Absorption of cadmium accompanied by EDTA varies according to tomato cultivar

Two tomato (Lycopersicum solanum) varieties, one high (YSL189) and one low (HZ903) cadmium (Cd) accumulator, were used in our experiment. We detected cadmium (Cd ion/Cd-EDTA) uptake rates in roots and the corresponding expression of the Cd transport genes IRT1, IRT2, ZIP, Nramp1, Nramp2 and Nramp3. Our data proved that both cultivars – YSL189 and HZ903 – showed higher Cd accumulation in plants and a higher Cd uptake rate in roots supplied with Cd ion than with Cd-EDTA. In roots of YSL189, the expression levels of IRT1, IRT2, ZIP, Nramp1, Nramp3 and Nramp2 (5, 10 and 20 µmolc L–1 Cd) treated with Cd ion were higher than those treated with Cd-EDTA, whereas in roots of HZ903, only two genes, IRT1 and Nramp1 (5, 10, 100 µmolc L–1 Cd), showed higher expression levels in plants treated with Cd ion than in those treated with Cd-EDTA. When the difference between the cultivars was considered, the Cd concentration in the plant and the Cd uptake rate in the roots of YSL189 were higher than those of YZ903 under the same Cd treatments (i.e. Cd ion or Cd-EDTA). The expression of IRT2 and ZIP in the roots of YSL189 was higher than that observed in HZ903 treated with all levels of ion-Cd. We attribute the higher Cd uptake rate and greater accumulation of ion-Cd compared with EDTA-Cd in YSL189 than those found in HZ903 partly to the genes that had higher expression levels. Our results indicate that the roles of transporters in the absorption of different forms of Cd vary according to plant genotype.

Internode length is an important agronomic trait affecting plant architecture and crop yield. However, few genes for internode elongation have been identified in tomato. In this study, we characterized an elongated internode inbred line P502, which is a natural mutant of the tomato cultivar 05T606. The mutant P502 exhibits longer internode and higher bioactive GA concentration compared with wild-type 05T606. Genetic analysis suggested that the elongated internode trait is controlled by quantitative trait loci (QTL). Then, we identified a major QTL on chromosome 2 based on molecular markers and bulked segregant analysis (BSA). The locus was designated as EI (Elongated Internode), which explained 73.6% genetic variance. The EI was further mapped to a 75.8-kb region containing 10 genes in the reference Heinz 1706 genome. One single nucleotide polymorphism (SNP) in the coding region of solyc02g080120.1 was identified, which encodes gibberellin 2-beta-dioxygenase 7 (SlGA2ox7). SlGA2ox7, orthologous to AtGA2ox7 and AtGA2ox8, is involved in the regulation of GA degradation. Overexpression of the wild EI gene in mutant P502 caused a dwarf phenotype with a shortened internode. The difference of EI expression levels was not significant in the P502 and wild-type, but the expression levels of GA biosynthetic genes including CPS, KO, KAO, GA20ox1, GA20ox2, GA20ox4, GA3ox1, GA2ox1, GA2ox2, GA2ox4, and GA2ox5, were upregulated in mutant P502. Our results may provide a better understanding of the genetics underlying the internode elongation and valuable information to improve plant architecture of the tomato.

Potassium depletion (KD) is a very common clinical entity often associated with adverse cardiac effects. KD is generally considered to reduce muscular Na-K-ATPase density and secondarily reduce K uptake capacity. In KD rats we evaluated myocardial Na-K-ATPase density, ion content, and myocardial K reuptake. KD for 2 wk reduced plasma K to 1.8 ± 0.1 vs. 3.5 ± 0.2 mM in controls ( P < 0.01, n = 7), myocardial K to 80 ± 1 vs. 86 ± 1 μmol/g wet wt ( P < 0.05, n = 7), increased Mg, and induced a tendency to increased Na. Myocardial Na-K-ATPase α2-subunit abundance was reduced by ∼30%, whereas increases in α1- and K-dependent pNPPase activity of 24% ( n = 6) and 13% ( n = 6), respectively, were seen. This indicates an overall upregulation of the myocardial Na-K pump pool. KD rats tolerated a higher intravenous KCl dose. KCl infusion until animals died increased myocardial K by 34% in KD rats and 18% in controls ( P < 0.05, n = 6 for both) but did not induce different net K uptake rates between groups. However, clamping plasma K at ∼5.5 mM by KCl infusion caused a higher net K uptake rate in KD rats (0.22 ± 0.04 vs. 0.10 ± 0.03 μmol·g wet wt−1·min−1; P < 0.05, n = 8). In conclusion, a minor KD-induced decrease in myocardial K increased Na-K pump density and in vivo increased K tolerance and net myocardial K uptake rate during K repletion. Thus the heart is protected from major K losses and accumulates considerable amounts of K during exposure to high plasma K. This is of clinical interest, because a therapeutically induced rise in myocardial K may affect contractility and impulse generation-propagation and may attenuate increased myocardial Na, the hallmark of heart failure.

