40 TIMING OF INSEMINATION WITH SEXED OR NONSEXED SEMEN ON PREGNANCY RATES OF JERSEY HEIFERS DETECTED IN HEAT BY RADIOTELEMETRY

The objectives of this study were to evaluate the follicular dynamics (experiment 1) and the effects of the timing of insemination with sexed or nonsexed semen on pregnancy rates (experiment 2) of Jersey heifers detected in heat by a radiotelemetric estrus detection system. In experiment 1, 43 Jersey heifers, around 12 mo old and BCS of 2.68 ± 0.11 (1 to 5 scale) were used. The Heat Watch (HW) system was utilized to detect the onset of estrus and mounting behavior associated with estrus. Ultrasound examinations to monitor follicular dynamics occurred every 12 h from estrus onset for 48 h. Statistical analyses were performed using GLM and GLIMMIX procedure of SAS (SAS Institute, Cary, NC, USA). The results of experiment 1 indicated a mean ovulatory follicle diameter of 14.1 ± 0.3 mm, ovulation rate of 86.1% (37/43), and an interval of 31.2 ± 0.9 h from onset of heat to ovulation. In experiment 2, 753 Jersey heifers were allocated in a 2 × 4 factorial with semen (sexed and nonsexed) and AI period (0 to 6, 6 to 12, 12 to 18, and 18 to 24 h after heat onset) as parameters. Semen from 3 bulls was used, with ejaculates divided in 2 fractions: one fraction was submitted to the traditional freezing procedure and the other was submitted to the sexing process and then frozen. The statistical analysis was performed using GLIMMIX procedure of SAS. There was no interaction among the semen, bull, and AI period. There were effects on pregnancy rate by type of semen [sexed 49.5% (189/382) and nonsexed 64.2% (238/371); P = 0.001] and by bull [bull A 53.5% (107/200)b, bull B 50.0% (108/216)b and bull C 63.4% (211/333)a; P = 0.008]. Semen from bull C resulted in a greater pregnancy rate for both sexed and nonsexed semen. Within semen type, there were no differences in pregnancy rates by AI moment [sexed: 0 to 6 h 48.2% (41/85), 6 to 12 h 48.7% (54/111), 12 to 18 h 49.5% (49/99), 18 to 24h 52.4% (44/84) and nonsexed: 0 to 6h 62.8% (49/78), 6 to 12h 60.6% (63/104), 12 to 18h 68.0% (68/100), 18 to 24h 64.8% (57/88); P = 0.77]. We conclude that the use of sexed semen resulted in a lower pregnancy rate than nonsexed semen, and that AI timing does not affect conception rate in Jersey heifers identified in estrus by radiotelemetric estrus detection system. Abstract The present study aimed to determine the effects of progesterone-based gonadotropin-releasing hormone (GnRH), prostaglandin F2α (PGF2α), and equine chorionic gonadotropin (eCG) injections on progesterone profiles and pregnancy rates in cows with no estrus symptoms within 60 days after parturition. A total of 80 cows were included in the study. All animals had the progesterone-releasing device PRID®Delta placed intravaginally for nine days with an injection of GnRH. On the eighth day, PGF2α was injected, and PRID®Delta was removed from the vagina on day nine. Artificial insemination was carried out 60 hours after PRID®Delta removal. In half of the animals (n = 40), 600 IU of eCG was injected when PRID®Delta was removed on the ninth day before artificial insemination 60 hours later. Blood samples were taken from the tail vein on days 0 and 8 to determine progesterone levels. The pregnancy rate in the group that received eCG was 37.5%, while it was 27.5% in those that did not (P = 0.4). While the dominant follicle diameter was 15.5 mm in cows injected with eCG during timed artificial insemination, the follicle diameter was 12.4 mm in cows with no eCG injection (P <0.001). There were no differences in serum progesterone values in blood samples taken until the time of artificial insemination. However, progesterone values in the blood taken during artificial insemination were 0.94 ng/ml in the eCG- group and 0.72 ng/ml in the eCG+ group (P <0.05). As a result, it was determined that eCG injections, in addition to progesterone-based GnRH and PGF2α applications, increased the pregnancy rates in cows without symptoms of estrus. The dominant follicle diameter was larger in cows treated with eCG during artificial insemination; however, follicle size did not increase the pregnancy rate.

