Research note: Micropropagation of Eucalyptus polybractea selected for key essential oil traits

A protocol for the micropropagation of Eucalyptus polybractea R.T. Baker (blue mallee) using axillary bud proliferation from lignotuber-derived explants is described. Three different ages of plants were used as explant sources: glasshouse-grown seedlings, field-grown saplings, and coppice of field-grown mature lignotubers. Explants from each source initiated successfully and no significant difference was observed for shoot proliferation, rooting success or hardening success between explant sources. Leaf oil quantity and quality for hardened clones transplanted to a field plantation were assessed after 3 months of growth. Ramets of all clones contained high quality oil with over 80% 1,8-cineole. For seedling-derived clones, foliar oil concentrations of ramets were higher than those of the ortets from which they were derived. For sapling and mature lignotuber derived clones the opposite was the case. This suggests that ontogenetic and physiological constraints may be influencing yield in the young ramets. The age of the explant source did not appear to influence the success of micropropagation, and as a result older plants (for which key oil traits are known) can be selected as elite plants for multiplying selected genotypes via micropropagation.

Abstract Laboratory experiments were conducted to determine the repellent activities of the ornamental plant Dioscorea sansibarensis leaf essential oil against the Bruchus chinensis bean weevil in the protection of stored legumes. The leaves exhibited potential repellency in a Y-tube olfactometer. The tabulated data show that 10µL of leaf oil exhibited a repellency of 41.33%, with a grouped median of 53.33%, which was better than 10 µL of Actellic 50 EC, which had a repellency of 36.00% and a grouped median of 40%. Statistically, there was no significant difference in the percentage repellency obtained from the leaf oil and Actellic 50 EC at different concentrations (10, 20, 30 and 40 µL/mL, X2(7, N=200) = 114.93, P< 0.05). The GC-MS analysis identified 16 compounds in the leaf oil. The main compounds and their percentage composition were phytol (19.46%), 1-epi-α-gurjunene (11.71%), palmitic acid (10.48%), ethyl palmitate (8.87%), methyl palmitate (7.72%), isophytol (5.99%), 2-heptadecanone (4.59%) and α-selinene (4.5%). The repellency caused by the by leaf oil may have been due to the presence of α-selinene, also known as naphthalene, 1,2,3,5,6,7,8,8a-octahydro-1,8a-dimethyl-7-(1-methylethenyl)-, [1S- (1.α.,7.α.,8a.α.)]-. Dioscorea sansibarensis may be a good choice for repellent formulations.

Objective The purpose of this study was to evaluate the quality of evidence of rhinology and rhinologic skull base surgery (RSBS) research and its evolution over the past decade. Study Design Review article. Setting We reviewed articles from 2007 to 2019 in 4 leading peer-reviewed otolaryngology journals and 3 rhinology-specific journals. Methods The articles were reviewed and levels of evidence were assigned using the Oxford Centre for Evidence-Based Medicine 2011 guidelines. High quality was defined as level of evidence 1 or 2. Results In total, 1835 articles were reviewed in this study spanning a 13-year period. Overall, the absolute number of RSBS publications increased significantly 22.6% per year, from 108 articles in 2007 to 481 in 2019 ( P < .001; 95% CI, 7.9-37.2). In 2007, only 13 articles, or 15%, were high quality, and this grew to 146 articles, or 39%, in 2019. A 14.0% per year exponential increase in the number of high-quality publications was found to be statistically significant ( P < .001; 95% CI, 7.2, 20.7). Overall, high-quality publications represented just 25.8% of RSBS articles overall. There was no significant difference in quality between rhinology-specific journals and general otolaryngology journals (χ2 = 3.1, P = .077). Conclusion The number of overall publications and of high-quality RSBS publications has significantly increased over the past decade. However, the proportion of high-quality studies continues to represent a minority of total RSBS research.

As one of the most commonly ordered imaging tests, the computed tomography (CT) scan comes with inevitable radiation exposure that increases cancer risk to patients. However, CT image quality is directly related to radiation dose, and thus it is desirable to obtain high-quality CT images with as little dose as possible. CT image denoising tries to obtain high-dose-like high-quality CT images (domain Y ) from low dose low-quality CT images (domain X ), which can be treated as an image-to-image translation task where the goal is to learn the transform between a source domain X (noisy images) and a target domain Y (clean images). Recently, the cycle-consistent adversarial denoising network (CCADN) has achieved state-of-the-art results by enforcing cycle-consistent loss without the need of paired training data, since the paired data is hard to collect due to patients’ interests and cardiac motion. However, out of concerns on patients’ privacy and data security, protocols typically require clinics to perform medical image processing tasks including CT image denoising locally (i.e., edge denoising). Therefore, the network models need to achieve high performance under various computation resource constraints including memory and performance. Our detailed analysis of CCADN raises a number of interesting questions that point to potential ways to further improve its performance using the same or even fewer computation resources. For example, if the noise is large leading to a significant difference between domain X and domain Y , can we bridge X and Y with a intermediate domain Z such that both the denoising process between X and Z and that between Z and Y are easier to learn? As such intermediate domains lead to multiple cycles, how do we best enforce cycle- consistency? Driven by these questions, we propose a multi-cycle-consistent adversarial network (MCCAN) that builds intermediate domains and enforces both local and global cycle-consistency for edge denoising of CT images. The global cycle-consistency couples all generators together to model the whole denoising process, whereas the local cycle-consistency imposes effective supervision on the process between adjacent domains. Experiments show that both local and global cycle-consistency are important for the success of MCCAN, which outperforms CCADN in terms of denoising quality with slightly less computation resource consumption.

