Prediction of cadmium concentrations in potato tubers (Solanum tuberosum L.) by pre-plant soil and irrigation water analyses

Potato tubers can accumulate high concentrations of cadmium (Cd) in edible portions, so that techniques to determine high risk Cd environments are required by growers. The use of combined soil and irrigation water analyses prior to crop planting was investigated as a means to predict risks of Cd accumulation in tubers. Soils and irrigation waters were analysed at 134 sites in the major potato production areas in Western Australia, South Australia, Tasmania, Victoria, and New South Wales. Irrigation waters were analysed for electrical conductivity (EC), major cations, and anions. Cadmium was extracted from soil using aqua regia (1 : 3 HNO3: HCl), EDTA (ethylenediamine-N,N,N′,N′-tetraacetate), DTPA (diethylene-triamine-pentaacetate), 0·01 M CaCl2, 0·01 M Ca(NO3)2, 0·1 M CaCl2, and 1·0 M NH4NO3. The preferred test procedure was validated in a subsequent sampling and analysis program at 39 sites. Irrigation water quality (EC or Cl concentration), measured prior to planting, explained the greatest variation in tuber Cd concentrations. Of the soil test procedures, only Cd extracted by 0·01 M CaCl2 significantly improved the predictive capacity of water EC. These 2 measures explained >55% of the variance in tuber Cd concentrations. The data set were transformed to generate a probability curve for exceeding Cd concentrations of either 0·05 or 0·1 mg/kg fresh weight, the latter being the current maximum permitted concentration (MPC) in Australia for potato tubers. The probability of producing potato tubers exceeding 0·05 and 0·1 mg/kg fresh weight was >50% once irrigation water EC increased above 1·4 and 3·0 dS/m, respectively. Using the relationships developed, growers should be able to quantify Cd risks by a simple test of irrigation water EC prior to planting and, if further precision is needed, also determine CaCl2-extractable Cd in soil.

Alkalinity of irrigation water affects pH of the plant growing media. High alkalinity water raises media pH and affects nutrient availability to plants and, thus, plant growth. Alkalinity levels in irrigation water vary in different regions. Knowledge of alkalinity levels would help growers and advisors in modifying cultural practices to suit the levels. To find out how the levels vary in different states, we searched our database of thousands of water analyses from across the country. Overall, 38% of the waters had <100 ppm CaCO3, a level not considered to be a concern for even small pots. About 74% of the waters had <200 ppm CaCO3, a level considered to be safe for large pots. The majority of the waters in the states along the eastern seaboard had <150 ppm CaCO3. In waters in the interior states, alkalinity levels varied. Pacific Northwest had most waters <150 ppm CaCO3. Detailed results—bar charts showing percent of waters at different alkalinity levels in each state and in the country will be shown.

SummaryThe present study concerned the reproducibility of the so-called prothrombin time as assessed with a series of more commonly used modifications of the Quick’s onestage assay procedure, i.e. the British comparative reagent, homemade human brain thromboplastin, Simplastin, Simplastin A, and Thrombotest. All five procedures were tested manually on pooled lyophilized normal and patients’ plasmas. In addition, Simplastin A and Thrombotest were investigated semiautomatically on individual freshly prepared patients’ plasmas. From the results obtained, the following conclusions may be drawn :The reproducibility of results obtained with manual reading on lyophilized plasmas is satisfactory for all five test procedures. For Simplastin, the reproducibility of values in the range of insufficient anticoagulation is relatively low due to the low discrimination power of the test procedure in the near-normal range (so-called low sensitivity of rabbit brain thromboplastins). The reproducibility of Thrombotest excels as a consequence of its particularly easily discerned coagulation endpoint.The reproducibility of Thrombotest, when tested on freshly prepared plasmas using Schnitger’s semiautomatic coagulometer (a fibrinometer-liJce apparatus), is no longer superior to that of Simplastin A.The constant of proportionality between the coagulation times formed with Simplastin A and Thrombotest was estimated at 0.64.Reconstituted Thrombotest is stable for 24 hours when stored at 4° C, whereas reconstituted Simplastin A is not.The Simplastin A method and Thrombotest seem to be equally sensitive to “activation” of blood coagulation upon storage.