This study was performed on an industrial dairy farm near Tehran province, Iran, on 666 lactating Holstein cows from September 2019 to June 2020 in order to eliminate the harmful effects of heat stress on the reproductive performance of the cows. The hypothesis of the recent study was that by increasing the serum concentration of progesterone (P4) in the luteal phase before insemination in the form of Presynch-Ovsynch+CIDR (POC) and G6G protocols, the pregnancy rate of cows would be increased compared to the Presynch-Ovsynch (PO) protocol. The aim of the present study was to evaluate the effects of Presynch-Ovsynch (PGF2α-14d- PGF2α-12d-Ovsynch (OVS: GnRH(GnRH1)- 7d-PGF2α(PG)-2d-GnRH)-18h-Fixed-time artificial insemination (FTAI), n=212), Presynch-Ovsynch+CIDR (similar to the PO protocol, plus the use of a CIDR for 7 days, from the start of the OVS+FTAI protocol to the time of PG injection, n=230), and G6G (PGF2α- 2d-GnRH-6d-OVS+FTAI, n=224) protocols on the reproductive performance and pregnancy rate of lactating Holstein cows. The average body condition score (BCS) for the cows was about 2.5 at the time of the onset of the OVS+FTAI program. The average daily milk production for cows at the time of the start of the OVS+FTAI program was 38.3 kg/day. On average, the studied cows were inseminated 86 days postpartum. Implementation of the G6G protocol in comparison to the PO and POC protocols increased the serum concentration of P4 at GnRH1 (P=0.04). The cows that received the G6G protocol had a greater number of corpuses luteum (CL) on their ovaries at PG in comparison to the cows in the PO and POC protocols (P=0.03 and P=0.05, respectively). For all treatment protocols, the pregnancy rate of cows with &gt;2 CLs on their ovaries at PG was significantly higher than for cows with ≤2 CLs on their ovaries at this time. The overall pregnancy rate for all cows studied was 42%. Separately, pregnancy rates for cows in PO, POC, and G6G protocols were calculated at 36.7%, 41.7%, and 47.3% respectively. The highest and the lowest pregnancy rates were obtained in the G6G and PO protocols, respectively (P=0.03). It can be concluded that the G6G protocol increased the pregnancy rate of cows in comparison to the PO but not to the POC protocol. This result can be attributed to the increase in serum concentration of P4 at GnRH1 and the greater number of CLs at PG following implementation of the G6G compared to the PO protocol. Although CIDR administration in the POC protocol led to an increase in the pregnancy rate of cows compared to the PO protocol, possibly by increasing the synchrony rate and the quality of the ovulatory follicle, this increase was not statistically significant (P=0.09).

Background: Beta-(β-) carotene, is the precursor to vitamin A, in particular, has some potential benefits on reproduction. The main objective of this study was to investigate the efficacy of β-carotene administration on fertility following either prostaglandin F2 alpha (PGF) induced estrus or Ovsynch protocol in lactating dairy cows.Methods: Cows with at 47±3 postpartum days were divided into two groups: β-carotene group (βC, n=139) was treated with injectable β-carotene while untreated cows served as control (CON, n=227). In both groups, PGF was administered and heatmount detectors were applied at 54±3 days postpartum. Cows detected in estrus after PGF were inseminated. Cows that had not been detected in estrus were divided into two groups 7 days after PGF administration; βC-OVS (n=137) and CON-OVS (n=89). Ovsynch protocol was initiated 4 days after β-carotene administration.Result: The estrus detection rate was similar between the βC and CON groups (P = 0.19). Pregnancy per AI (P/AI) on d 31 was also similar between groups (P = 0.93). In the Ovsynch protocol, ovulation to the first GnRH and ovulatory follicle diameter at the time of insemination did not differ between groups. No difference was observed in P/AI at d 31 (P = 0.13). The results of this study indicated that β-carotene administration had no effect on fertility either PGF induced estrus or Ovsynch protocol in dairy cows.