Objective: To delineate the response of migrating chondrogenic progenitor cells (CPCs) that arose from the surface of mechanically injured articular cartilage to proinflammatory damage-associated-molecular-patterns (DAMPs). Design: Bovine CPCs and non-CPC chondrocytes isolated from either impacted or scratched articular cartilage were studied. Those 2 types of cells were treated with mitochondrial DAMPs (MTDs; 10 nM fMLF and 10 µg/mL CpG DNA), or 10 nM HMGB1, or 10 ng/mL IL-1b for 24 hours. At the end of experiments, conditioned media and cell lysates were collected for analysis of expression levels of matrix metalloproteinases (MMPs), chemokines, and cytokines that are associated with cartilage degeneration with Western blotting and quantitative polymerase chain reaction. The difference of expression levels was compared by Welch’s t-test. Results: Our data indicated that HMGB1 and MTDs remarkably upregulated pro-MMP-13 expression in CPCs. Compared with non-CPCs, CPCs expressed significantly more baseline mRNAs of MMP-13, CXCL12, and IL-6. MTDs greatly increased the expression of MMP-13 and IL-6 in CPCs by over 100-fold ( P < 0.001). MTDs also significantly increased IL-8 expression in CPCs to a similar extent ( P < 0.001). However, when IL-1b was present, CPCs expressed less MMP-3 and active MMP-13 proteins as well as less CCL2 and IL-6 than did non-CPCs. Conclusions: We concluded that CPCs were more sensitive than non-CPCs in response to DAMPs, especially MTDs. The proinflammatory nature of CPCs implied their critical role in the early phase of posttraumatic osteoarthritis development.

Abstract. We report here the results of three experiments, which are slight variations of the 15N method (JGOFS protocol) for determination of new production. The first two test the effect of (i) duration of incubation time and (ii) concentration of tracer added on the uptake rates of various N-species (nitrate, ammonium and urea) by marine phytoplankton; while the third compares in situ and deck incubations from dawn to dusk. Results indicate that nitrate uptake can be underestimated by experiments where incubation times shorter than 4h or when more than 10% of the ambient concentration of nitrate is added prior to incubation. The f-ratio increases from 0.28 to 0.42 when the incubation time increases from two to four hours. This may be due to the observed increase in the uptake rate of nitrate and decrease in the urea uptake rate. Unlike ammonium [y{=}2.07x{-}0.002\\, (r2=0.55)] and urea uptakes [y{=}1.88x{+}0.004 (r2=0.88)], the nitrate uptake decreases as the concentration of the substrate (x) increases, showing a negative correlation [y{=}-0.76x+0.05 (r2=0.86)], possibly due to production of glutamine, which might suppress nitrate uptake. This leads to decline in the f-ratio from 0.47 to 0.10, when concentration of tracer varies from 0.01 to 0.04μ M. The column integrated total productions are 519 mg C m-2 d-1 and 251 mg C m-2 d-1 for in situ and deck incubations, respectively. The 14C based production at the same location is ~200 mg C m-2 d-1, which is in closer agreement to the 15N based total production measured by deck incubation.

Objective. To investigate the influence of early bladder imaging (EBI) in experimental rabbits on the quantitative calculation of glomerular filtration rate (GFR) by the Gates method. Methods. We retrospectively analyzed the data of dynamic renal scintigraphy (DRS) in experimental rabbits. We calculated renal uptake during minutes 1-2 and 2-3 by correcting bladder radioactivity and computed the split GFR by renal uptake. Then, the EBI and GFR between 1-2 min and 2-3 min were compared, respectively. Results. The EBI proportion (57.3%) at 2-3 min of DRS was higher than that (8.5%) at 1-2 min ( P < 0.05 ). The correlations between the 1-2 min and 2-3 min uptake rates of unobstructed kidneys after correction ( r = 0.952 ‐ 0.979 ) were higher than those before correction ( r = 0.859 ‐ 0.936 ). However, the correlation between the two in obstructed kidneys was not improved ( r before = 0.967 versus r after = 0.968 ). For unobstructed kidneys, the difference in GFR based on 2-3 min uptake between before and after correction was significant ( P < 0.05 ), but not in obstructed kidneys ( P > 0.05 ). For GFR based on 1-2 min uptake, the difference between before and after correction was not significant in obstructed or unobstructed kidneys ( P > 0.05 ). Before correction, the GFR of unobstructed kidneys of 10.5% of the rabbits in the protein load test was lower than that in the baseline status, but not so after correction. Conclusion. The 2-3 min EBI on DRS has a significant influence on the GFR calculated by the Gates method in experimental rabbits. Controlling water intake or calculating the GFR by 1-2 min renal uptake helps to avoid the influence of EBI on GFR.

