520. RELATIONSHIP BETWEEN THE OVARIAN STATUS ON THE INITIATION DAY OF SUPEROVULATION AND THE OVULATORY FOLLICLES AT ESTRUS IN SHALL IRANIAN EWES

The objective of this experiment was to determine the influence of the ovarian status on the initiation day of superovulation (day 14) on superovulatory response in terms of the number of ovulatory follicles (F, ≥4mm) at estrus (day 16). Estrous cycles of 50 Shall Iranian ewes were synchronized by CIDR. The day of CIDR insertion was considered as the starting point of the experiment (day 0). At the time of CIDR removal (day 14), 650 IU of eCG was administered to ewes. Ultrasonography was performed on days 14 and 16. Data were analyzed by means of the GLM procedure of the SAS. In these cases, the number of follicles were determined in each of the three studied classes (<2, 2 to 4, and ≥4 mm) on day 14. Subsequently, each of these classes was divided into two subgroups (Low, High) based on the number of follicles observed on day 14 (Table 1). The number of large follicles at estrus in each of the three studied classes were (3.96±0.28 vs. 4.78±0.26, p<0.05; 4.31±0.35 vs. 4.41±0.24, p>0.05; and 4.50±0.23 vs. 4.07±0.37, p>0.05) respectively. The number of medium and large follicles on day 14 did not affect the superovulatory response. However, the high number of small follicles on the initiation day of superovulation increased the number of ovulatory follicles at estrus. Since, 23 out of 50 ewes had a low number of small follicles on day 14 (Table 1), which may suggest that an injection of FSH 1-2 days before initiation of eCG treatment enhance the superovulatory response.

Introduction: The kisspeptin gene Kiss1 is expressed in two hypothalamic areas: anteroventral periventricular nucleus/periventricular nucleus (AVPV/PeN) and arcuate nucleus (ARC), and also in gonads. Evidences suggest that gamma-amino butyric acid B receptors (GABAB) signaling can regulate Kiss1 expression. Here we inhibited GABAB signaling from PND2-PND21 and evaluated the hypothalamic-pituitary-gonadal (HPG) axis. Methods: BALB/c mice were treated on postnatal days 2-21 (PND2-PND21) with CGP55845 (GABAB antagonist), and evaluated in PND21 and adulthood: gene expression (qPCR) in hypothalamus and gonads, hormones by radioimmunoassay, gonad histochemistry (H&E), puberty onset, estrous cycles. Results: At PND21, CGP inhibited Kiss1 and Tac2 and increased Pdyn and Gabbr1 in the ARC of both sexes and decreased Th only in female AVPV/PeN. Serum follicle-stimulating hormone (FSH) and testis weight decreased in CGP-males and puberty onset was delayed. In adults, Kiss1, Tac2, Pdyn, Pgr, Cyp19a1, Gad1 were downregulated, while Gabbr1 was upregulated in the ARC of both sexes. In the AVPV/PeN, Kiss1, Th, Cyp19a1 and Pgr decreased while Gad1 increased in CGP-females, whereas Cyp19a1 increased in CGP-males. Serum FSH increased in CGP-males while prolactin increased in CGP-females. Testosterone and progesterone increased in ovaries from CGP-females, in which Kiss1, Cyp19a1 and Esr1 were downregulated while Hsd3b2 was upregulated, together with increased atretic and decreased ovulatory follicles. Testes from CGP-males showed decreased progesterone, increased Gabbr1, Kiss1, Kiss1r, Esr2 and decreased Cyp19a1 and clear signs of seminiferous tubules atrophy. Conclusion: These results demonstrate that appropriate GABAB signaling during this critical prepubertal period is necessary for the normal development of the HPG axis.

Follicular dynamics of Zebu Cattle are characterized by lower maximum diameter and persistence of dominant follicles, when compared to European breeds. However, the main effects of follicular dominance on subordinated follicle population, and consequently on oocyte recovery and embryo production, have been shown to be similar among breeds. The aim of this study was to evaluate the dominant follicle establishment period, indicated by follicle divergence, in Zebu cows. Pluriparous, non-lactating Guzera breed (Bos indicus) cows (n=5), presenting a mean body weight of 518.0±48.5kg and similar body score condition (ranging from 3.5 to 4, in a 1 to 5 scale), were used. In a first trial, follicular dynamics were evaluated daily during 1 (n=2) or 2 (n=3) estrous cycles, using a portable ultrasound device equipped with a linear rectal 5MHz transducer (Aloka SSD500, Aloka Co., Tokyo, Japan). In a second trial, estrous cycles were synchronized using progesterone intravaginal-releasing devices (CIDR, Pharmacia, Sao Paulo, Brazil), and follicular dynamics were evaluated every 6h after ovulation, during the first follicular wave, until the largest follicle achieved a diameter of 10mm. During each evaluation ovaries were mapped, and evolution of follicles followed individually. Data are normalized for the moment of follicular divergence, and analyzed by ANOVA. Results are presented as mean±SD. Data from the first evaluations confirmed the occurrence of cycles with three (4/8) and four (1/8) follicular waves, despite a mean cycle length of 19.1±1.9 days, with a maximum diameter of 11.6±2.4mm for non-ovulatory follicles, and 14.4±0.5mm for ovulatory follicles. Follicular deviation during the first follicular wave occurred 49.2±5.0h after ovulation, when the largest follicle presented a diameter of 7.3±0.6mm, and the second largest follicle 6.8±0.7mm. Growth rates of the dominant and the largest subordinated follicles were similar before (0.35mm/6h vs. 0.33mm/6h, respectively;; P&gt;0.05), but significantly different after deviation (0.52mm 6h v. −0.04mm 6h; P&lt;0.05). These results shows that the lower maximum diameter and persistence of dominant follicles, in Zebu cattle, is partially compensated by an earlier dominant follicle deviation.