Fertility in dairy cows has decreased over the last fifty years while milk production per cow has increased. Furthermore, dairy herds become larger resulting in fewer employees per cow, which might also influence reproductive performance. To elucidate the situation in Germany, selected parameters (conception rate, service rate, pregnancy rate, days to first insemination and days open) were studied using data on 148 herds. For statistical analysis the herds were categorized concerning milk yield: (1) &lt; 30 kg, (2) 30‒35 kg, and (3) &gt; 35 kg/cow/day as well as concerning herd size: (1) &lt; 200, (2) 200‒400, (3) 400‒1000 and (4) &gt; 1000 milking cows. There was no difference in conception rate among herds. That means on dairy farms of larger size or with high milk yield the chance of an inseminated cow to become pregnant is the same as in small herds or in herds with low milk yield. Small herds (&lt; 200 cows) had lower pregnancy rates than larger herds (200–400 cows). The pregnancy rate in herds with different milk yield was not statistically different. Though there was a trend that dairy farms with higher milk yield (&gt; 30 kg) had higher pregnancy rates. The statistical differences and trends of pregnancy rates are due to higher service rate in larger herds as well as in herds with high milk yield. Therefore, service rate is the key factor for high reproductive performance on dairy farms. Poor fertility is not associated with high milk yield on herd basis or large herd size but may represent inappropriate farm management.

Asymmetry in the cow affects ovarian function and pregnancy. In this work we studied ovarian and uterine asymmetry. Synchronised animals, in which in vitro-produced embryos (n = 30–60) had been transferred on Day 5 to the uterine horn ipsilateral to the corpus luteum (CL), were flushed on Day 8. Ovulatory follicle diameter, oestrus response and total protein flushed did not differ between sides. However, a corpus luteum in the right ovary led to plasma progesterone concentrations that were higher than when it was present in the left ovary. Fewer embryos were recovered from the left than the right horn. Among 60 uterine proteins identified by difference gel electrophoresis, relative abundance of nine (acyl-CoA dehydrogenase, very long chain; twinfilin, actin-binding protein, homologue 1; enolase 1; pyruvate kinase isozymes M1/M2 (rabbit); complement factor B Bb fragment ; albumin; fibrinogen gamma-B chain; and ezrin differed (P < 0.05) between horns. Glucose concentration was higher, and fructose concentration lower, in the left horn. In a subsequent field trial, pregnancy rates after embryo transfer did not differ between horns (51.0 ± 3.6, right vs 53.2 ± 4.7, left). However, Day 7 blood progesterone concentrations differed (P = 0.018) between pregnant and open animals in the left (15.9 ± 1.7 vs 8.3 ± 1.2) but not in the right horn (12.4 ± 1.3 vs 12.4 ± 1.2). Progesterone effects were independent of CL quality (P = 0.55). Bilateral genital tract asymmetry in the cow affects progesterone, proteins and hexoses without altering pregnancy rates.

Treatments with GnRH and PGF2α for synchronization of ovulation has resulted in acceptable pregnancy rates after fixed-time artificial insemination in dairy cows without estrus detection. The objective of the present study was to evaluate the practicability of ovulation synchronization (Ovsynch, Pursley JR et al. 1995 Theriogenology 44, 915–923) in dairy cattle using GnRH and PGF2α for the embryo transfer recipients. Dairy cattle (cows; n=100, heifers; n=88) were randomly allocated to one of two groups. The control group (cows; n=45, heifers; n=37) was composed of cows in natural estrus. The ovulation synchronization group (cows; n=55, heifers; n=51) was treated with an intramuscular injection of 100μg of GnRH at a random stage of the estrous cycle. Seven days later, the cattle received PGF2α (Cows; 25–30mg) or PGF2α analog (Heifers; 0.5mg) in order to regress the corpora lutea (CL). Forty-eight hours later, cows and heifers received a second injection of 100μg GnRH. Embryo transfer was carried out 7 days after the second injection of GnRH in the ovsynch group and 7 days after estrus in the control group. The