Significant population genetic structure detected for a new and highly restricted species of Atriplex (Chenopodiaceae) from Western Australia, and implications for conservation management

Atriplex sp. Yeelirrie Station (L. Trotter & A. Douglas LCH 25025) is a highly restricted, potentially new species of saltbush, known from only two sites ~30 km apart in central Western Australia. Knowledge of genetic structure within the species is required to inform conservation strategies as both populations occur within a palaeovalley that contains significant near-surface uranium mineralisation. We investigate the structure of genetic variation within populations and subpopulations of this taxon using nuclear microsatellites. Internal transcribed spacer sequence data places this new taxon within a clade of polyploid Atriplex species, and the maximum number of alleles per locus suggests it is hexaploid. The two populations possessed similar levels of genetic diversity, but exhibited a surprising level of genetic differentiation given their proximity. Significant isolation by distance over scales of less than 5 km suggests dispersal is highly restricted. In addition, the proportion of variation between the populations (12%) is similar to that among A. nummularia populations sampled at a continent-wide scale (several thousand kilometres), and only marginally less than that between distinct A. nummularia subspecies. Additional work is required to further clarify the exact taxonomic status of the two populations. We propose management recommendations for this potentially new species in light of its highly structured genetic variation.

Abstract We used DNA fingerprinting to assess genetic structure of populations in Cory's Shearwater (Calonectris diomedea). We analyzed mates and parent-offspring relationships, as well as the amount and distribution of genetic variation within and among populations, from the level of subcolony to subspecies. We found no evidence of extrapair fertilization, confirming that the genetic breeding system matches the social system that has been observed in the species. Mates were closely related, and the level of genetic relatedness within populations was within the range usually found in inbred populations. In contrast to previous studies based on allozymes and mtDNA polymorphism, DNA fingerprinting using microsatellites revealed consistent levels of genetic differentiation among populations. However, analyzing the two subspecies separately revealed that the pattern of genetic variation among populations did not support the model of isolation by distance. Natal dispersal, as well as historic and/or demographic events, probably contributed to shape the genetic structure of populations in the species.

Abstract. Sea sandwort (Honckenya peploides) was one of the first plants to successfully colonize and reproduce on the volcanic island Surtsey, formed in 1963 off the southern coast of Iceland. Using amplified fragment length polymorphic (AFLP) markers, we examined levels of genetic variation and differentiation among populations of H. peploides on Surtsey in relation to populations on the nearby island Heimaey and from the southern coast of Iceland. Selected populations from Denmark and Greenland were used for comparison. In addition, we tested whether the effects of isolation by distance could be seen in the Surtsey populations. Using two primer combinations, we obtained 173 AFLP markers from a total of 347 plant samples. The resulting binary matrix was then analysed statistically. The main results include the following: (i) Surtsey had the highest proportion of polymorphic markers as well as a comparatively high genetic diversity (55.5% proportion of polymorphic loci, PLP; 0.1974 HE) and Denmark the lowest (31.8% PLP; 0.132 HE), indicating rapid expansion during an early stage of population establishment on Surtsey and/or multiple origins of immigrants; (ii) the total genetic differentiation (FST) among Surtsey (0.0714) and Heimaey (0.055) populations was less than half of that found among the mainland populations in Iceland (0.1747), indicating substantial gene flow on the islands; (iii) most of the genetic variation (79%, p < 0.001) was found within localities, possibly due to the outcrossing and subdioecious nature of the species; (iv) a significant genetic distance was found within Surtsey, among sites, and this appeared to correlate with the age of plant colonization; and (v) the genetic structure analysis indicated multiple colonization episodes on Surtsey, whereby H. peploides most likely immigrated from the nearby island of Heimaey and directly from the southern coast of Iceland.

The Epicopeiidae is a small geometroid family distributed in the East Palaearctic and Oriental regions. It exhibits high morphological diversity in body size and wing shape, while their wing patterns involve in various complex mimicry rings. In the present study, we attempted to describe a new genus, and a new species from Vietnam, with comments on two assumed congeneric novel species from China and India. To address its phylogenetic affinity, we reconstructed the phylogeny of the family by using sequence data of COI, EF-1α, and 28S gene regions obtained from seven genera of Epicopeiidae with Pseudobiston pinratanai as the outgroup. We also compared the morphology of the new taxon to other epicopeiid genera to affirm its taxonomic status. The results suggest that the undescribed taxon deserve a new genus, namely Mimaporia gen. n. The species from Vietnam, Mimaporia hmong sp. n., is described as new to science. Under different tree building strategies, the new genus is the sister group of either Chatamla Moore, 1881 or Parabraxas Leech, 1897. The morphological evidence, which was not included in phylogenetic analyses, however, suggests its potential affinity with Burmeia Minet, 2003. This study also provides the first, although preliminary, molecular phylogeny of the family on which the revised systematics and interpretation of character evolution can be based.

During the investigation of fungal microbiome associated with boxwood in the Czech Republic, samples from Buxus sempervirens L. (Buxaceae) plants were collected and used for isolation. Two fungal strains were proposed as a new species Pyrenochaetopsis kuksensis based on morphology as well as phylogenetic analyses of ITS, LSU, rpb2, and tub2 sequence data. Detailed descriptions and phylogenetic relationships of the new taxon are provided.

