Because of the severe decline in grey wolf (Canis lupus) abundance, preservation of genetic diversity within the species is essential to prevent inbreeding depression. Cryopreservation is a valuable tool in endangered species conservation, because it allows genetic material to be rescued from rare individuals for future use. The ability to preserve gonadal tissue is particularly useful in seasonal breeders, such as grey wolves, when mature gametes are not available during the non-breeding season. Optimization of species-specific cryopreservation protocols is necessary due to the toxicity of cryoprotectants. Here, we aimed to investigate three protocols to cryopreserve grey wolf testicular tissue, as a model for endangered canid species. Testicular tissue samples from four adult grey wolves were obtained and cryopreserved using slow-freezing (SF) and needle vitrification (nVT) techniques. Slow-frozen samples were exposed to 15% dimethylsulfoxide (DMSO) or 7.5% DMSO + 7.5% ethylene glycol (EG) in minimum essential medium (MEM) at room temperature, placed in a Mr. Frosty Freezing Container (Thermo Fisher Scientific) and cooled to −80°C overnight, before storage in liquid nitrogen. Needle-vitrified samples were equilibrated in a 7.5% DMSO + 7.5% EG solution in MEM for 10min at 4°C, then treated with 15% DMSO + 15% EG + 0.5M sucrose for 10min at 4°C before plunging into liquid nitrogen. Each sample was thawed using serial dilutions of sucrose (1, 0.5, and 0.25) into MEM at 37°C for 5min each, fixed in 4% paraformaldehyde, and embedded in paraffin wax. Five-micron-thick cross-sections of tissue samples were stained using haematoxylin and eosin. Tubule areas and quantity of each cell type (Sertoli cells, spermatogonia, spermatocytes, spermatids, and abnormal cells) were recorded and compared with a fresh control group for each wolf. Cell populations and tubule areas were analysed using a nonparametric Wilcoxon test through JMP statistical software (SAS Institute Inc.), and are presented as means±standard deviations. There was no difference in tubule areas between the treatment groups (fresh=5200±2870, nVT=5655±2370, SF DMSO=3607±814, SF DMSO+EG=3905±1450µm2; P>0.05). Although there was an increase in abnormal cells/tubule area (µm2) for nVT, SF DMSO, and SF DMSO+EG compared with the fresh samples (0.0015±0.0014, 0.0014±0.0013, 0.0016±0.0011, and 0.0007±0.0004, respectively; P>0.05), differences were not statistically significant. Moreover, the number of Sertoli cells, spermatogonia, and spermatocytes were reduced in the cryopreserved groups compared with fresh tissues, but the differences were not statistically different (P>0.05). The data suggest that the three tested protocols preserved testicular structure and cell populations of cryopreserved testicular tissue and could be useful in the genetic preservation of grey wolves.

Background: Hyperglycemia is a common effect of uncontrolled DM. The main impact of DM on male infertility is the result of hyperglycemia which causes an increasing in free radicals which cause oxidative stress in the testes, and then attacks Sertoli cells. Increasing of free radicals will cause disruption and damage to this cell, including apoptosis and atrophy and then caused decreasing in the number of Sertoli cells. Kebar grass contains antioxidants compounds, such as flavonoids and vitamin E. There are synergy effects between vitamin E and flavonoids which reinforce both in working to improve infertility as an antioxidant. Objective: To compared the number of Sertoli cells between groups which receiving extract kebar grass (Biophytum petersianum Klotzsch) and the control group in diabetes mellitus mice (Mus musculus) model. Methods: The research subjects used DM male mice which were divided into 5 groups: 2 control groups (K- and K+) and 3 treatment groups (P1, P2, P3). Day 1 to day 5, all groups were induced by STZ. The 11th day to the 45th day, the control group (K+) is given metformin 2 mg/head /day, the P1, P2 and P3 groups is given kebar grass with dose of 67,5; 135 and 270 mg/kg/day and metformin 2 mg/head/day. The 11th day to the 31th day, the control group (K-) is given CMC-Na 1% suspension. The 46th day is done surgery and sampling testicular tissue. Examination of the number of Sertoli cells is done through examination of the histological picture of testicular tissue that has been given Haematoxylin-Eosin (HE) staining. Results: Statistical tests showed that there was significant differences with a value of p = 0.000 (<0.05) in the number of Sertoli cells. Mean and standard deviation of Sertoli cells in each group were K- = 8.63 ± 0.50; K+ = 9.87 ± 1.52; P1 = 11.40 ± 0.77; P2 = 14.75 ± 1.97 and P3 = 14.97 ± 2.00. Conclusion: Kebar grass extract can maintain the number of Sertoli