cattle judged to have CL 17mm were classified as acceptable recipients. The size of the follicles and the CL were determined to be of estrus stage and embryo transfer by means of ultrasonography. The mean numbers of follicles and CL were analyzed by ANOVA, while pregnancy rates were analyzed by chi-square test. The results are presented in the Table. The proportion of cows and heifers determined to be acceptable embryo transfers was not different between the control group and the ovsynch group. There were no differences in the proportion of acceptable embryo transfers between the control group and the ovsynch group. Follicle diameter at the time of estrus in the control group (cows; 20.7±0.7mm, heifers; 16.8±0.5mm) were significantly larger than that of the ovsynch group (cows; 18.0±1.0mm, heifers; 14.7±0.2mm) (P&lt;0.05). Although CL diameter at the time of embryo transfer in heifers showed no differences between the control group and the ovsynch group (25.0±1.0mm v. 22.8±1.5mm), The CL diameter of the control cow group was larger than that of the ovsynch group (29.8±0.7mm v. 26.1±1.0mm, P&lt;0.05). However, no differences in pregnancy rate were seen between the control group and the ovsynch group. These results suggest that ovsynch can be effectively applied in an embryo transfer program for cattle. Abstract Apparent ruminal digestibility of forage soybean-based silages, with and without pearl millet, was determined along with evaluation of silages on heifer performance and reproductive function. Fermenters were utilized in a Latin square design and randomly assigned to 1 of the following treatments: 1) control diet of alfalfa haylage (CON), 2) soybean silage (SB) or 3) soybean and pearl millet silage (SB×PM). All diets were formulated to meet or exceed nutrient requirements of replacement beef heifers targeted to gain 0.79 kg/d. These same diets were fed to 90 Angus-Simmental beef replacement heifers [body weight (BW) = 366 kg; body condition score (BCS) = 5.53; age = 377 ± 11 d] 65 d prior to timed artificial insemination (TAI). Heifers were randomly allotted by breed, BCS and BW to 1 of the 3 treatments, with 3 reps/treatment. Diets were terminated 21 d post-TAI and heifers were commingled and placed on a common diet. Pubertal status was determined by progesterone concentrations of 2 blood samples taken 10 d apart prior to both trial initiation as well as initiation of estrous synchronization. Ovulatory follicle diameter was determined at time of breeding by ultrasonography. Pregnancy diagnosis was accomplished 35 and 66 d post-TAI, respectively, to calculate TAI and end of season pregnancy rates. Neither SB nor SB×PM had an effect (P &gt; 0.37) on apparent ruminal digestion of nutrients compared to the CON. Final BW (414 kg; P ≥ 0.10) and BCS (5.28; P ≥ 0.26) for the heifers were similar among treatments. Likewise, there were no differences in TAI (48%; P &gt; 0.43) or overall breeding season (93%; P &gt; 0.99) pregnancy rates. Ovulatory follicle diameters (11.7 mm) was not different (P &gt; 0.19) among treatments. In summary, forage soybean-based silages, with and without pearl millet, was an acceptable alternative forage for developing replacement beef heifers.

The aim of this study was to evaluate the effect of presence of corpus luteum (CL) at the onset of timed AI (TAI) protocol, size of ovulatory follicle and oestrus expression on pregnancy rate using E2 cypionate (EC) or GnRH as ovulation inductor in Angus heifers. On day 0, heifers [n = 414; 317 ± 23 kg; 3.08 ± 0.21 body condition score (1 to 5 scale) and 22–24 months of age] received a 1-g progesterone intravaginal device (Primer®, Agener União, Brazil) and 2 mg of oestradiol benzoate, IM, (RIC-BE®, Agener União, Brazil). On day 8, the device was removed, were administered 500 μg of sodium cloprostenol, IM, (Estron®, Agener União, Brazil) and heifers were divided in 2 groups: EC [n = 213; 0.5mg EC, IM, (SincroCP®, Ouro Fino, Brazil) on day 8] and GnRH [n = 201; 25 μg of Licerelin, IM, (GestranPlus®, Agener União, Brazil) at TAI] TAI was performed 50 h after device removal. Heifers were tail painted (Tell Tail®, GEA, New Zealand) to control oestrus expression from device removal to TAI. Heifers were