Several factors influence the limited application of assisted reproductive technologies (ART) in the canine species. Most problems arise because of the complex nature of reproductive physiology of the dog. For example, dogs are monoestrus, generally exhibiting oestrus only once every 6 month to 1 year. In the canine species, there has been little research on the ART because of difficulties associated with anatomy and reproductive physiology. Because in vitro maturation of canine oocytes has been particularly difficult, in vivo matured oocytes have been used in somatic cell nuclear transfer. However, the number of oocytes that can be obtained using this approach is limited, with ∼6 to 10 good oocytes being obtained per collection. The present study was undertaken to evaluate the effects of different dosages of eCG on folliculogenesis in the dog and to determine the number of oocytes that might be obtained after ovulation. The experimental design involved 3 groups that were treated with different dosages of eCG at the early stage of proestrus; Group A was a nontreatment (control) group, Group B received 200-IU eCG SC injections every day, and Group C was injected with 500 IU of eCG every 2 days until reaching 2 to 3 ng mL–1 serum progesterone concentration, respectively. Dogs in Groups B and C received a 1 000-IU hCG SC injection when progesterone concentrations reached 2 to 3 ng mL–1. The serum progesterone concentration was examined with a Radioimmunoassay Kit (Diagnostic Systems Laboratories Inc., Webster, TX, USA). The day of ovulation was considered as the day when serum progesterone concentration reached 4.0 to 7.2 ng mL–1. Approximately 70 to 76 h after ovulation, the dogs were subjected to the oocyte collection procedure. One-way ANOVA followed by Duncan’s multiple range tests was performed. The significance level was <0.05. In total, 446 oocytes were recovered from 60 bitches, with an average of 6.4 oocytes/dog in Group A (from 49 bitches), 16.6 oocytes/dog in Group B (from 5 bitches), and 8.5 oocytes/dog in Group C (from 6 bitches). The oocytes collection rate (number of oocytes per dog) in Group B was higher (P < 0.05) than those in the other 2 groups. In conclusion, the results showed that eCG treatment with hCG in early-proestrus-stage bitches can result in a greater number of recovered in vivo matured oocytes. This technology could become a useful research tool for canine cloning and ART.

In order to increase the effectiveness of assisted reproductive technologies (ART) programs, it is essential to improve and develop conditions of embryo culture prior its transfer or cryopreservation of expanded blastocysts on the day 5 or 6. The aim of the study was to assess the effect of human blastocysts’ expansion timing on clinical pregnancy rate (CPR), miscarriage rate (MR) and take-home baby rate (THBR) in frozen-thawed cycles during ART programs. The study involved 2275 frozen embryo transfers (FET) of blastocysts expanded on the day 5 (group A) and 170 FET of blastocysts expanded on the day 6 (group B). The pregnancy rates in both groups were 50.8% and 46.5% respectively. There were no statistically significant differences in clinical pregnancy rate 37.4% and 37.0%, miscarriage rate 26.0% and 21.5% in both groups, respectively. THBR, as the main indicator of efficiency in the programs with transfer of post thawed expanded blastocysts on the day 5 (group A) or 6 (group B) were 36.5% and 35.2%, respectively (the difference is insignificant). In conclusion, in cryoprotocols the day of blastocyst expansion (day 5 or 6 of development) does not statistically affect PR, MR and THBR. In FET programs the quality of blastocyst (excellent and good) should be prioritized regardless of the day of cryopreservation.

