion were considered to have a major psychotic disorder; and one fifth remained under a CommTO for more than a year without inpatient care. Among the psychiatrists, there was a high level of agreement that, when used appropriately, the benefits of CommTOs outweigh their coercive impact on the patients; the most strongly supported indicator for use was the promotion of compliance with medication. The rate of use of CommTOs in Otago is remarkably similar to the rate in Victoria, Australia. Conclusions: Records suggest that a significant proportion of patients under CommTOs are not soon readmitted; and many clinicians in New Zealand consider CommTOs to be a useful strategy for managing the community care of long-term patients with schizophrenia and major affective disorders.

To study marsupial developmental mechanisms and their application to fertility control, it is necessary to develop reliable procedures for breeding, colony maintenance, reproductive monitoring for obtaining known-age embryos and, if possible, an induced ovulation protocol. These procedures also provide means to enhance conservation of endangered species. Such procedures are examined in the stripe-faced dunnart, an excellent model for developmental analysis, and the common brush-tail possum, an agricultural and ecological pest species in New Zealand that has become a model for fertility control in marsupials. A long-term colony of the stripe-faced dunnart has been in existence for 21 years, and the procedures for its maintenance and continued survival are outlined, and include minimal reproductive contributions from wild-caught animals, and development of an appropriate timetable of development and induced-ovulation protocols. Common brushtail possum colonies are relatively frequent but have regular input from wild-caught animals. Procedures that minimise competition for prized resources and allow successful group housing of possums are outlined. For both species the available development timetables, in vitro techniques and induced ovulation protocols are essential tools for the study of developmental mechanisms and fertility control, respectively, and also have considerable implications for conservation of these and other species.

ABSTRACT Long term problems associated with future immunological methods of fertility control include the following: (1) reversibility, (2) unscrupulous use, (3) unphysiological intervention giving rise, among other possibilities, to carcinogenesis, (4) deleterious genetical selection operating on antigens, and (5) deleterious genetical selection operating on the immune response. The first three of these constitute a hazard to individuals, and the last two to populations. None is considered to constitute a threat so serious as to inhibit further research on the subject. We should keep under review future developments in relevant areas of immunology, including particularly cell-cell cooperation as a mechanism in autoimmunity, and immune response genes as the objects of immunoselection. It will be important also to come to terms with the social, political, and economic consequences of any cheap, widely-applicable form of birth control, such as might develop from immunological research. The possibility is raised of wide-spread immunisation against a synthetic immunological determinant, as an aid to fertility control. Another possibility is to make women grandchildless.

The article deals with a theoretical model  of the mechanism of the spontaneous regression of tumor, involving the activation of innate immune response. After stimulation the cells of the immune system, use glycolysis for ATP synthesis.  Lactic acid, the final product of glycolysis, acts as inhibitor of such activation. Therefore, highly glycolytic cancerous cells form the environment (hypoglycemia with high level of lactic acid), which is strongly inhibitory to immune cells activation. Starvation, KD (ketogenic diet) and Coley’s toxin induce hypoglycemia and in the long term should also lower lactic acid level. After a few days of such treatment the glucose level could increase naturally due to gluconeogenesis or by the application of significant dose of glucose. Such procedure should induce activation of immune cells response (high glucose and low lactic acid levels) and a spontaneous regression of cancer. If lactic acid concentration remained still too high and thus inhibit the activation of immune cells, the organism would require alkalization because sodium lactate is not inhibitory  to the activation of immune cells. The model explains the cases of spontaneous regression described in literature and could be easily verified  experimentally.

This article describes a high-level evaluation of a scenario exercise that took place in the New Zealand health sector in 1997 and derives some lessons for future evaluations. By extension, such an evaluation tests the efficacy of scenario development and futures thinking (foresight) in general. Context for the evaluation is provided by a brief reflection on scenarios as a technique, both generally and in the health field. Then a discussion of the process used in 1997 to develop five scenarios is followed by a description of the logic and methodology for the evaluation itself. Findings suggest that the process used to develop the 1997 scenarios was valuable in opening up decision-makers’ minds to possibilities without them needing to feel threatened or defensive, but it may not have been inclusive enough for the New Zealand context. Using criteria identified by Schoemaker the scenarios themselves were relevant, credible, and coherent, but not particularly archetypal or long term. Their impact on strategic decision-making was short-lived, but they were prescient in many respects and have been referred to within academia. Future considerations of health futures should be clearer as to purpose, get more explicit buy-in of key decision-makers and draw on a more diverse range of inputs. We also suggest that rather than being carried out during a discrete time period, scenario development should be a continuous and constantly updated process.