Cadmium (Cd) is present in many soils and, when entering the food chain, represents a major health threat to humans. Reducing Cd accumulation in plants is complicated by the fact that most known Cd transporters also operate in the transport of essential nutrients such as Zn, Fe, Mn, or Cu. This work summarizes the current knowledge of mechanisms mediating Cd uptake, radial transport, and translocation within the plant. It is concluded that real progress in the field may be only achieved if the transport of Cd and the above beneficial micronutrients is uncoupled, and we discuss the possible ways of achieving this goal. Accordingly, we suggest that the major focus of research in the field should be on the structure–function relations of various transporter isoforms and the functional assessment of their tissue-specific operation. Of specific importance are two tissues. The first one is a xylem parenchyma in plant roots; a major “controller” of Cd loading into the xylem and its transport to the shoot. The second one is a phloem tissue that operates in the last step of a metal transport. Another promising and currently underexplored avenue is to understand the role of non-selective cation channels in Cd uptake and reveal mechanisms of their regulation.

Abstract Aims Perinatal depression threatens the health of maternal women and their offspring. Although screening programs for perinatal depression exist, non-uptake of referral to further mental health care after screening reduces the utility of these programs. Uptake rates among women with positive screening varied widely across studies and little is known about how to improve the uptake rate. This study aimed to systematically review the available evidence on uptake rates, estimate the pooled rate, identify interventions to improve uptake of referral and explore the effectiveness of those interventions. Methods This systematic review has been registered in PROSPERO (registration number: CRD42019138095). We searched Pubmed, Web of Science, Cochrane Library, Ovid, Embase, CNKI, Wanfang Database and VIP Databases from database inception to January 13, 2019 and scanned reference lists of relevant researches for studies published in English or Chinese. Studies providing information on uptake rate and/or effectiveness of interventions on uptake of referral were eligible for inclusion. Studies were excluded if they did not report the details of the referral process or did not provide exact uptake rate. Data provided by observational studies and quasi-experimental studies were used to estimate the pooled uptake rate through meta-analysis. We also performed meta-regression and subgroup analyses to explore the potential source of heterogeneity. To evaluate the effectiveness of interventions, we conducted descriptive analyses instead of meta-analyses since there was only one randomised controlled trial (RCT). Results Of 2302 records identified, 41 studies were eligible for inclusion, including 39 observational studies (n = 9337), one quasi-experimental study (n = 43) and one RCT (n = 555). All but two studies were conducted in high-income countries. The uptake rates reported by included studies varied widely and the pooled uptake rate of referral was 43% (95% confidence intervals [CI] 35–50%) by a random-effect model. Meta-regression and subgroup analyses both showed that referral to on-site assessment or treatment (60%, 95% CI 51–69%) had a significantly higher uptake rate than referral to mental health service (32%, 95% CI 23–41%) (odds ratio 1.31, 95% CI 1.13–1.52). The included RCT showed that the referral intervention significantly improved the uptake rate (p < 0.01). Conclusions Almost three-fifths of women with positive screening results do not take up the referral offers after perinatal depression screening. Referral to on-site assessment and treatment may improve uptake of referral, but the quality of evidence on interventions to increase uptake was weak. More robust studies are needed, especially in low-and middle-income countries.

Our new technique allows the decline in ambient ammonium concentration to be followed continuously during phytoplankton uptake experiments. A self-cleaning, in-line filter permits the continuous separation of cells from the medium for nutrient determination by an autoanalyzer. The technique works well with laboratory cultures of the marine diatoms Thalassiosira pseudonana and Skeletonema costatum. Changes in uptake rate on time scales of 1 min or longer can be resolved, the resolution being limited by the response characteristics of the autoanalyzer.
