155 NOVEL RECIPIENT SYNCHRONIZATION REGIMENS FOR SUCCESSFUL EMBRYO TRANSFER IN THE BRAZILIAN OCELOT FOLLOWING LONG-TERM FROZEN EMBRYO STORAGE

The Brazilian ocelot (Leopardus pardalis mitis) is threatened with extinction in southern Brazil due to habitat loss and poaching. As one component of a bi-national conservation program, efforts have been initiated to establish a Brazilian ocelot population in North American zoos through a combination of natural breeding and assisted reproduction. With improved efficiency, embryo freezing and transfer might be useful as a management tool to help achieve this conservation goal. The objectives of this study were to (1) compare two novel ovarian stimulation regimens for embryo recipient synchronization in ocelots and (2) assess embryo transfer success following long-term (7 yrs) storage of frozen ocelot embryos. Adult female ocelots (n = 8), housed individually at a breeding facility in southern Brazil, were used as recipients. Ovarian activity was monitored noninvasively by assessing fecal estrone metabolite concentrations via a validated enzyme immunoassay. Initial fecal monitoring indicated that all females displayed active ovarian cyclicity. For embryo transfer, fecal samples were collected from each cat and assayed daily over a three week period. Females (n = 4) showing two or more consecutive days of increasing estrone were classified as estrual and treated (Trt 1) with two injections of porcine luteinizing hormone (pLH, 3000 IU dose–1, IM, 13 h interval). Females (n = 4) exhibiting consecutive days of basal estrone levels were considered interestrual and received a combination regimen (Trt 2) of equine chorionic gonadotropin (eCG, 400 IU, IM) followed 85 h later by pLH (3000 IU, IM). Females were anesthetized 50 h after the second hormone injection and evaluated laparoscopically to assess ovarian response. All females exhibited at least one fresh corpus luteum (CL) indicating proper timing of ovulation induction. The number (mean ± SEM) of ovarian follicles (0 v. 3.0 ± 1.5) and CLs (1.0 ± 0.0 v. 3.8 ± 1.8) did not differ (P > 0.05) between treatments. Upon confirmation of ovulation, 3 or 4 frozen embryos (24 embryos total), produced by IVF and frozen in ethylene glycol for liquid nitrogen storage 7 years earlier, were immediately thawed and transferred laparoscopically into one oviduct of each female. At 83 to 84 days post-transfer, two females (one per Trt) each gave birth to one healthy kitten, whereas a third female (Trt 2) experienced dystocia, requiring a C-section to deliver a single healthy offspring. Overall, 38% (3/8) of recipients became pregnant with 33% (3/9) of transferred embryos in pregnant females developing to term. Results indicate that these novel recipient synchronization regimens produce consistent ovulation and a suitable maternal environment for ocelot embryo transfer and that frozen ocelot embryos retain developmental competence after years of storage. Our findings suggest that frozen embryo transfer in ocelots has adequate efficiency for applied usage, allowing international shipment of frozen embryos to be used as a viable alternative to the transport of living ocelots for genetic management (NCRR 015338).

Obesity has been associated with altered reproductive activity in mares, and may negatively affect fertility. To examine the influence of long-term high-energy (HE) feeding on fertility, Shetland pony mares were fed a diet containing 200% of net energy (NE) requirements during a three-year study. The incidence of hemorrhagic anovulatory follicles (HAF) and annual duration of cyclicity were compared to those in control mares receiving a maintenance diet. Day-7 embryos were flushed and transferred between donor and recipient mares from both groups; the resulting conceptuses were collected 21 days after transfer to assess conceptus development. HE mares became obese, and embryos recovered from HE mares were more likely to succumb to early embryonic death. The period of annual cyclicity was extended in HE compared to control mares in all years. The incidence of HAFs did not consistently differ between HE and control mares. No differences in embryo morphometric parameters were apparent. In conclusion, consuming a HE diet extended the duration of cyclicity, and appeared to increase the likelihood of embryos undergoing early embryonic death following embryo transfer.

AbstractRadon (Rn), a radioactive element, has especial interest in medical geology because long-term exposure to high concentration is related to lung cancer. In this study, outdoor and indoor radon measurements were conducted in dwellings of the Piquiri Syenite Massif, located in southern Brazil, given the relative high Rn content in soils of this region. Measurements were done using CR-39 detectors and placing them inside and outside dwellings. Moreover, a one-dimensional diffusion model was performed in order to quantify the natural transport of Rn to the air in confined and aerated environments. Results indicate that the region presents relatively low air Rn concentrations, within the environmental limits; however, the health risk might increase in confined and ill-ventilated environments because of transfer from soil and exhalation from ornamental rock-material often found inside dwellings. The main north facies of the syenite, where most of the rock extractions are located, was found to have the highest air Rn concentration because of the higher soil Rn concentration, compared to other facies of the syenite.

Abstract A new approach to define heavy and extreme rainfall events based on cluster analysis and area-average rainfall series is presented. The annual frequency of the heavy and extreme rainfall events is obtained for the southeastern and southern Brazil regions. In the 1960–2004 period, 510 (98) and 466 (77) heavy (extreme) rainfall events are identified in the two regions. Monthly distributions of the events closely follow the monthly climatological rainfall in the two regions. In both regions, annual heavy and extreme rainfall event frequencies present increasing trends in the 45-yr period. However, only in southern Brazil is the trend statistically significant. Although longer time series are necessary to ensure the existence of long-term trends, the positive trends are somewhat alarming since they indicate that climate changes, in terms of rainfall regimes, are possibly under way in Brazil.