cells in diabetes mellitus mice models. Tujuan: Membandingkan jumlah sel Sertoli mencit (Mus musculus) model diabetes melitus antara kelompok yang mendapat ekstrak etanol rumput kebar (Biophytum petersianum Klotzsch) dan kelompok kontrol. Bahan dan cara: Subyek penelitian menggunakan mencit jantan DM, dibagi 5 kelompok yaitu 2 kelompok tatist (K- dan K+) dan 3 kelompok perlakuan (P1, P2, P3). Hari ke-1 sampai hari ke-5, semua kelompok diinduksi STZ. Hari ke-11 sampai hari ke-45, kelompok tatist (K-) disonde tatisti CMC-Na 1%, kelompok tatist (K+) disonde metformin 2 mg/ekor/hari, kelompok P1, P2 dan P3 disonde rumput kebar dosis 67,5, 135, 270 mg/kg BB/hari dan metformin 2 mg/ekor/hari, Hari ke – 46 dilakukan pembedahan dan pengambilan sampel jaringan testis kanan dan kiri. Pemeriksaan jumlah sel Sertoli dilakukan melalui pengamatan gambaran histologi jaringan testis yang telah diberikan pewarnaan Haematoxylin-Eosin (HE). Hasil: Uji statistik menunjukkan bahwa terdapat perbedaan bermakna dengan nilai p = 0.000 (< 0,05) pada jumlah sel Sertoli. Mean dan standar deviasi sel Sertoli pada tiap kelompok adalah K- = 8,63 ± 0,50; K+ = 9,87 ± 1,52; P1 = 11,40 ± 0,77; P2 = 14,75 ± 1,97 and P3 = 14,97 ± 2,00. AbstractIntroduction: Radioactive iodine (RAI) is commonly used for the treatment of hyperthyroidism caused by Graves’ disease or thyroid nodules. However, information available on the impact of RAI therapy on male gonadal function is scarce. This study aimed to determine any possible damage to testicular tissue and sperm quality caused by RAI therapy, and the radioprotective effect of amifostine against such damage.Material and Methods: In total, 36 rats were randomly allocated to three groups, including a control group, RAI group (111 MBq Iodine-131), and RAI + amifostine group (111 MBq Iodine-131 and a single dose of 200 mg/kg amifostine). Blood and epididymal sperm samples were taken for hormone analyses and the evaluation of spermatological parameters. The TUNEL assay and haematoxylin-eosin were used to stain testicular tissue samples to detect histological changes and apoptosis.Results: The groups differed insignificantly for the testicular mass index and spermatozoa concentration. However, spermatozoa motility and percentage of viable spermatozoa were higher in the RAI + amifostine group, compared to the RAI group. Sperm DNA fragmentation and the index of apoptotic germ cells significantly decreased in the amifostine group, in comparison to the radioiodine group. While the testosterone levels showed no significant change, the follicle stimulating hormone (FSH) levels significantly decreased in the RAI + amifostine group.Conclusion: All histopathological parameters and some spermatological parameters showed that RAI therapy caused statistically significant damage of testicular tissue and this damage was reduced by amifostine.

Abstract Background Androgens are well known to be necessary for spermatogenesis. The purpose of this study was to determine Sertoli cell responsiveness to androgens according to age from birth to puberty. Results Testicular tissue samples were studied in a population of 84 control boys classified into seven groups according to age: group 1 (1–30 days), group 2 (1–3 months), group 3 (3–6 months), group 4 (0.5–3 years), group 5 (3–6 years), group 6 (6–12 years), and group 7 (12–16 years). We compared these data with those of 2 situations of pathology linked to androgens: 1/premature secretion of testosterone: 4 cases of Leydig cell tumor (LCT) in childhood; and 2 /defect of androgen receptors (AR): 4 cases of complete form of insensitivity to androgen syndrome (CAIS). In control boys, AR immunoreactivity (ir) in Sertoli cells appeared between 4.6 and 10.8 years of age, Anti-Mullerian Hormone (AMH) ir in Sertoli cells disappeared between 9.2 and 10.2 years of age. Connexin 43 (Cx43) ir in Sertoli cells and histological features of the onset of spermatogenesis appeared between 10.8 and 13,8 years of age. Cx43 ir was significantly higher in 12–16 year-olds than in younger boys. In case of CAIS, no spermatogenesis was observed, both AR and Cx43 ir were undetectable and AMH ir was elevated in Sertoli cells even at pubertal age. In the vicinity of LCTs, spermatogenesis occurred and both AR and Cx43 ir were strongly positive and AMH ir in Sertoli cells was low for age. Conclusions Androgen action on Sertoli cells is required for onset of spermatogenesis and premature androgen secretion by LCT can induce spermatogenesis in the vicinity of the tumor. AR ir appeared earlier than onset of spermatogenesis, with large interindividual variability. The timing and mechanisms of Sertoli cell responsiveness to androgens are important issues for understanding the induction of spermatogenesis at puberty.