The article presents an analysis of literature data and the results of own prospective studies of the application of the local iatrogenic injury method with the parallel introduction of plasma enriched with platelets into the basal layer of the endometrium in pregravid preparation for treatment with assisted reproductive technologies (ART) in patients with tubal peritoneal infertility and marked lag development of the endometrium. The objective: was to determine the optimal time for the injection of scratch scratching in pregravid preparation before the next cycle of ART treatment in patients with tubal peritoneal infertility and marked lag in the development of the endometrium. Materials and methods. The study included 109 patients with unsuccessful attempts to treat ART with a tubal peritoneal factor of infertility with a marked degree of lag in the development of the endometrium according to absolute indications. The subjects were divided into three groups. Group A – 47 patients who experienced a marked lag in the development of the endometrium during the control stimulation of superovulation during ART (ETC + ET) and suggested the application of the procedure of injection scaling of the endometrium on the 7th day of the stimulation cycle (CA). Group B – 32 patients who underwent the procedure of injection endometrial scratching on the 22nd day of the menstrual cycle (MC), which preceded the repeated cycle of CGI + ET. Group B – 30 patients who underwent the procedure of injection endometrial scratching on the 8th day of the MC, which preceded the repeated cycle of CCN + ET. Results. The use of injection scratches is more effective in improving the clinical outcomes of ART by using the methods of ART (CAS + ET) in the case of a pronounced delay in the development of the endometrium in the follicular phase of the CA on the 6th to 7th day of gonadotropin use. During treatment with ART, the frequency of pregnancy for each transfer of embryos (CHNBPE) was significantly lower, amounting to 9.4% (3 of 32) in group B and 7.1% (2 of 28) in group B than CHNBPE in group A – 27.7% (13 of 47). The frequency of early reproductive losses in group A was 23.1% (3 of 13), in group B – 66.7% (2 of 3), in group B – 50% (1 of 2). Conclusion. During the study of the optimal period for the appointment of injection scratching, it was found that injection endometrial scratching using autoplasma enriched with platelets demonstrates the best pregnancy rate for each transfer of embryos and low early reproductive loss if performed in the follicular phase of the stimulation cycle by 6–7th day of gonadotropins). Introduction and Aim.Exceptional properties of the NiTi archwires may be jeopardized by the oral cavity; thus its long-term effect on the mechanical and physiochemical properties of NiTi archwires was the aim of work.Material and Methods. Study group comprised sixty 0.016×0.022 NiTi archwires from the same manufacturer evaluated (group A) after the first 12 weeks of orthodontic treatment. 30 mm long pieces cut off from each wire prior to insertion formed the control group B. Obeying the strict rules of randomization, all samples were subjected to microscopic evaluation and nanoindentation test.Results.Both groups displayed substantial presence of nonmetallic inclusions. Heterogeneity of the structure and its alteration after usage were found in groups B and A, respectively.Conclusions.Long-term, reliable prediction of biomechanics of NiTi wires in vivo is impossible, especially new archwires from the same vendor display different physiochemical properties. Moreover, manufacturers have to decrease contamination in the production process in order to minimize risk of mutual negative influence of nickel-titanium archwires and oral environment.

Abstract Immunosuppressive agents have been recognized as factors that induce changes and modifications in bone metabolism. The purpose of this study was to evaluate the effect of ozonated plant extracts (herein termed ozonated oil) under the influence of Cyclosporin A (CsA) on osseointegration. A total of 20 dental implants were placed in 20 rabbit tibiae assigned to Group A or B. CsA was injected at an immunosuppressive dose in Groups A and B as a single-dose treatment. At the day of surgery, Group A received a single topical ozonated oil treatment (0.55 mL) around dental implants; Group B, the control group, received no ozonated oil. Animals were sacrificed after 8 weeks. Radiographs were obtained at implant surgery and on the day of sacrifice. Bone quality was compared between the 2 groups. Radiographically, osseointegration was microscopically evaluated using scanning electron and light microscopies. In ozonated Group A specimens, light microscopic examination demonstrated evidence of more organized mature bone compared with Group B. Within the limits of this study, the results suggest that short-term administration of CsA, when administered with topical ozonated oil, may influence bone density and the quality of dental implant osseointegration. Therefore, topically applied ozonated oil may influence bone density and the quality of osseointegration around dental implants.