Ocimum gratissimum L. leaf oil exhibited many pharmacological properties. This study aimed to formulate and evaluate the physical and chemical stability of O.gratissimum leaf oil nanoemulsion. O.gratissimum leaf oil was extracted by hydrodistillation. The major component of the essential oil eugenol, was analyzed by UV-Vis spectrophotometry. Nanoemulsions of O.gratissimum leaf oil were formulated using polysorbate 80, hyaluronic acid, poloxamer 188, and deionized water by phase inversion composition method. The hydrodynamic diameter, polydispersity index, and zeta potential value of O.gratissimum leaf oil nanoemulsion was evaluated by a dynamic light scattering technique. The %remaining of eugenol in the nanoemulsion was analyzed by UV-Vis spectrophotometry. The essential oil extracted from of O. gratissimum leaf oil was a clear, pale yellow color. The %yield of the essential oil was 0.15 ± 0.03% v/w. The size of the nanoemulsion was less than 106 nm. The polydispersity index of the nanoemulsion was ranging from 0.303 - 0.586 and the zeta potential value of the nanoemulsion was closely to zero, depending on the formulation component. O. gratissimum leaf oil at concentrations ranging from 0.002 - 0.012% v/v contained 35 - 41% of eugenol. The size of nanoemulsion was significantly decreased after storage at 4 °C, while significantly increased upon storage at 45 °C. The size of nanoemulsion stored at 30 °C did not significantly change. The %remaining of eugenol in the nanoemulsion was more than 90% after storage at 4 °C and 30 °C for 28 days. The percentage of eugenol remaining in the nanoemulsion stored at 45 °C was more than 85 - 90%, suggesting that the temperature affected the stability of eugenol in the nanoemulsion.

Even though the Lavandula species may be propagated by seeds, it should not be the preferred propagation method because it causes a great lack of uniformity. On the other hand, asexually propagated lavender crops would provide more homogeneous crops, and clones from high quality plant material would increase the odds for obtaining a higher quality essential oil. However, problems such as poor rooting and restrict market availability for superior clones have been a problem in vegetative propagation of the Lavandula species. The objective of this work was to define which type and size of cutting is more adequate for cutting propagation of L. dentata, a very productive Lavandula species. Cuttings with 5, 8, 10 or 13 cm and from the apical or basal parts of stems cut from L. dentata stock plants were placed in Plantmax HT® filled polystyrene foam trays and kept under intermittent mist system for two months. Averages of root number, length of the longest root, fresh and dry root weight, and percentage of rooting were evaluated. Apical cuttings combined 97.9% rooting with an average of 13.2 roots per cutting and basal cuttings 93.7% rooting with 2.98 roots per cutting. Apical cuttings with at least 10 cm in length were considered the most adequate for cutting propagation of L. dentata.

The chemical constituents of leaf and peel essential oil of Citrus medica L. were analysed by gas chromatography mass spectroscopy (GC-MS). Nineteen components accounting for 99.9% of the oil were identified in leaf oil. The major constituents are erucylamide (28.43%), limonene (18.36%) and citral (12.95%). The peel oil contains forty three components accounting for 99.8% of the total oil and the major components are isolimonene (39.37%), citral (23.12%) and limonene (21.78%). Objective: To verify if the use of ylang ylang essential oil by cutaneous application or inhalation alters the anxiety and self-esteem perception and physiological parameters as blood pressure and temperature. Method : A pilot study with 34 professionals from a nursing group randomized in three groups: one received the ylang ylang essential oil by cutaneous application, the second received through inhalation and the third (placebo) received the ylang ylang essence through cutaneous application. The assessment was done by an Anxiety Inventory (IDATE) and the Dela Coleta self-esteem scale, applied on baseline, after 30, 60 and 90 days and after 15 days post-intervention (follow up). Results : In the pre and post-intervention intergroup analysis, there was a significant difference in self-esteem for the three groups (p values: G1=0.014; G2=0.016; G3=0.038). There were no differences in the analysis between groups for anxiety or for physiological parameters. Conclusion : It was found significant alterations only to the intergroup perception of self-esteem for the three groups.

This study aims to examine the relationship between feedback environment practices and creativity in the educational organizations. This study is a quantitative study. A total of 40 polytechnic students from Polytechnic Kuching Sarawak are randomly selected to participate in this study. Data were obtained using a set of questionnaire consisting of three parts. The data obtained were analyzed descriptively and inferentially. Inferential analysis involves <em>t</em> test and Pearson correlation. The findings have shown that the level of feedback environment based on students' perception is high. Simultaneously, the level of creativity among the students is also high. The findings indicated that there is no significant difference in feedback environment between genders. Furthermore, the study also showed a significant and positive relationship between feedback environment and students’ creativity. The results of this study have several implications directly to the formation of high-quality students as feedback environment and students’ creativity are interrelated and inseparable in educational organizations.

Recent developments in cereal breeding for forage production have given the potential to greatly increase annual forage dry matter (DM) production. This paper reports on the findings from two cereal forage production trials on irrigated Canterbury land. Trial 1 studied the potential of a range of single and multi-grazed cereal forages over a 9 month period to produce high yields and quality from both grazing and whole-crop silage forage. Trial 2 studied the suitability of different cereal/ legume combinations for green-chop silage grown over a three month summer period. These two cereal forage systems, when combined in a 12 month period, produced in excess of 25 tonnes of high quality forage per hectare, almost double the DM production achieved using current perennial pasture based systems. Trial 1 showed no significant difference in the total DM produced by either single or multi-graze treatments. In Trial 2 pea/cereal combinations produced over 6 t DM/ha. Due to an earlier final harvest the multi-graze system is more easily combined with the summer crop and more likely to produce a lower cost and more sustainable forage system. Animal performance on forage produced in Trial 1 showed that dairy calves can grow well on cereal forages during winter. Keywords: cereal, feed supplements, forage, forage yield, legume, silage
