Disorders related to oxidative metabolism, which occur in the production cycle of high-producing dairy cows (HPC) are more frequent during the transition period and early lactation. Blood concentrations of the enzyme superoxide dismutase (SD) are closely related to the activity of stabilizing conditions of oxidative stress (OS). It is very important to know the main mechanisms involved in regulation of metabolic events, to develop new strategies to increase the productive and reproductive performance of HPC. The objective of this study was to evaluate differences in pregnancy rates and pregnancy losses of HPC with different concentrations of SD with fixed-time AI (FTAI). The experiment was conducted on a commercial farm with 140 HPC (multiparous pregnant Holsteins). The evaluations of concentrations of SD were done from 21 days before birth to the day of the first-service FTAI (75 days in milk), with intervals of 14 days. The animals were retrospectively divided into 2 groups (G1 and G2) in accordance with the average output from all cows in peak lactation (4–8 weeks). Data were analyzed using logistic regression to determine the pregnancy rates between groups. There was a time effect and group effect for the concentrations of SD (P < 0.05) only on the first day of FTAI (3054.4a ± 37.4 and 3662.4 ± 42.1b) for G1 and G2, respectively. But this was not reflected in differences in pregnancy rates between the experimental groups (G1 and G2) and 30 (26.7 v. 31.4) and 60 (20 v. 19.6) days. Although the dismutation activity was high, it seems that the plasma concentration of this enzyme alone does not explain the effects of oxidative stress on fertility in HPC.

The objective of the present study was to evaluate the effects of an antioxidant and anti-inflammatory compound found in rosemary plants (Salvia rosmarinus) named carnosic acid during the transition period of dairy cows. From day 1 to 3 after calving, 16 multiparous Holstein cows received a daily intravenous infusion of either 500 mL of saline (NaCl 0.9%; Saline; n = 8) or carnosic acid at a rate of 0.3 mg/kg of BW supplied in 500 mL of saline (CA; n = 8). Blood samples were taken at –7, 2, 5, 7, 14, and 21 d relative to parturition, then analyzed for metabolites related to energy metabolism, muscle mass catabolism, liver function, inflammation, and oxidative stress. CA infusion tended to improve milk performance; however, DMI was unaffected by treatment. At 2 d relative to parturition, CA cows had lower blood concentrations of haptoglobin, paraoxonase, FRAP, and NO2– than saline cows. After treatment infusions, haptoglobin remained lower in CA cows than saline at 5 d relative to parturition. Our results demonstrate that carnosic acid promoted positive responses on inflammation and oxidative stress biomarkers and may promote beneficial effects on lactation performance in peripartal dairy cows.

Previous studies in the field of acid-base and oxidative status in cows have mainly focused on the transition period (three weeks before and three weeks after calving). The aim of this study was to determine the differences in the parameters of acid-base and oxidative status and electrolyte balance in cows not only during the transition period, but also at other stages of the production cycle. Holstein-Friesian cows were divided into four numerically equal groups (n = 6): early lactating cows (9?2 days in milk - DIM), peak lactating cows (50?5 DIM), late lactating cows (170?10 DIM) and dry cows (10?1 days before calving). Venous blood samples were taken from the cows to analyze acid-base status, electrolyte concentrations and oxidative stress parameters, and to compare group means. Significantly higher pH was observed in early lactating cows than in late lactating cows. Sodium (Na+) concentration was significantly lower in early lactating and peak lactating cows compared to dry cows, while chloride (Cl-) concentration was also lower in late lactating cows compared to dry cows. Plasma glutathione peroxidase (GSH-Px) activity was higher in early lactating cows compared to peak lactating cows. Thus, the highest blood pH, lowest Na+ and Cl- concentrations and highest serum GSH-Px activity were observed in early lactating cows compared to other groups, indicating the highest metabolic and oxidative stress during this period. In conclusion, it would be useful to consider the inclusion of these parameters in standard health assessment procedures in intensive dairy production.

