Using plant wax markers to estimate diet composition and intakes of mixed forages in sheep by feeding a known amount of alkane-labelled supplement

The feeding of known amounts of supplements to grazing animals can be accomplished relatively easily. If the supplement and the other diet components have distinctive profiles of cuticular wax n-alkanes, then the supplement intake and the alkane profiles of the supplement, other dietary components and faeces can be used to estimate the proportions and hence intakes of several forages by the grazing animal. However, this method requires knowledge of recoveries of n-alkanes in faeces. Twenty four wethers were fed one of four diets comprising equal combinations of 1, 2, 3 or 4 forages. Forages used were subterranean clover, phalaris, annual ryegrass and wheat straw. Forages were chopped using a chaff cutter and fed with solvent-extracted cottonseed meal (CSM) labelled with beeswax and synthetic C28 alkane to provide a characteristic alkane profile. Faecal grab samples were taken from sheep from 14 to 23 days after administration of an intra-ruminal controlled-release device (CRD) containing 1 g of each of C32 and C36 alkane. Total faeces were collected from half the sheep on each treatment in order to measure alkane recoveries in individual sheep. Faecal concentrations of the n-alkanes C25 to C31 and C33 were corrected for recovery using the individual sheep value, the treatment mean or the grand mean for all four treatments. Dietary compositions were then estimated from corrected faecal concentrations of n-alkanes using a least-squares procedure and, together with the known supplement intakes, were used to estimate the intakes of all other diet components. Estimates from this ‘labelled supplement’ method were compared with the amounts fed or those estimated using the alkanes derived from the CRD. The labelled supplement method accurately and precisely estimated dietary component proportions and intake for all treatments when measured recoveries for individual sheep were used. Precision declined when recovery was based on estimated recoveries for treatment means or the grand mean. Estimates of intake based on dietary C33 and the measured release rates of C32 or C36 alkanes from the CRD did not differ from measured intakes. Estimates based on the C32 : C31 alkane pair over-estimated intake. Estimates of whole-diet digestibility based on the various ways of estimating intake were all very close to the digestibilities calculated from directly-measured intakes and faecal outputs. It is concluded that the feeding of a known amount of supplement can be successfully used to estimate dietary proportions and hence intakes of diet components, in mixed diets with up to five ingredients, but this approach requires estimates of faecal alkane recovery.

Knowledge of the feeding ecology of mammalian herbivores is fundamental in predicting their responses to habitat change. Where native woodlands are newly established in open moorland, the extent to which trees form part of the diet of mountain hares (Lepus timidus) is unknown. This information is necessary for predicting the potential effects of mountain hare browsing on woodland establishment. The n-alkanes and a long-chain fatty alcohols found in the cuticular wax of diet plants and faeces (N = 240) were used as markers to estimate the composition of the diet of mountain hares in an area of moorland with newly established Pinus sylvestris and Betula pubescens woodland. During winter, the diet of mountain hares was dominated by Calluna vulgaris, but there was a seasonal shift to a diet dominated by grasses, sedges, and rushes in summer. Pinus sylvestris and B. pubescens were minor dietary components in all seasons. A higher proportion of grasses, sedges, and rushes was found in the diet of lactating females. Results suggest that when an alternative browse species such as C. vulgaris is widely available, mountain hares may not have a large impact on the establishment of native woodland. The dietary results from this study are in broad agreement with those from previous studies using other techniques.