scanned (7.5-MHz linear transducer MediSono® P3V, USA) to determine presence of CL (Day 0), ovulatory follicle diameter (OFD; Day 10) and ovulation (Day 11). Pregnancy was diagnosed 30 days after TAI. Oestrus expression was confirmed when marking ink was removed on day 10. Data were analysed using logistic regression and ANOVA. Oestrus expression was higher (P < 0.05) in EC (93.90%, 200/213) compared to GnRH group (67.66%, 136/201). The OFD differed (P < 0.05) according to presence of CL on Day 0 (10.3 mm) or not (9.6 mm), but was similar in treatment groups. Ovulation rate did not differ in EC (85%, 17/20) and GnRH (95%, 19/20) groups. Pregnancy rates were similar (P > 0.05) between EC (52.58%, 112/213) and GnRH (59.20%, 119/201) groups; however, in heifers that expressed oestrus, rates were higher (P < 0.05) (EC = 56%, 112/200; GnRH = 71.3%, 97/136) than in those did not show (EC = 0%, 0/13; GnRH = 33.8%, 22/65). Presence of CL at onset of TAI protocol was associated with higher pregnancy (P < 0.05) in GnRH group (68%, 68/100) compared with EC (54.46%, 61/112) group, but in heifers without CL, pregnancy were similar for GnRH (50.5%, 51/101) and EC (50.5%, 51/101) groups. Heifers were also classified in 3 groups according OFD: small (≤9.5 mm), medium (9.6–11.5 mm), and large (≥11.6 mm) to verify its effect on oestrus expression and pregnancy. Medium and large OFD were implicated in higher (P < 0.05) oestrus expression [(EC = 100%, 51/51; 96.97%, 32/33) and (GnRH = 83.02%, 44/53; 97.44%, 38/39)] than small OFD [(EC = 85%, 51/60) and (GnRH = 23.8%, 15/63)]. Pregnancy was lower (P < 0.05) in small OFD groups (EC = 28.3%, 17/60; GnRH = 25.4%, 16/63). In medium and large OFD, pregnancy was similar in EC (72.55%, 37/51; 54.55%, 18/33) groups, but differed (P < 0.05) between GnRH large (82.05%, 32/39) and medium (62.26%, 33/53). Higher pregnancy was achieved in heifers with CL in the beginning of TAI protocol treated with GnRH. Size of ovulatory follicle was associated with higher oestrus behaviour and pregnancy rate in EC and GnRH treatments.

Two experiments in Nelore cows subjected to conventional protocol for timed AI (TAI) were designed to compare the influence of antral follicle count (AFC; 1) on the ovarian follicular dynamics and (2) conception rates. First, multiparous cows with high (G-high, ≥45 follicles; n = 43) and low AFC (G-low, ≤15 follicles; n = 32) were selected to receive a conventional oestradiol/progesterone-based TAI protocol for monitoring ovarian follicular dynamics. Second, AFC from 962 multiparous suckling cows (40 ± 15 days postpartum) subjected to a conventional protocol for TAI were classified as G-high (≥45 follicles; n = 194), G-intermediate (≥20 and ≤40 follicles, n = 397), and G-low AFC (≤ 15 follicles; n = 243). Pregnancy status was evaluated 30 to 35 days later. Data were analysed using ANOVA followed by the Tukey test and model of logistic regression (P ≤ 0.05). In the first study, the AFC were greater (P ≤ 0.05) in G-high than in G-low, but remained stable in both groups during the evaluations. The ovarian diameter, perimeter, and area were greater (P ≤ 0.05) in G-high than in G-low (28.3 ± 3.9 v. 20.5 ± 3.2 mm, 100.9 ± 13.2 v. 73.8 ± 13.9 mm, and 67.5 ± 16.4 v. 37.7 ± 11.7 mm2; respectively). Follicular growth rate, time of ovulation, corpus luteum diameter, and progesterone concentration were similar (P > 0.05) for cows with low or high AFC. Dominant follicles showed greater (P ≤ 0.05) diameter in G-low than G-high at D4 (7.3 ± 2.2 v. 6.2 ± 1.4 mm; P = 0.069, tendency), D8 (11.2 ± 1.8v. 9.5 ± 1.8 mm), D9 (12.3 ± 1.7 v. 10.6 ± 1.7 mm), and D10 (13.4 ± 1.3 v. 12.2 ± 1.8 mm), and the estimated diameter of the ovulatory follicle tended (P = 0.08) to be higher in G-low than in G-high (14.4 ± 1.5 v. 13.4 ± 2.1 mm). In the second study, AFC of G-low cows showed higher (P ≤ 0.05) conception rate (61.7%) than those of G-intermediate (52.9%) and G-high (49.5%) cows. In Nelore cows subjected to ovulation synchronization protocol, cows with high AFC showed higher ovarian measurements, but those with low AFC had large follicular diameters. Additionally, the cows with low AFC showed higher conception rate than groups with intermediate and high counts.