Abstract Activation of physiological stress responses have been associated with decreased pregnancy rates in beef cattle. Therefore, alternative cattle restraint methods have been developed to reduce this negative impact. However, little evidence exists comparing pregnancy outcomes of differing methods. Thus our objective was to determine if a difference was present in pregnancy rate to fixed-time artificial insemination in Bos taurus beef females when using a breeding box (b) versus a squeeze chute (c). Cows and heifers from two separate locations (Virginia Tech University, VT; and R.A. Brown Ranch, RAB) were exposed to FTAI protocols at location of origin while restrained in a squeeze chute (n = 169) or a breeding box (n = 162). Females were exposed to the 7 day Co-Sync + CIDR protocol pre-breeding. At day of breeding, females were randomly assigned to treatment. Immediately following breeding, blood was collected via coccygeal venipuncture for assessment of plasma cortisol concentration using an automated ELISA assay (Immulite 2000 XPi, Siemens). Chute and Exit Score (1–5) were assessed at the time of breeding by a trained technician at each location. Pregnancy was verified thirty days post-breeding via rectal ultrasonography. Pregnancy rate (PROC FREQ) and cortisol (PROC GLIMMIX) were analyzed using SAS version 9.4. Also, a Linear Regression Model (SAS version 9.4) was used to determine relationships between chute score, exit score, and cortisol. It was found that there was no difference in pregnancy rate between the breeding box and the squeeze chute (61.73% vs 64.71%, P = 0.31, respectively). Cortisol showed no difference between b and c (59.44±10.17 vs 58.61±10.16, P = 0.68, respectively). There was no linear relationship between cortisol, exit score, and chute score (P = 0.14). It can be concluded that stress responses are similar between the breeding box and squeeze chute. Therefore females will have similar pregnancy rates when exposed to FTAI in either restraint mechanism.

The application of Fixed Time Artificial Insemination (FTAI) protocols has strongly boosted artificial insemination in Brazilian bovine herds. The control of the estrous cycle with protocols based on progesterone and estrogen involves synchronizing the emergence of a follicular wave, controlling the progesterone phase, and inducing ovulation in a synchronized manner. The increase in pregnancy rates starts with the preparation of the breeding season and the choice of protocols and females. Thereafter, based on the characteristics of the property and the lots of animals, small adjustments can be made to the hormonal protocols as needed. Finally, resynchronization can increase usability by allowing multiple inseminations within short periods. The search for new strategies is of paramount importance for the development of the FTAI protocol and the increase in final pregnancy rates. Thus, this review sought to highlight the main critical points and adjustments of the FTAI protocols and their applicability in the reproduction of beef cattle.

Postpartum anestrous cows are usually treated with progesterone/progestagen protocols in order to induce ovulation and sustain pregnancy. In the present work, the efficiency of a protocol (modified GPE), in which temporary calf removal (TCR) and/or administration of eCG, replaced the use of progesterone/progestagens, was evaluated in anestrous animals. Anestrous Nelore cows (40 to 60 days postpartum, n = 22) were randomly allocated to two groups: GPE/eCG and TCR/GPE/eCG. At a random stage of the estrous cycle (D0), animals from Group GPE/eCG were treated with GnRH (50 �g, licereline, i.m., Gestran Plus�; Tecnopec, Sao Paulo, Brazil) and seven days later (D7) they received PGF2� (150 mcg, d-cloprostenol, i.m., Prolise�) and eCG (300 UI, i.m., Novormon�). On D8, estradiol benzoate (EB, 1 mg, Estrogin�; Tecnopec) was administered, and 30 to 36 h afterwards all cows were inseminated at fixed-time (FTAI), without estrus detection. The animals from group TCR/GPE/eCG received the same treatment described above, but with temporary calf removal (during 48 h) before beginning hormonal treatments. The ovaries were examined by ultrasonography (Aloka SSD 500, 7.5 MHz probe) before (D-12, D-2), during (D0, D7, D8), and after (D9, D10, D11, D17, D60) hormonal treatments. The absence of CL, during the ultrasonography performed before starting the treatments, was the criterion used to classify the cows as in postpartum anestrus. Administration of GnRH (D0) induced ovulation (determined by the presence of CL on D7) in 6 of 11 cows (54.5%) from Group GPE/eCG and TCR/GPE/eCG. In relation to synchronization of ovulation after EB administration, in Group GPE/eCG, two cows (18.1%) ovulated until 12 h after FTAI, and four (36.3%) 12 h afterwards (24 h after FTAI). In Group TCR/GPE/eCG, one cow (9%) ovulated approximately 6 h before FTAI, two (18.1%) 12 h after FTAI, and two (18.1%) 12 h afterwards. Preovulatory follicles had a diameter of 11.0 � 1.3 and 11.5 � 2.6 mm in Groups GPE/eCG and TCR/GPE/eCG, respectively. Pregnancy rates, determined by ultrasonograpy 40 to 46 days after FTAI, were 27.2% (3/11) and 45.4% (5/11), respectively. The results indicate that association of TCR with protocol GPE/eCG did not improve synchronization of ovulation or pregnancy rates (P > 0.005). However, increase in pregnancy rates was observed in another experiment with a large number of animals (Pinheiro et al. 2005 Reprod. Fert. Dev. 17, 161, abstr), in which cows from Group TCR/GPE/eCG had higher pregnancy rates (51.5%; 34/66) when compared to animals from Group GPE/eCG (28.3%, 21/74; P < 0.05). The objective of the experiment was to evaluate the effects of recombinant bovine somatropin (rbST), and the reuse of the progesterone (P4) releasing devices in resynchronization, on the pregnancy rates of Nellore cows submitted to fixed-time artificial insemination. A group of 123 multiparae Nellore cows, was submitted to a resynchronization protocol: on day 0 a Bovine Intravaginal Device (DIB® ) with 1,0g of P4 was implanted, associated with intramuscular administration of 2,0mg of estradiol benzoate (IM); on day 8 DIB was removed; and 1,0mg of estradiol cypionate, 0,15mg of prostaglandin F2? and 300 UI of equine chorionic gonadotropin were administered; on day 10, fixed-time artificial insemination was conducted (FTAI). The cows were randomized into G1 (n=12) – without rbST / with used Bovine Intravaginal Device, G2 (n=50) – without rbST / with new DIB, G3 (n=11) - with rbST / with used DIB and G4 (n=50) – with rbST/ with new DIB. rbST was introduced on the eighth day of the protocol. Sixty days after TAI, pregnancy diagnoses were conducted, via rectal palpation. Blood samples were taken on day 0, 8 and 10 of the protocol, in order to assess P4 plasma concentrations. Pregnancy rates were statistically evaluated through Generalized Linear Models Theory and their significance was tested with Analysis of Deviance. Pregnancy rates were 58%, 40%, 81% and 48% for G1, G2, G3 and G4, respectively, with significant statistical difference for G3. Plasma concentrations of P4 were not statistically different among groups, or collections. In view of the results obtained, we concluded that the administration of rbST in association with P4 DIB, used for the third time, improves pregnancy rates. Estrus resynchronization and re-insemination positively impacted pregnancy rates.