Abstract. In compliance with the development of the Water Framework Directive, there is a need for an integrated management of water resources, which involves the elaboration of reservoir management models. These models should include the operational and technical aspects which allow us to forecast an optimal management in the short term, besides the factors that may affect the volume of water stored in the medium and long term. The climate fluctuations of the water cycle that affect the reservoir watershed should be considered, as well as the social and economic aspects of the area. This paper shows the development of a management model for Rules reservoir (southern Spain), through which the water supply is regulated based on set criteria, in a sustainable way with existing commitments downstream, with the supply capacity being well established depending on demand, and the probability of failure when the operating requirements are not fulfilled. The results obtained allowed us: to find out the reservoir response at different time scales, to introduce an uncertainty analysis and to demonstrate the potential of the methodology proposed here as a tool for decision making.

In Florianópolis, southern Brazil, the venerid clam Anomalocardia brasiliana has supported subsistence and small-scale commercial fisheries for decades. The introduction of a hand dredge (gancho) since 1987 led to the development of a significant fishery supplying both local and regional shellfish markets. In 1992 one of the main fishing areas in the region was designated as the first Brazilian Marine Extractive Reserve (Pirajubaé RESEX), a federal form of governance intended to promote sustainable exploitation of natural resources by assigning exclusive fishing rights to traditional users. However, excessive fishing effort, institutional shortcomings and lack of a negotiated management plan have resulted in the overexploitation of the species since 2000. This study was aimed at evaluating the efficiency and selectivity of the hand dredge currently in use at the RESEX, through a field experiment conducted in October 2006. Quantitative samples of A. brasiliana were collected before and after dredging 15 experimental plots. Additional samples were obtained inside the dredge (catch) and respective cover cod-end (discard) for selectivity analysis. A single haul of the hand dredge can dislocate up to 76% of the individuals present in the sediment irrespective of their size, and retain up to 69% of the commercial-sized organisms. The gear has a knife-edge selection pattern, which enables the use of the minimum spacing between the iron bars of the dredge's basket as an effective management tool.

Objective: In the present study, we aimed to retrospectively evaluate the cumulative live birth rate (LBR) after up to three consecutive embryo transfer (ET) cycles, either fresh or frozen, in women with expected poor ovarian response (ePOR). Methods: We selected 115 women who entered the first <i>in vitro</i> fertilization (IVF) cycle between August 2013 and July 2016. The women were divided into an ePOR group (37 women) and a non-ePOR group (78 women). All women in the ePOR group were ≥40 years old or had serum anti-Müllerian hormone levels of less than 1.1 ng/mL at the time of the first IVF cycle. Live birth outcomes were monitored until December 2017. The cumulative LBR (with both conservative and optimistic estimates) was calculated according to the serial number of ET cycles. Results: After up to three ET cycles, the overall cumulative LBR was significantly lower in the ePOR group than in the non-ePOR group (conservative estimate, 10.8% vs. 44.9%, respectively; optimistic estimate, 14.7% vs. 56.1%, respectively; log-rank test, <i>p</i>=0.003). Conclusion: Women with ePOR exhibited a lower cumulative LBR than women in the non-ePOR group, and this information should be provided to ePOR women during counseling before starting IVF.

Abstract BackgroundThe outcome of in vitro fertilization-embryo transfer is often determined according to follicles and estradiol levels following gonadotropin stimulation. However, there is no accurate indicator to predict pregnancy outcome, and it has not been determined how to choose subsequent drugs and dosage based on the ovarian response. This study aimed to make timely adjustments to follow-up medication to improve clinical outcomes based on the potential value of estradiol growth rate. MethodsSerum estradiol levels were measured on the day of gonadotrophin treatment (Gn0), four days later (Gn4), seven days later (Gn7), and on the trigger day (HCG). The ratio was used to determine the increase in estradiol levels. According to the ratio of estradiol increase, the patients were divided into four groups: group A1 (Gn4/Gn0≤6.44), group A2 (6.44˂Gn4/Gn0≤10.62), group A3 (10.62˂Gn4/Gn0 ≤21.33), and group A4 (Gn4/Gn0>21.33); group B1 (Gn7/Gn4≤2.39), group B2 (2.39˂Gn7/Gn4≤3.03), group B3 (3.03˂Gn7/Gn4≤3.84), and group B4 (Gn7/Gn4>3.84). We analyzed and compared the relationship between data in each group and pregnancy outcome. ResultsIn the statistical analysis, the estradiol levels of Gn4 (P = 0.029, P = 0.042), Gn7 (P< 0.001, P = 0.001), and HCG (P< 0.001, P = 0.002), as well as the ratios of Gn4/Gn0 (P = 0.004, P = 0.006), Gn7/Gn4 (P = 0.001, P = 0.002), and HCG/Gn0 (P< 0.001, P< 0.001) both had clinical guiding significance, and the lower one significantly reduced the pregnancy rate. The outcomes were positively linked to groups A (P = 0.040, P = 0.041) and B (P = 0.015, P = 0.017). The logistical regression analysis revealed that group A1 (OR = 0.440 [0.223–0.865]; P = 0.017, OR = 0.368 [0.169–0.804]; P = 0.012) and B1 (OR = 0.261 [0.126–0.541]; P< 0.001, OR = 0.299 [0.142–0.629]; P = 0.001) had opposite influence on outcomes. ConclusionMaintaining a serum estradiol increase ratio at least above 2.39 on Gn7/Gn4 may result in a higher pregnancy rate. When estradiol growth is not ideal, gonadotrophin dosage should be adjusted appropriately to ensure the desired outcome.
