Implementing the use of a biobank in the endangered black-footed ferret (Mustela nigripes)

In the current global health climate, many conservation biologists are managing crisis situations, including increased species extinction rates. One strategy for securing wildlife populations into the future is to preserve biomaterials in genome resource banks (GRB; or ‘biobanks’). However, for GRBs to be successful we must understand the fundamental reproductive biology of species, along with developing assisted reproductive techniques (ARTs), including AI and semen cryopreservation. ART has been successfully used for several taxa, from amphibians to mammals, including ungulates, carnivores and primates. Not all these success stories implemented the use of a biobank, but one example that discussed herein is the black-footed ferret (Mustela nigripes) GRB. From a founder population of seven individuals, this species has been breeding in a managed setting for nearly 30 years. The goal of the breeding program is to maintain genetic integrity by ensuring each individual has the opportunity to pass his/her genes onto the next generation, while simultaneously providing animals for release into the wild. Scientists have used ART (e.g. AI) in the recovery program. Recently, semen from an individual of the founder population that was cryopreserved for up to 20 years was used successfully for AI, which improved the genetic diversity of the population. The black-footed ferret recovery program can serve as a model for other endangered species and demonstrates the usefulness of ART and GRBs to maintain highly endangered species into the future.

Pet cats receive bad press from conservationists. This is partly because there is an obvious link between pet cats and feral cats, and predation by feral cats is widely regarded as a key threatening process for endangered species of small vertebrates in Australia. There is also a perception, however, that pet cats are directly responsible for declines in wildlife populations, particularly around urban areas. As a consequence, many local government authorities have introduced regulations aimed at controlling cats, ranging from night-time curfews and confinement to complete prohibition of cat ownership. Greenaway (2010) discusses the approaches taken by local and state governments to control pet cats throughout Australia.

We discuss 12 themes that emerged from the set of case studies comprising the text, namely: (1) mechanisms matter for conservation; (2) physiology is just one source of knowledge; (3) physiology and behaviour are intertwined; (4) new tools and technologies should be embraced; (5) physiology can be valuable in captive settings; (6) conservation physiology extends across scales; (7) physiology can be incorporated into long-term monitoring programmes; (8) conservation physiology is applicable to invertebrates; (9) non-imperilled species deserve attention; (10) successful application is increased by co-production; (11) sharing success stories is important; and (12) findings should be communicated across a variety of platforms. We end the chapter with a discussion of some of the challenges currently being faced in the discipline, and with a message of optimism for the future.

The use of assisted reproductive techniques for endangered species is a major goal for conservation. One of these techniques, testis tissue xenografting, allows for the development of spermatozoa from animals that die before reaching sexual maturity. To assess the potential use of this technique with endangered species, testis tissue from six Iberian lynxes (one fetus, two perinatal cubs, two 6-month-old and one 2-year-old lynx), two Cuvier’s gazelle fetuses and one 8-month-old Mohor gazelle were transplanted ectopically into nude mice. Tissue from the lynx fetus, perinatal cubs and 2-year-old donors degenerated, whereas spermatogonia were present in 15% of seminiferous tubules more than 70 weeks after grafting in transplanted testis tissue from 6-month-old donors. Seminal vesicle weights (indicative of testosterone production) increased over time in mice transplanted with tissue from 6-month-old lynxes. Progression of spermatogenesis was observed in xenografts from gazelles and was donor age dependent. Tissue from Cuvier’s gazelle fetuses contained spermatocytes 40 weeks after grafting. Finally, round spermatids were found 28 weeks after transplantation in grafts from the 8-month-old Mohor gazelle. This is the first time that xenotransplantation of testicular tissue has been performed with an endangered felid and the first successful xenotransplantation in an endangered species. Our results open important options for the preservation of biological diversity.

This paper should begin with a brief defense of its title. “Variety” and “unsuccessful” are doubtful if not dirty words to most economists and many economic historians. The “success stories” of rapid development in Western Europe, Russia and Japan have been the most frequent subject of this Journal's articles on non-American topics. And the discovery of uniformity in the past, rather than variety, is admittedly essential to the development economist's search for predictability in the future that has informed so much of the economic history written since the Second World War.