Summary Cryptaphelenchus baujardi n. sp. is described and illustrated based on two populations from Golestan province, northern Iran. The new species is characterised by female and male body length of 224 (190-261) and 215 (195-229) μm, respectively, offset cephalic region with a shallow constriction, delicate stylet 6.6 (6.0-7.0) μm long, four incisures in the lateral field, presence of a rudimentary post-uterine sac in the female gonoduct, elongate-conoid female tail ending in an acute to finely rounded terminus, male tail conical, male with seven caudal papillae and delicate spicules. The new species shows a strong resemblance to C. varicaudatus and C. iranicus, but has a sclerotised mass near the tip of the spicule and an indistinct spermatheca. Phylogenetic analyses based on both partial SSU and LSU rDNA sequences confirm its status as a new taxon.

An ongoing survey for novel phytoplasmas and viruses that affect palms (Arecaceae) is being conducted in Costa Rica along with potential vectors. During that survey, a new species of derbid planthopper (Hemiptera: Fulgoroidea) in the genus Omolicna Fennah was detected from Heredia and Limón State and is here described as Omolicna xavieri sp. n. This new taxon was first found on declining coconut palms (Cocos nucifera) and subsequently on healthy coconut palms in Tortuguero National Park. Also, two additional species of Omolicna, O. brunnea and O. triata, were collected on coconut in the same habitat and represent new country records. There are no previously published records of Omolicna from Costa Rica. Sequence data for the cytochrome c oxidase subunit I (COI) and the 18S ribosomal RNA gene were obtained. In addition, novel 18S primers specific to auchenorrhynchan insects were developed and presented. The molecular data show high pairwise distances between O. xavieri sp. n. and other Omolicna species (24% to 31% and 10% using COI and 18S, respectively). Based on both morphological and sequence data, we erect and briefly describe the new subgenus Agoo for the new species to reflect the observed differences. A checklist of Omolicna species is presented.

Genetic variation between and within populations of Trifolium glomeratum (cluster clover) was studied using seed collected from 2 sites in Western Australia: Mount Barker in the south and Kwelkan in the wheatbelt. Seed was collected at 64 subplots within each site and the material was grown at the University Field Station at Shenton Park, Perth. Seventeen morphological characters were scored and the results were analysed using analysis of variance, principal components analysis, and cluster analysis. Within-site variation was much greater than had previously been shown, and a considerable amount of between-site variation was present. It is suggested that within-site variation is due to a small amount of heterozygosity, as a result of limited outbreeding, being present in each population. The 2 populations are shown to be distinct from each other, with the population from Mount Barker containing more within-site variation. It is suggested that this is a result of climatic stress influencing and reducing the amount of variation being maintained in the Kwelkan population.

ABSTRACT Tudivasum Rosenberg & Petit, 1987 is a morphologically distinct gastropod genus of low diversity. All but one species are known from Australia and they occur from the intertidal zone down to hundreds of metres on the continental shelf. These carnivorous gastropods are thought to have intracapsular development. The six currently recognized extant species are reviewed here and their geographical ranges clarified. Two new species, Tudivasum chaneyi n. sp. and T. ashmorense n. sp., are described from Ashmore Reef, Western Australia, and are characterized by differences in protoconch colour and shell sculpture. The third new species, T. westrale n. sp., is described from the mid-west coast of Western Australia, where it has long been misidentified as T. spinosum (H. Adams & A. Adams, 1864). We generated a molecular phylogeny based on mitochondrial DNA sequence data to test morphological species concepts and reconstruct relationships among four of the described species. High levels of divergence within one of the new species could indicate an additional cryptic species.

A new species of the eyeless eleotrid genus Milyeringa is described from wells sunk on Barrow Island, Western Australia. Milyeringa justitia n. sp. is the third species of the genus to be named. Morphological data and cytochrome c oxidase subunit I (COI) DNA sequence data from a wide sample of localities at which the genus occurs was used to evaluate relationships and species limits. Milyeringa veritas is redescribed, and M. brooksi is synonymised with M. veritas. The unique form and ecology of these fishes, plus the threats to their survival, warrants immediate and continuing attention in management.

The systematics of the genus Hannia Vari 1978, endemic to freshwater habitats of remote north-western Australia, is revised in light of recent collections in the region and a molecular study of the group that identified an undescribed candidate species. A new freshwater fish species (Hannia wintoni sp. nov) is described based on analysis of multiple nuclear genetic markers (53 allozyme loci), mitochondrial DNA sequence data (601 bp cytochrome b) and morphology (examination of a suite of 66 morphometric and meristic characters). Head profile, postorbital length, maximum length, preopercular spines and pectoral-fin rays are characters that best distinguish H. wintoni sp. nov from its only congener, H. greewayi. While the existing description of H. greenwayi is robust and accurate, we present a number of additional characters that enhance to the original description, based on type and fresh material. Information on the known distribution, habitats and conservation status of the two species is summarised. The new species is a narrow-range endemic.