At present most medical microprobe analysis is conducted on insoluble particulates such as asbestos fibers in lung tissue. Cryotechniques are not necessary for this type of specimen. Insoluble particulates can be processed conventionally. Nevertheless, it is important to emphasize that conventional processing is unacceptable for specimens in which electrolyte distributions in tissues are sought. It is necessary to flash-freeze in order to preserve the integrity of electrolyte distributions at the subcellular and cellular level. Ideally, biopsies should be flash-frozen in the operating room rather than being frozen several minutes later in a histology laboratory. Electrolytes will move during such a long delay. While flammable cryogens such as propane obviously cannot be used in an operating room, liquid nitrogen-cooled slam-freezing devices or guns may be permitted, and are the best way to achieve an artifact-free, accurate tissue sample which truly reflects the in vivo state. Unfortunately, the importance of cryofixation is often not understood. Investigators bring tissue samples fixed in glutaraldehyde to a microprobe laboratory with a request for microprobe analysis for electrolytes.

The research on the sterlet roe artificial insemination using cryopreserved sperm was carried out in the research base of the RAS Southern Scientific Centre (the Rostov region). Reproductive cells (including cryopreserved cells), larvae, sterlet fry ( Acipenser ruthenus Linnaeus, 1758) were taken as an object of research. A half of the roe (1.7 kg) taken from female starlet was inseminated by native sperm (control group); another half was inseminated by defrosted sperm of two males, which was stored in liquid nitrogen at -196ºC during 3 years (pilot group). Incubation lasted 5 days at water temperature 14.5-18.2ºC, with daily fluctuations of temperature 1.9ºC. Roe insemination in the control group made 90%, in the pilot group - 70%. Roe embryonic growth in the control group was faster, but embryogenesis duration in the pilot group met the standard time limits. Hatching prolarvae in the control group started one hour earlier, than in the pilot group; it made 75% and 60% of all incubated roe, correspondingly. Waste during the period of larvae maturing before they pass to mixed feeding was negligible - 2% in the control group and 3.4% in the pilot group. According to the test results, "open field" of reactivity of the central nervous system in the pilot group fry didn’t change from the control group fry, but more active response to stimuli was noted in the pilot group, which is very important for fry adaptation to the conditions in natural water basins. It was established that sterlet offspring obtained with use of defrosted sexual cells does not differ from the offspring obtained using native sperm and has higher morphometric characteristics. The test results prove the possibility and practicability of using sexual cells stored in liquid nitrogen for artificial restoration and formation of sturgeon fish broodstocks.

Periode emas tumbuh kembang seorang anak dimulai sejak anak dalam kandungan hingga berusia dua tahun. Wanita hamil dan anak-anak usia dibawah 5 tahun berada pada risiko tertinggi micronutrient deficiencies (MNDs), masalah pertumbuhan, penurunan intelektual, komplikasi perinatal dan meningkatnya risiko morbiditas dan mortalitas. Oleh karena itu ibu hamil memerlukan program edukasi tentang nutrisi yang tepat untuk kesehatan ibu dan janin. Edukasi berbasis keluarga sesuai program keluarga sehat dapat meningkatkan intensi untuk mengubah perilaku kesehatan. Menurut Theory of Planned Behavior (TPB), bahwa intensi untuk mengubah perilaku dipengaruhi oleh sikap, norma subjektif dan persepsi kontrol perilaku. Penelitian ini bertujuan mengidentifikasi pengaruh edukasi berbasis keluarga terhadap intensi ibu hamil untuk optimalisasi nutrisi pada 1000 Hari Pertama Kehidupan di wilayah kerja Puskesmas Neglasari, Kota Bandung. Desain penelitian menggunakan quasi eksperiment, pre-test and posttest with control group design. Pengambilan sampel dengan teknik total sampling seluruh ibu hamil yang terdata di Puskesmas sampai bulan November 2016 berjumlah 44 orang yang kemudian dibagi dalam kelompok intervensi dan kelompok kontrol. Kegiatan edukasi dilaksanakan melalui tiga kali pertemuan kunjungan rumah. Analisis data menggunakan Wilcoxon test dan Mann whitney test. Hasil penelitian menunjukkan bahwa ada pengaruh yang signifikan antara edukasi berbasis keluarga terhadap intensi ibu hamil untuk optimalisasi nutrisi pada 1000 hari pertama kehidupan (p = 0.00). Program edukasi berbasis keluarga adalah efektif meningkatkan intensi ibu hamil. Dengan demikian, disarankan penerapan edukasi berbasis keluarga dalam intervensi keperawatan untuk meningkatkan kesehatan ibu dan bayi yang dikandungnya.Kata kunci: Edukasi berbasis keluarga, intensi, ibu hamil, nutrisi, 1000 