The phenylthiohydantoins (PTHs) derived from amino acids were hydrolysed in boiling hydriodic acid for 24 hr. Good yields of free amino acids were obtained for all PTH derivatives except methionine. In contrast to hydrolysis with hydrochloric acid, hydrolysis with hydriodic acid converts PTH-threonine, PTH-serine, and PTH-tryptophan respectively to oc-amino-n-butyric acid, alanine, and a mixture (approx. 2: 1) of glycine and alanine. This procedure provides a useful adjunct to thin-layer chromatography and ultraviolet spectroscopy for quantitative identification of the PTH derivative.

We isolated the steroidal saponins from the rhizomes of Polygonatum odoratum (Mill.) Druce with an efficiency of 4.50% which represents 7 fractions identified by thin-layer chromatography (TLC), of which 3 were furostanics and 4 spirostanics. After the acid hydrolysis of the saponins, one aglycone (sapogenine) was identified by TLC. Further, we have determined the physico-chemical and the biological properties of the isolated saponins.

Nineteen free amino acids and four ninhydrin-positive compounds were detected in the haemolymph of fifth-instar nymphs of Rhodnius prolixus by the technique of thin-layer chromatography. On the sixth day after feeding, tyrosine and tryptophan appeared in the haemolymph. Their concentration increased until the 12th day when testing stopped. The concentrations of alanine, asparagine, cystine, isoleucine, phenylalanine, and threonine fluctuated moderately from day to day. The results obtained by thin-layer chromatography differed from those of a similar study in which the technique of paper chromatography was used.

1. A system of separation using buffered Celite columns is described that enables the pipsyl derivatives of most of the common amino acids to be separated. 2. The reaction of pipsyl chloride with several amino acids not included in previous studies has been investigated. In particular, knowledge of the acid-soluble pipsyl derivatives of arginine, histidine, lysine, tyrosine and cysteic acid has been extended. 3. Reproducible factors have been obtained that enable corrections to be applied for the breakdown of pipsylamino acids on acid hydrolysis. 4. The reaction of pipsyl chloride with peptides has been studied under various conditions. 5. The extent of the reaction between pipsyl chloride and insulin depends on the nature of the solvent-buffer system, and under the best conditions so far found is about 75% complete. 6. In an Appendix, the separation of pipsylamino acids by thin-layer chromatography is described.

Quantitative investigation of Sceliphron caementarium venom, obtained by electrical excitation of the wasp, has shown an unexpectedly small protein fraction of the dried venom. A few percent of the venom consists of the free amino acids histidine, methionine, and pipecolic acid. Three other components have been isolated by paper and thin-layer chromatography and are found to give weak ninhydrin reactions, in addition to certain color tests characteristic of alkaloids.
