A preliminary survey of zearalenone and other mycotoxins in Australian silage and pasture

The oestrogenic mycotoxin zearalenone can occur in pasture and fodder infected with various Fusarium spp., and at concentrations exceeding 1 mg/kg, it has been associated with a reduction in the fertility of grazing ruminants. Pasture and fodder samples collected in four small studies from cattle and sheep farms in south-eastern Australia were tested for zearalenone and other mycotoxins using high performance liquid chromatography. Zearalenone in winter pasture was detected (viz. >0.1 and up to 5.0 mg/kg DM) in 8 of 22 pastures; three exceeded the tolerance concentration, 1.0 mg/kg. Zearalenone was detected in 15 of 24 samples of silage that were submitted by farmers for nutritional assessment, but that were not suspected of toxicity; 12 were in the range 1.0–80 mg/kg. Zearalenone was correlated with crude protein concentration (r = 0.59, P < 0.05); it was high in legume-dominant compared with grass-dominant silage (P < 0.04). In a separate investigation of 28 feed samples submitted for mycotoxin screening by nutritionists/veterinarians, zearalenone was detected in 7 of 13 pastures, in two of two hays and 9 of 13 silages (6 of 28 exceeded 1 mg/kg). Deoxynivalenol was detected in 6 of 13 pastures, one of two hays and 10 of 13 silages. Deoxynivalenol was high (0.64–1.76 mg/kg) in five silages of excellent/average appearance. Ergovaline and lolitrem B were detected at above tolerable concentrations in four of eight perennial ryegrass-dominant samples examined. Aflatoxins, ochratoxin A and fumonisins were detected in some samples although the concentrations were low. Two samples contained either zearalenone and ergovaline, or zearalenone and deoxynivalenol, where both mycotoxins exceeded tolerable concentrations.

Abstract The research material included 96 slaughter rabbit carcasses. Half of them came from the animals managed in small-scale backyard farming units where animals were fed a natural ingredient diet, while the other half was from rabbits kept under commercial production conditions and fed commercial rabbit pellets. The thigh and saddle muscle samples were collected from each carcass to establish a content of ubiquinone (CoQ10) and crude protein along with its collagen level. Determination of tissue coenzyme Q10 (UQ10) was carried out by high-performance liquid chromatography with some modification. Crude protein concentration was estimated using Kjeldahl procedure, while total collagen content by the method of Stegemann modified by Hurych-Chvapil, using hydrolysis according to Möhler and Volley. Ubiquinone level in slaughter rabbit tissue ranged between 76 and 127 μg/g tissue. The studies indicated that rabbit rearing system and muscle type are determinants of CoQ10 content. Meat of rabbits managed under the traditional backyard farming system exhibited higher CoQ10 concentration as compared to that determined in rabbits from the commercial rabbitry. Additionally, the CoQ10 level in the saddle was significantly higher than that in the thigh muscles, and the relationships was noted in both types of rabbit production systems. When the CoQ10 content was expressed per gram of fibrillar protein, there were not significant differences between saddle and thigh muscles. The correlation coefficient between ubiquinone and fibrillar protein averaged to 0.94. The studies also demonstrated a higher protein level in the saddle than in thigh muscles. However, no differences in protein concentration were reported in respect to the rabbit farming system. The protein composition in the saddle muscles, irrespective of a rabbit production system, revealed significantly lower collagen content compared to the proteins in thigh muscles. The obtained results and data from literature provide evidence that rabbit meat, especially from the traditional (organic) management system, is one of the best sources of animal protein and ubiquinone Q10.

Background. Lynch Syndrome (LS) is characterized by germline mutations in the DNA mismatch repair (MMR) genesMLH1,MSH2,MSH6,andPMS2. This syndrome is inherited in an autosomal dominant pattern and is characterized by early onset colorectal cancer (CRC) and extracolonic tumors. The aim of this study was to identify mutations inMMRgenes in three Mexican patients with LS.Methods. Immunohistochemical analysis was performed as a prescreening method to identify absent protein expression. PCR, Denaturing High Performance Liquid Chromatography (dHPLC), and Sanger sequencing complemented the analysis.Results. Two samples showed the absence of nuclear staining for MLH1 and one sample showed loss of nuclear staining for MSH2. The mutations found inMLH1gene were c.2103+1G>C in intron 18 and compound heterozygous mutants c.1852_1854delAAG (p.K618del) and c.1852_1853delinsGC (p.K618A) in exon 16. In theMSH2gene, we identified mutation c.638dupT (p.L213fs) in exon 3.Conclusions. This is the first report of mutations in MMR genes in Mexican patients with LS and these appear to be novel.