Wapiti are seasonally polyestrous. The transition into and out of the breeding season is marked by resumption of ovulation in autumn and cessation of ovulation in winter. Onset of ovulatory cyclicity is distinct and associated with aggressive breeding behavior of stags in rut. Cessation of ovulation at the end of the breeding season is not distinguished by behavioral patterns. The objective of the present study was to characterize follicular and luteal dynamics in wapiti during the transitional periods into and out of the breeding seasons. Transition from anestrus to estrus was monitored in 15 hinds, aged 2 to 14 years, over two successive seasons (11 in year 1, with 5 hinds from year 1 used again in year 2 along with 4 new hinds; n = 20 observations). Transition from estrus to anestrus was monitored in 11 hinds over 1 season (n = 11 observations). Hinds were maintained on a farm near Saskatoon, Saskatchewan (52°07′N, 106°38′W). The ovaries were examined daily during September through October by transrectral ultrasonography using a B-mode ultrasound machine and a 7.5 MHz linear array transducer for transition to estrus, and December through April for transition to anestrus. The first ovulation was recorded on September 15 and all hinds had ovulated for the first time by October 7. In 17 of 20 observations, the duration of the first interovulatory interval (IOI) was 9.3 ± 0.4 days (mean ± SEM). With one exception, these IOIs were characterized by one wave of follicular development. The remaining three IOIs ranged from 16 to 23 days and consisted of two or three waves of follicle development. The second ovulation occurred by October 15 in hinds with a short IOI and by October 17 in all remaining hinds. The mean dates of first and last ovulation were September 25 and February 7, respectively, an interval of 135 days. The median date of the last ovulation was February 15 and the range was from December 3 to March 22. Duration of the last IOI of the season (21.2 ± 0.6 days) was similar to the notional 21-day cycle for wapiti, but longer (P < 0.05) than the duration of the first IOI (10.9 ± 1.0 days). Maximum diameters of the first 2 ovulatory follicles were similar (11.3 ± 0.4 vs. 11.3 ± 0.2 mm), but were larger (P < 0.05) than the last 2 ovulatory follicles of the breeding season (10.3 ± 0.3 vs. 10.1 ± 0.4 mm). Maximum diameter of the corpus luteum (CL) tended (P = 0.06) to be smaller for the short IOI than for longer IOI of the first and last cycles. Diameter of the last CL of the season was not different from that of the previous CL (12.8 ± 0.6 vs. 12.5 ± 0.6 mm); however, it was detected for a longer period (22.3 ± 1.2 vs. 19.3 ± 0.7 days; P < 0.05). Estrous cycles during transition into the breeding season have been described as being irregular and those out of the breeding season as increasingly long. In the present study, the transition periods were characterized by regular events. Transition to regular estrous cycles was preceded by one short (9 days) IOI. The last IOI of the breeding season was the same as that reported during the rut. Transition to anestrus occurred most commonly in February and was marked by a failure of the dominant follicle to ovulate after luteal regression.

The purpose of this experiment was to determine the influence of the ovarian status on the initiation day of superovulation (d 0) on superovulatory responses in terms of the number of large follicles (≥7mm) at estrus (d 4) and CL 7 days after estrus (d 11). Ultrasonography was performed on d 0, 4 and 11of the experiment. Animals conventionally superovulated (400 mg Folltropin-V) between d 8 to 12 of the estrous cycles and 102 superovulation cycles were considered in the present experiment. Data were analyzed using GLM procedure of the SAS. In these cases, the number of follicles was determined in each of the 2 studied classes (SF, 4 to 6 mm; and LF, ≥7mm) on d 0. Subsequently, each of these classes was divided into two subgroups (Low, High) based on the number of follicles observed on d 0 (Table 1). The number of LF at estrus in each of the 2 studied classes (Low vs. High) were (13.02±1.02 vs. 19.7±1.69, p<0.01; and 16.23±1.59 vs. 14.75±1.48, p>0.1) and the number of CL on d 11 were (9.75±0.57 vs. 12.75±0.98, p<0.01; 10.91±0.8 vs. 10.85±0.65, p>0.1). Therefore, the ovarian status on d 0 in terms of the number of SF affected the number of LF (d 4) and CL (d 11). In addition, the number of superovulatory cycles with low number of SF was 2 times more than the ones with a high number of SF on d 0; also 49 out of 102 superovulatory cycles had at least 2 LF on the initiation day of superovulation (Table 1). These results suggested that follicular puncture 1–2 days before a conventional superovulation treatment may increase the superovulatory responses.