ABSTRACT Objectives Oil pulling is an age-old practice that has gained modern popularity in promoting oral and systemic health. The scientific verification for this practice is insufficient. Thus, this study evaluated the effect of coconut oil pulling on the count of Streptococcus mutans in saliva and to compare its efficacy with that of Chlorhexidine mouthwash: in vivo. The null hypothesis was that coconut oil pulling has no effect on the bacterial count in saliva. Materials and methods A randomized controlled study was planned and 60 subjects were selected. The subjects were divided into three groups, Group A: Study Group: Oil pulling, Group B: Study Group: Chlorhexidine, and Group C: Control Group: Distilled water. Group A subjects rinsed mouth with 10 ml of coconut oil for 10 minutes. Group B subjects rinsed mouth with 5 ml Chlorhexidine mouthwash for 1 minute and Group C with 5 ml distilled water for 1 minute in the morning before brushing. Saliva samples were collected and cultured on 1st day and after 2 weeks from all subjects. Colonies were counted to compare the efficacy of coconut oil and Chlorhexidine with distilled water. Results Statistically significant reduction in S. mutans count was seen in both the coconut oil pulling and Chlorhexidine group. Conclusion Oil pulling can be explored as a safe and effective alternative to Chlorhexidine. Clinical significance Edible oil-pulling therapy is natural, safe and has no side effects. Abstract Background. The oral cavity is potentially high-risk transmitter of COVID-19. Antimicrobial mouthrinses are used in many clinical pre procedural situations for prophylactic purposes. An evident-based investigation for an effective mouthwash solution against salivary SARS-CoV-2 is urgently required for the exposure reduction during dental procedures. Aims. This study aimed to evaluate in vivo virucidal efficacy of 2 mouthwashes: 1% Povidone-iodine and 0.2% Chlorhexidine as a dental preprocedural oral disinfection against salivary SARS-CoV-2. Materials and Methods. In this randomized-controlled clinical trial, studied group comprised laboratory-confirmed COVID-19 positive patients through nasopharyngeal swabs. Participants were divided into 3 groups. For 30 seconds, group A gargled with 1% Povidone-iodine, group B mouthrinsed with 0.2% Chlorhexidine and control group C mouthrinsed with distilled water. Saliva samples were collected before and 5 minutes after mouthwash. SARS-CoV-2 rRT-PCR was then performed for each sample. Evaluation of the efficacy was based on difference in Ct value. The analysis of data was carried out using GraphPad Prism version 5 for Windows. Paired t test and unpaired t test were used. A probability value of less than 0.05 was regarded as statistically significant. Results. Sixty-one compliant participants (36 female and 25 male) with a mean age 45.3 plusmn 16.7 years-old were enrolled. A significant mean Ct value difference (p < 0.0001) between the paired samples in group A (n = 25) and also in group B (n = 27) (p < 0.0001) was found. In contrast, no significant difference (p = 0.566) existed before and after the experiment in the control group C (n = 9). Moreover, a significant difference was noted between the delta Ct of distilled water wash and each of the 2 solutions 1 % Povidone-iodine (p = 0.012) and Chlorhexidine 0.2% (p = 0.0024). No significant difference was found between the delta Ct of patients using 1% Povidone-iodine and Chlorhexidine 0.2% solutions (p = 0.24). Conclusion. Chlorhexidine 0.2% and 1% Povidone-iodine oral solutions are effective preprocedural mouthwashes against salivary SARS-COV-2 in dental treatments. Their use as a preventive strategy to reduce the spread of COVID-19 during dental practice should be systematically implemented.

In the canine species, assisted reproductive technologies, especially in vitro maturation (IVM) and IVF, are still ineffective. The main limiting factor remains the immaturity of the oocytes collected from anestrus ovaries. The ability of an oocyte to reach the MII stage in vitro is linked to the diameter of its follicle and anestrus oocytes, collected from small (<1 mm) follicles, are profoundly immature (De Lesegno et al. 2008). The objective of this study was to improve cytoplasmic quality by mimicking in vivo conditions; that is, to test the effect of pure preovulatory follicular fluid (FF) on survival and IVM rates of anestrus dog oocytes, in order to improve the nuclear and cytoplasmic maturation of these immature oocytes. Follicular fluids samples were collected from 54 Beagle bitches at 2 stages: before the LH peak (n = 23 bitches) and after the LH peak (n = 31 bitches). Only follicular fluid samples from large (>4 mm) follicles were collected and pooled by stage. Control oocytes were matured in 20% FCS/M199 medium. Groups of 5 oocytes were in vitro matured in 30 μL of follicular fluid, in half-area 96-well plates (5% CO2, 38°C). After 72 h of IVM, oocytes were denuded, fixed, and stained for DNA and tubulin before observation by confocal microscopy, and nuclear stages were classified as GV-A to GV-E, MI, and MII (Reynaud et al. 2012). A total of 460 oocytes were collected from 13 anestrus bitches and allocated to either the control medium (n = 155), the Pre-LH FF (n = 145) or the Post-LH FF (n = 160) groups. After 72 h of IVM, the morphology of the cumulus–oocyte complexes (COC) in the post-LH group was different from that of the others: cumulus cells appeared more compact and darker. Analysis of the nuclear stages showed that the degeneration rate was significantly higher (P < 0.05) in the post-LH group (58.7%) than in the pre-LH (40.9%) or in the control group (34.4%). No significant differences (P > 0.05) were observed between the 3 groups in the rate of immature GVA-B oocytes (36.4, 28.5, and 25.3% in the control, Pre-LH, and Post-LH groups, respectively), in the rate of meiotic resumption (GV-C/D/E, MI, MII stages, 44.4, 51.9, and 38.7% in the control, Pre-LH, and Post-LH groups, respectively). Metaphase II rates were not significantly different (12.1, 8.6, and 4.8% in the control, Pre-LH, and Post-LH groups, respectively). In conclusion, canine COC may survive when exposed to IVM in pure follicular fluid, but the degeneration rate was higher in the post-LH group. The presence of follicular fluid did not inhibit meiosis resumption, but did not significantly improve IVM rates. To better mimic in vivo conditions, IVM in a sequence of media, such as IVM in follicular fluid followed by IVM in oviducal fluid remains to be tested.