hari pertama kehidupan. Effect Of Family-Based Education Towards Pregnant Mothers’ Intention to Optimize The Nutrition at 1000 First Day Of LifeAbstractChildren “Golden Period” starts since in the womb until two years after birth. The data were analyzed using Wilcoxon test and Mann Whitney testThe findings showed that there was a significant effect between the family-based education towards pregnant mothers’ intention to optimize the nutrition of 1000 first days of life (p = 0.00). Attachment behaviors are behaviors experienced by children when they would leave their homes and families to join his friends at school he regarded as strangers. This behavior can interfere with the functioning life of the child so that the child can not be independent, and parents should be involved more in the activities of children. The study aimed to determine the effect of economic token therapy for reducing the attachment behavior among Kindergarten Students. The design of this study was quasi experimental with pre and post with control group. By using multistage random sampling and consecutive sampling technique with a total sample of 68 people who experienced attachment behavior. Consisted of 34 peoples as intervention group and 34 peoples as control group. Bivariate analysis using Wilcoxon test and mann-whitney test. The results showed that there were significant differences of attachment behavior before and after entering the token economy intervention in the intervention group compared with those in the control group (p = 0.000). Token economy therapy has positive effect to reduce attachment behavior of kindergarten student at the beginning of learning process in the kindergarten school. The application of economic token is recommended to overcome the attachment behavior of existing preschool children in schools and in the general public health services.   KEY WORDS: Attachment behavior, Token economy, Kindergarten students.

ABSTRACT Toilet training is an effort to train children to be able to control and urinate (BAK) and defecate (BAB). Toilet training is one of the main tasks of children at toddler age. Toilet training is one of the main tasks of children in toddler age which is very important to be done to create independence in children in controlling BAK and BAB and children know the parts of the body and their functions. Data in 2012 shows that ± 60% of parents do not teach toilet training to children from an early age. The aim of the study was to find out the effectiveness of toilet training education on maternal behavior and toilet skills in toddler age training (18-36 months). The study was conducted in July-August 2018. This type of quantitative research used the design of the Quasy pretest and posttest experiment with non-equivalent control group design. Samples were 36 mothers and 36 children with purposive sampling technique. Data analysis used Paired t test, Wilcoxon test, Man-Whitney test an Independent t test. The results showed that toilet training education through lecture methods, modules and maze games was more effective than toilet training education through lecture and leaflet methods on children's knowledge and abilities. Conversely, for the role of mothers in supervision there is no significant difference in effectiveness. Health education is recommended in health promotion programs to increase maternal knowledge, the role of mothers and the ability of toilet training children independently. Background: Uric acid is a final product or a waste that is resulted from the metabolism of purines. A high level of uric acid (hyperuricemia) will cause several health problems, such as vascular inflammation, smooth muscle proliferation, and vascular lesion in kidneys. The syzygium polyanthum leaves contain bioactive substances that may affect the level of uric acid in blood. Objective: This study aimed to determine the influence of boiled water of syzygium polyanthum leaves to the changes of uric acid levels in the target area of Puskesmas Pandak 1 Bantul. Methods: This study employed pre- and post-test without control group design. The population consisted of all patients with hyperuricemia in the target area of Puskesmas Pandak 1 Bantul. Sample was selected with a concecutive sampling, gaining a total number of 24 respondents. Data were analyzed with the Wilcoxon test. The dose of boiled water of syzygium polyanthum leaves intake was 0.36g/ KgBW, once a day for 14 days. Result: This research showed that the boiled water of syzygium polyanthum leaves decreased hyperuricemia (uric acid levels), along with the significancy value of 0.009 (p <0.05). At the pre-test time, the average level of uric acid reached 7.279 mg/dl, and after the treatment, it decreased to 6.76 mg/dl. Conclusion: This study has established evidence that the boiled water of syzygium polyanthum leaves is able to decrease hyperuricemia (uric acid level in blood).