SUMMARYIn a factorial experiment the effect of two protein intakes and three patterns of feeding in the second pregnancy of 48 Large White x Wessex Saddleback sows was examined. The high protein (HP) diet (19·5% crude protein) contained 15% white fish meal. The low protein (LP) diet (10·5% crude protein) contained cereal protein only. Nutrient components of the diets differed in protein only. The pattern treatments involved allowances of 1·8 kg (L), 2·7 kg (C) and 3·6 kg (H) per day, the three pregnancy patterns being HL, C and LH with the changeovers made from the 49th to the 63rd day post coitum (p.c). Sows on the three pattern treatments received the same total amount of feed from 0–112 days p.c. and were treated alike at farrowing and during lactation.Fertility and parturition results were similar for all treatments, but the number of piglets alive after birth (when weighed) was least for LP sows on the HL pattern. At 3 weeks of age the size and weight of litters on HP sows were significantly greater than those on LP sows (P < 0·05 and < 0·001 respectively). More piglets were weaned by HP sows than LP sows (P < 0·05). HP sows gained more weight in pregnancy (P < 0·001) which was slightly longer, and lost more weight in lactation (P < 0·05) than LP sows.The HL pattern of feeding was associated with smaller live weight gains in pregnancy than the LH pattern (P < 0·001) and the total birth weight of HL litters was lighter than LH (P < 0·05), mean piglet weights being similar. Lactation performance was unaffected by pattern treatment.The main conclusion is that a low intake, particularly during the latter half of pregnancy, of protein which is of vegetable origin, is associated with decreased viability of the piglets at birth and in early suckling life, and with lower capacity of the sows for milk production.

Purpose – The purpose of this paper is to understand how management and employee perspectives of high-performance work systems (HPWS) relate to employee discretionary behaviour. In addition, the paper examines to what extent the relationship between employees’ perception of the HPWS and discretionary behaviour varies among different organizations/groups. Design/methodology/approach – Two samples were used in the study. The first sample included data from 51 managers and 1,023 employees from 26 manufacturing companies. The second sample included 52 managers and 6,382 employees from 42 manufacturing companies. Findings – The study shows that employee rated HPWS mediates the relationship between management rated HPWS and individual-level discretionary behaviour. Moreover, results showed that the effect of employee rated HPWS on discretionary behaviour varies among different organizations/groups. Practical implications – Results show that employee perceptions of the HPWS more strongly predict employees’ discretionary behaviour than management rated HPWS. Moreover, it shows that employees’ perceptions of the same HPWS, but operating in different organizational contexts exhibit different levels of discretionary behaviour. Originality/value – The study differentiates between management and employee perspectives of the HPWS. It also examines the variability of the relationship between HPWS and discretionary behaviour. Multilevel structural equation modelling is used to test the hypotheses.

1. The results of over 500 determinations of the heat of combustion of the urine produced by cattle and sheep have been analysed statistically. 2. The analytical errors for nitrogen, carbon and heat of combustion were ±0.54, ±1.4 and ±2.2%. The error attached to an estimate of the heat of combustion of the urine produced by an individual sheep in 4 days was ± 10%.3. At the maintenance level of feeding, the heat of combustion of the urine (U kcal/ 100 kcal food) was related to the crude protein content of the diet (P%)by the equationU = 0.25P+1.6with a residual standard deviation of ±0.88 kcal/100 kcal.4. Regression analysis of the relation between the heat of combustion of urine and its N content showed significant differences with diet. The heat of combustion of the urine of sheep was 9.7 kcal/g C and of cattle 10.3 kcal/g C, and did not vary with diet. 5. It is shown that the variation in the heat of combustion of urine/g N and its relative constancy/g C arises largely from variation, from diet to diet, in the proportion of the N excreted as hippurate. 6. The results have been combined with the results of a similar analysis (Blaxter & Clapperton, 1965) of methane production by sheep to show that the ratio of metabolizable energy to digested energy varies very little from mean values of 0.82 for roughages, 0.85 for cereals and 0.79 for oilseed cakes and meals.

The effect of replacing wilted grass silage (GS) with pea-barley intercrop silage (PBS) on feed intake, diet digestibility and milk production was studied with 8 multiparous Ayrshire-cows in a replicated 4 × 4 Latin square experiment. Proportion of PBS was 0 (PBS0), 33 (PBS33), 67 (PBS67) or 100 (PBS100) % of silage dry matter (DM). The DM content was 559 and 255 g kg-1 for GS and PBS. Crude protein content was 131 and 170 g kg-1 DM, respectively. Pea-barley silage was more extensively fermented than GS with total fermentation acid content of 120 vs. 12 g kg-1 DM. Silage was fed for ad libitum intake and supplemented with on the average 13 kg concentrate per day. Silage DM intake was 9.2 (PBS0), 9.7 (PBS33), 9.0 (PBS67) and 7.1 (PBS100) kg per day (Pquadr. < 0.05). The energy corrected milk yield [30.3 (PBS0), 29.8 (PBS33), 30.3 (PBS67), 31.3 (PBS100) kg per day] was not significantly affected by the treatment. Milk protein concentration decreased linearly (P < 0.05) in response to feeding PBS. It is concluded that PBS can replace up to two thirds of wilted, moderate quality GS in the feeding of dairy cows because in this experiment pure pea-barley silage reduced silage intake.

ABSTRACTAn exact pole inversion method is used to calculate the complete chain-axis orientation distribution from a single inter-chain reflection. A brief outline of the formulation and computation method is given. This method is demonstrated by comparing the measured chain-axis distribution from a (002) reflection of oriented polyethylene samples, to the calculated distribution from (110) and (200) reflections of the same sample. The error in the Hermans Orientation function is less than 0.5% for the two samples tested.