This study examined whether individual differences in the vividness of visual imagery influenced performance on a novel long-term change detection task. Participants were presented with a sequence of pictures, with each picture and its title displayed for 17  s, and then presented with changed or unchanged versions of those pictures and asked to detect whether the picture had been changed. Cuing the retrieval of the picture's image, by presenting the picture's title before the arrival of the changed picture, facilitated change detection accuracy. This suggests that the retrieval of the picture's representation immunizes it against overwriting by the arrival of the changed picture. The high and low vividness participants did not differ in overall levels of change detection accuracy. However, in replication of Gur and Hilgard (1975) , high vividness participants were significantly more accurate at detecting salient changes to pictures compared to low vividness participants. The results suggest that vivid images are not characterised by a high level of detail and that vivid imagery enhances memory for the salient aspects of a scene but not all of the details of a scene. Possible causes of this difference, and how they may lead to an understanding of individual differences in change detection, are considered.

Flupropanate (sodium 2,2,3,3 tetrafluoropropanate), a slow-acting lipid bio- synthesis-inhibiting herbicide, was recently registered in New Zealand as Taskforce (745 g/L flupropanate as the sodium salt) for the selective and long-term control of Nassella trichotoma (nassella tussock) in pastures. In five dose-response experiments in permanent hill pastures in Canterbury, conducted between 2012 and 2016, we measured the efficacy of the herbicide against established plants of N. trichotoma and its residual activity against recruiting seedlings. Mortality, as an average across the five sites, was 93% 1.5 years after applying 1.49 kg flupropanate/ha (the label-recommended rate), and 100% at 2.98 kg/ha. This indicates that an application rate higher than the label rate will be necessary for complete control of a N. trichotoma infestation. The presence of 1,000 and 6,250 visible seedlings of N. trichotoma/ha in the autumn 3.2 and 2.1 years after applying 1.49 kg flupropanate/ha (at a Greta Valley and Scargill site respectively) indicates that the herbicide’s soil residues had decayed within 12 months to a concentration lower than necessary to kill the germinating seedlings of N. trichotoma.

BACKGROUND Approximately 10% to 12% of New Zealand children and young people have long-term physical conditions (also known as chronic illnesses) and are more likely to develop psychological problems, particularly anxiety and depression. Delayed treatment leads to worse physical and mental healthcare, school absence, and poorer long-term outcomes. Recently, electronic health (eHealth) interventions, especially those based on the principles of Cognitive Behavior Therapy (CBT), have been shown to be as good as face-to-face therapy. Biofeedback techniques have also been shown to enhance relaxation during the treatment of anxiety. However, these modalities have rarely been combined. Young people with long-term physical conditions have expressed a preference for well-designed and technologically-based support to deal with psychological issues, especially anxiety. OBJECTIVE This study aimed to co-design and evaluate the (i) acceptability and (ii) usability of a CBT and biofeedback-based, 5-module eHealth game called ‘Starship Rescue’ and (iii) to provide preliminary evidence regarding its effectiveness in addressing anxiety and quality of life in young people with long-term physical conditions. METHODS Starship Rescue was co-designed with children and young people from a tertiary hospital in Auckland, New Zealand. Following this, 24 young people aged 10 to 17 years were enrolled in an open trial, during which they were asked to use the game for an 8-week period. Acceptability of the game to all participants was assessed using a brief, open-ended questionnaire, and more detailed feedback was obtained from a subset of 10 participants via semi-structured interviews. Usability was evaluated via the System Usability Scale (SUS) and device-recorded frequency and duration of access on completion of the game. Anxiety levels were measured prior to commencement, on completion of the game, and 3 months later using the Generalized Anxiety Disorder 7-item scale (GAD-7) and Spence Child Anxiety Scales (SCAS), and at the start of each module and at the end of the game using an embedded Likert/visual analog scale. Quality of life was measured prior to commencement and on completion of the game using the Pediatric Quality of Life Scale (PEDS-QL). RESULTS Users gave Starship Rescue an overall rating of 5.9 out of 10 (range 3-10 and a mean score of 71 out of 100 (SD 11.7; min 47.5; max 90) on the System Usability Scale (SUS). The mean time period for use of the game was just over 11-weeks (78.8 days, 13.5 hours, 40 minutes). Significant reductions in anxiety were noted between the start and end of the game on the GAD-7 (-4.6 (p=0.000)), SCAS (-9.6 (p=0.005)), and the Likert/visual analogue scales (-2.4 (p=0.001)). Quality of life also improved on the PedsQL scale (+4.3 (p=0.042)). All changes were sustained at 3-month follow-up. CONCLUSIONS This study provides preliminary evidence for Starship Rescue being an acceptable, usable and effective eHealth intervention for addressing anxiety in young people with long-term physical conditions. Further evaluation is planned via a more formal randomized controlled trial.