Recently, interest in the problem of free radical oxidation in biological membranes, which is directly related to both the normal functioning of cells and the occurrence, course and outcome of many pathological conditions, has increased again in clinical medicine. The aim was to determine the role and impact of antioxidant defense in boys with hypoandrogenism. The study involved 75 adolescents with hypoandrogenism aged 13–18 years, who underwent a complex of clinical and laboratory examinations. All patients were conducted complex of anthropometric research and determination of the degree of delayed puberty, laboratory and instrumental examination. Free radical oxidation was determined by the levels of malondialdehyde, conjugated dienes, carbonated proteins, superoxide dismutase and catalase in the serum, and restored glutathione and glutathione peroxidase in whole blood. Based on their determination, the coefficient of oxidative stress was calculated. Statistical processing of results was performed using parametric and nonparametric methods. The study of indicators of the free radical oxidation process found that adolescents with hypoandrogenism have multidirectional changes in the oxidation of proteins and lipids, namely: the level of conjugated dienes increases, the concentration of malondialdehyde remains at the level of the control group, and the level of carbonated proteins tends to decrease. As for the activity of antioxidant protection enzymes, a significant decrease in the level of glutathione peroxidase was detected, while the level of superoxide dismutase and catalase remained at the level of normative indicators. Oxidative stress accompanies and is one of the pathogenetic links in the formation or maintenance of the state of hypoandrogenism in boys. This requires the use of antioxidants, the complex of which must be selected individually.

Endometriosis is described as a gynecological disorder characterized by the presence of endometrial tissue outside the uterus; extensively explored because of its increasing incidency, with an indubitable diagnostic only after invasive surgery, with no efficient treatment, it has still many aspects to be elucidated. A growing body of facts sustain oxidative stress as a crucial factor between the numerous incriminated factors implicated in endometriosis ethiopathogeny. Reactive oxygen species(ROS) act to decline reproductive function. Our study intends to determine if an experimental model of endometriosis may be useful to assess the impact of oxidative stress on endometrial cells; we have used a murine model of 18 adult Wistar female rats. A fragment from their left uterine horn was implanted in the abdominal wall. After 4 weeks, a laparatomy was performed, 5 endometrial implants were removed, followed by biochemical tissue assay of superoxide dismutase(SOD) and catalase(CAT). At the end of the experiment, the rats were sacrificed, the implants were removed for histopathological exam and biochemical assay of antioxidant enzymes. The results revealed decreased levels of antioxidant enzymes, pointing on significant oxidative stress involvement.