Metallic clusters are interesting from various points of view, e.g. as a mean of spreading expensive catalysts on a support, or following heterogeneous and homogeneous catalytic events. It is also possible to study nucleation and growth mechanisms for crystals with the cluster as known starting point.Gold-clusters containing 55 atoms were manufactured by reducing (C6H5)3PAuCl with B2H6 in benzene. The chemical composition was found to be Au9.2[P(C6H5)3]2Cl. Molecular-weight determination by means of an ultracentrifuge gave the formula Au55[P(C6H5)3]Cl6 A model was proposed from Mössbauer spectra by Schmid et al. with cubic close-packing of the 55 gold atoms in a cubeoctahedron as shown in Fig 1. The cluster is almost completely isolated from the surroundings by the twelve triphenylphosphane groups situated in each corner, and the chlorine atoms on the centre of the 3x3 square surfaces. This gives four groups of gold atoms, depending on the different types of surrounding.

Root extracts of a Cameroon medicinal plant, Dorstenia psilurus, were purified by screening for AMP-activated protein kinase (AMPK) activation in incubated mouse embryo fibroblasts (MEFs). Two isoprenylated flavones that activated AMPK were isolated. Compound 1 was identified as artelasticin by high-resolution electrospray ionization mass spectrometry and 2D-NMR while its structural isomer, compound 2, was isolated for the first time and differed only by the position of one double bond on one isoprenyl substituent. Treatment of MEFs with purified compound 1 or compound 2 led to rapid and robust AMPK activation at low micromolar concentrations and increased the intracellular AMP:ATP ratio. In oxygen consumption experiments on isolated rat liver mitochondria, compound 1 and compound 2 inhibited complex II of the electron transport chain and in freeze–thawed mitochondria succinate dehydrogenase was inhibited. In incubated rat skeletal muscles, both compounds activated AMPK and stimulated glucose uptake. Moreover, these effects were lost in muscles pre-incubated with AMPK inhibitor SBI-0206965, suggesting AMPK dependency. Incubation of mouse hepatocytes with compound 1 or compound 2 led to AMPK activation, but glucose production was decreased in hepatocytes from both wild-type and AMPKβ1−/− mice, suggesting that this effect was not AMPK-dependent. However, when administered intraperitoneally to high-fat diet-induced insulin-resistant mice, compound 1 and compound 2 had blood glucose-lowering effects. In addition, compound 1 and compound 2 reduced the viability of several human cancer cells in culture. The flavonoids we have identified could be a starting point for the development of new drugs to treat type 2 diabetes.

Purpose This clinical focus article provides an overview of clinical models currently being used for the provision of comprehensive aural rehabilitation (AR) for adults with cochlear implants (CIs) in the Unites States. Method Clinical AR models utilized by hearing health care providers from nine clinics across the United States were discussed with regard to interprofessional AR practice patterns in the adult CI population. The clinical models were presented in the context of existing knowledge and gaps in the literature. Future directions were proposed for optimizing the provision of AR for the adult CI patient population. Findings/Conclusions There is a general agreement that AR is an integral part of hearing health care for adults with CIs. While the provision of AR is feasible in different clinical practice settings, service delivery models are variable across hearing health care professionals and settings. AR may include interprofessional collaboration among surgeons, audiologists, and speech-language pathologists with varying roles based on the characteristics of a particular setting. Despite various existing barriers, the clinical practice patterns identified here provide a starting point toward a more standard approach to comprehensive AR for adults with CIs.

Abstract: We review research on response-latency based (“implicit”) measures of attitudes by examining what hopes and intentions researchers have associated with their usage. We identified the hopes of (1) gaining better measures of interindividual differences in attitudes as compared to self-report measures (quality hope); (2) better predicting behavior, or predicting other behaviors, as compared to self-reports (incremental validity hope); (3) linking social-cognitive theories more adequately to empirical research (theory-link hope). We argue that the third hope should be the starting point for using these measures. Any attempt to improve these measures should include the search for a small-scale theory that adequately explains the basic effects found with such a measure. To date, small-scale theories for different measures are not equally well developed.

Abstract: Solidarity in the classic sense pertains to a cohesion among humans that entails physical contact, shared emotions, and common goals or projects. Characteristic cases are to be found among families, close friends, or co-workers. The present paper, in contrast, treats a phenomenon of the solidarity of distance, a solidarity based in fear of certain others and in incompetence to interact with them. The starting point for this analysis is the person who is motivated to interact with others who are unfamiliar or fear-provoking. Given that the fear and momentary social incompetence do not allow a full interaction to ensue, the individual will move toward solidarity with those others on a symbolic level. In this manner the motivation to approach the others is acted upon while physical and emotional distance is retained.