Background Recently, some authors introduced a water glass (WG, sodium-silicate glass; Na2O·SiO2·nH2O) coating over tricalcium phosphate (TCP) bioceramic to modulate its resorption rate and enhance the bone cell behaviors. In this study, four different types of granular samples were prepared to evaluate the ability of new bone formation in vivo using micro-computed tomography and histology. Methods Four types sample groups: group A (pure HA as a negative resorption control); group B (pure TCP as a positive resorption control); group C (WG-coated TCP as an early resorption model); and group D (same as group C but heat-treated at 500°C as a delayed resorption model). Cylindrical tube-type carriers with holes were fabricated with HA by extrusion and sintering. Each carrier was filled densely with each granular sample. Four types of tubes were implanted into the medial femoral condyle and medial tibial condyle of New Zealand White rabbits. Results The HA group (A) showed the lowest amount of new bone formation. All the TCP sample groups (B, C, and D) showed more new bone formation. On the other hand, among the TCP groups, group C (early resorption model) showed slightly more bone formation. The amount of residual bioceramics was most abundant in the HA group (A). All the TCP sample groups showed less residual bioceramics than group A. Among the TCP groups, group C showed slightly more residual bioceramics. Group B showed the lowest amount of residual bioceramics. Conclusions The WG-coated TCP sample (group C) is the best bone substitute candidate because of its proper biodegradation rate and the Si ions release because the WG-coated layer reduces the material resorption and enhances the new bone formation. That is, the WG-coated TCP is believed to be the best material for the application of an artificial bone substitute material.

The aim of this study was to investigate whether the nanohydroxyapatite/collagen/poly(L-lactic acid) (nHAC/PLA) composite is suitable to be compounded with VEGF to enhance the axial vascularization in vivo. Thirty rabbits were divided into 2 groups of 15 animals each. In control group, a nHAC/PLA scaffold slice was vascularized axially by an inserted ligated femoral arteriovenous (AV) bundle in the animal. In experimental group, a slice compounded with VEGF gel was applied. The rabbits were sacrificed at 2 weeks, 6 weeks, and 10 weeks after surgery; the specimens of scaffold slices underwent histomorphometric examination; analysis of the microvessel density (MVD) of both groups was done. The combination with VEGF (Group B) did not enhance the vascularization in early phase (2 and 6 weeks,P>0.05) but worked in later phase (10 weeks,P<0.05). The data of the experiment demonstrated the suitability of the nHAC/PLA composite as carrier for the growth factor VEGF, enabling its sustained release in bioactive form with enough binding efficacy.

Thromboembolic complications occur frequently in patients who have undergone cardiac valve replacement and platelets are known to be actively involved in the pathogenesis of such thrombi, A method of in-vivo assessment of platelet function would be useful in the management of such patients. βTC and PF4 Levels, the indicators of platelet release reaction in-vivo were measured in 100 patients who had cardiac valve replacement at least six months previously) Group A-54 patients had porcine heterograft, Group B-32 patients had disc valves and Group C-14 patients had double valve replacement. Findings of these patients were compared with 50 controls. In the control group, the mean βTG was 27.6ng/ml, and PF4 9.0 ng/ml. In each of the three groups of operated patients, both mean βTC and PF 4 levels were significantly raised (p < 0.05). Group A 61.5 ng/ml and 26.5 ng/ml, Grout B 57.0 ng/ml and 19.6 ng/ml and Group C 63.0 ng/ml and 36.6 ng/ml respectively. Of the 100 cases studied, 57 involved mitral, 29 aortic and 14 multiple valve replacement. No significant differences were observed in βTC or PF4 plasma levels with the different type of valve involvement. The clinical implications of these results will be discussed.