ABSTRACT Cauliflower is a brassica produced and consumed in Brazil, whose cultivation depends on the adequate supply of water and nutrients. The objective of this study was to evaluate the effect of irrigation depths and nitrogen doses on the production components and water yield of cauliflower hybrid Barcelona CMS. The treatments consisted of five irrigation water depths (0, 75, 100, 125 and 150% of the crop evapotranspiration) combined with five nitrogen doses (0, 75, 150, 300 and 450 kg ha-1). The experiment was conducted in a completely randomized design with a split-plot arrangement. The effects of these factors were evaluated using the response surface methodology. The water yield of the crop decreases with increasing irrigation water depth; therefore, the yield is higher when water replenishment is lower than the recommended. The highest estimated total inflorescence yield is 24,547.80 kg ha-1, with a inflorescence mean diameter of 19.60 cm, a inflorescence mean height of 12.25 cm, and an inflorescence fresh weight of 858.90 g plant-1, obtained with an irrigation water depth equivalent to 132.09% of the crop evapotranspiration (ETc) and a nitrogen dose of 450 kg ha-1. The highest inflorescence diameter and height are obtained with an irrigation depth equivalent to 128.70 and 108.20% of ETc, respectively, and a nitrogen dose of 450 kg ha-1. Therefore, the best productivity response of the Barcelona CMS cauliflower hybrid can be obtained using an irrigation depth greater than the crop evapotranspiration, regardless of the nitrogen doses.

Up to 25 proteins have been found in the sap of potato tubers by polyacrylamid electrophoresis. The mode of preparation and concentration of the plant material was investigated as well as the pretreatment of the polyacrylamid gel to avoid undue retardation of the migrating bands.Sulfite was the most active inhibitor of sap-browning compared with ascorbic acid and cysteine. which did not influence the protein pattern except for one line. The juice had to be dialyzed or filtered through Sephadex G 25 to remove an unkown acidic material which interferes with the more acidic proteins and causes spreading and diffusing of bands. Proteins were concentrated by means of Carbowax MW 20000. For best results ionic contaminations inherent in the gels was removed by electrophoresis before applying the proteins or dyes. Very few proteins were affected by decreasing the pore size of the gel. Ammonium sulfate (40% saturation) precipitated mainly those proteins which are similar in electrophoretic behavior to the albumin- and transferrinregion of human plasma.

Potato (Solanum tuberosum L.) has various biotic constraints in its production due to pest attack. Among these, common scab caused by streptomyces scabies is an important disease in potato which causes economic loss with respect to plant yield and quality of tubers. The present study was conducted to determine the pathogenicity, pathogenic variation, characterization of morphological, physiological and biochemical aspects of Streptomyces associated with potato tubers grown in Rawalpindi district. Severity data and pathogenic variation of disease was studied by using different isolation and characterization techniques.

Haemorrhagic fever caused by the Ebola virus is a highly hazardous infectious disease with a mortality rate of 50– 90 %. Heterologous immunoglobulins with a high virus-neutralizing titer are an important element of the WHO-endorsed set of measures for emergency prevention and treatment of the disease. Specific activity of these products is largely determined by their fractional composition, and, in particular, by molecular mass distribution (MMD). The size-exclusion-high-performance liquid chromatography (SEC-HPLC) has traditionally been used for determination of the MMD of the target protein in human immunoglobulin-based products. The use of this method for evaluation of molecular parameters of heterologous immunoglobulin requires confirmation of its specificity, accuracy and precision, and establishment of the chromatographic system suitability criteria in the context of a new test object.The aim of the study was to test the applicability of the SEC-HPLC method to the assessment of molecular parameters of anti-Ebola immunoglobulin derived from horse serum.Materials and methods: three batches of purified equine anti-Ebola immunoglobulin were used in the study. Normal equine and human immunoglobulins of the IgG isotype were used as reference standards. The HPLC test procedures described in the European Pharmacopoeia 9.6 and State Pharmacopoeia of the Russian Federation, 14th ed., were used for determination of monomers and other immunoglobulin fractions. An Agilent 1260 Infinity (Agilent, USA) HPLC system with a diode array detector and an Agilent Bio SEC-3 HPLC column were used for quality evaluation of the tested products.Results: the resolution factor between IgG monomer and dimer peaks (1.69 and 2.10), and the chromatographic column efficiency (>2000) make it possible to use the SEC-HPLC system for evaluation of molecular parameters of heterologous immunoglobulin. The study demonstrated reproducibility of the test procedure.Conclusions: the study confirmed the applicability of the SEC-HPLC procedure for evaluation of molecular parameters of anti-Ebola immunoglobulin derived from horse serum. It demonstrated the compliance of the purified immunoglobulin to the national and international quality requirements in terms of «Molecular parameters».