Seedlings of 3 perennial grasses, Danthonia linkii Kunthcv. Bunderra, D. richardsonii Cashmore cv. Taranna(wallaby grasses), and Phalaris aquatica L. cv. Sirosa,were each grown in replacement series mixtures with seedlings ofTrifolium repens L. (white clover),Trifolium subterraneum L. var. brachycalycinum (Katzn.et Morley) Zorahy & Heller cv. Clare (subterraneanclover), and Lolium rigidum L. (annual ryegrass). Plantswere sown 5 cm apart in boxes (45 by 29 by 20 cm) at a density of 307plants/m2. Maximum likelihood estimates were usedto derive parameters of a non-linear competition model using the dry matterweights of perennial grasses and competitors at 3 harvests, approximately 168,216, and 271 days after sowing. Intra-plant competition was examined inmonocultures of each species, grown at plant spacings of 2, 5, and 8 cm apartwith plants harvested at the above times.Competition occurred in all perennial grass–competitor mixtures, exceptin those of each perennial grass with white clover and thephalaris–subterranean clover mixture (Harvest 1) and those withD. richardsonii and phalaris grown with white clover(Harvest 2). For D. richardsonii (Harvests 1 and 2) andD. linkii (Harvest 1 only) grown with white clover andthe phalaris–subterranean clover (Harvest 1), the two species in themixture were not competing. In the phalaris–white clover mixture, eachspecies was equally competitive (Harvests 1 and 2). These differences incompetition and aggressiveness reflected differences in individual plantweights in monocultures where there was an effect (P < 0.05) of species ondry matter weight per box, but no significant effect of plant spacing.These data indicated that for successful establishment,D. richardsonii and D. linkiishould not be sown in swards with either subterranean clover or white clover,or where populations of annual ryegrass seedlings are likely to be high.Phalaris was more compatible with both white clover and subterranean clover,but aggressively competed with by annual ryegrass.

Nutrigenomics and lifestyle factors play a vital role in health management and represent an exceptional prospect for the improvement of personalized diets to the individual at risk of developing diseases like cancer. Concerning cancer as a multifactorial genetic ailment, several aspects need to be investigated and analysed. Various perspectives should be researched and examined regarding the development and prognosis of breast and colon cancer. Malignant growth occurrence is anticipated to upsurge in the impending days, and an effective anticipatory strategy is required. The effect of dietary components, basically studied by nutrigenomics, looks at gene expression and molecular mechanisms. It also interrelates bioactive compounds and nutrients because of different 'omics' innovations. Several preclinical investigations demonstrate the pertinent role of nutrigenomics in breast and colon cancer, and change of dietary propensities is conceivably a successful methodology for reducing cancer risk. The connection between the genomic profile of patients with breast or colon cancer and their supplement intake, it is conceivable to imagine an idea of personalized medicine, including nutrition and medicinal services.

Barley grass (Hordeum spp.) is a relatively short lived annual that provides high quality grazing early in the season, but its seed heads cause contamination of wool and carcasses, and may irritate the mouth, eyes and nose of sheep. Treatments were imposed on established subterranean clover (Trifolium subterraneum L.) annual pasture in the same plots for three consecutive years (2015 to 2017) to evaluate changes in barley grass content. Treatments included: grazing alone (G), herbicide followed by grazing (HG), or a forage conservation harvest in early October, late October or early November consistent with an early silage harvest (ES), late silage harvest (LS) or hay cut (H). Grazing plus herbicide markedly reduced (P &lt; 0.05) barley grass numbers compared with all other treatments, but increased (P &lt; 0.05) the growth of annual ryegrass (Lolium rigidum L.). ES reduced (P &lt; 0.05) barley grass and increased (P &lt; 0.05) subterranean clover compared with H, but broadleaf weed content benefitted by LS in contrast to either ES or H. Although herbicide application was the most effective method for barley grass control, forage harvest timing could be used to beneficially manipulate pasture composition.

SummaryAn experiment was carried out to evaluate the use of alkanes for estimating diet composition of goats and sheep offered three different dietary treatments. Twelve animals as two groups of 4 crossbred goats (G1, 24 kg live weight; G2, 22 kg) and 4 crossbred sheep (S, 26 kg live weight), were housed in metabolism pens. Animals were offered daily a total of 1 kg DM/100 kg live weight. G1 received 70% ryegrass (Lolium perenne) and 30% gorse (Ulex gallii), G2 received 70% ryegrass and 30% heather (Erica sp.) and S group ate 100% ryegrass. Diet composition was estimated from the alkane concentrations (using all alkanes from C23 to C36 or only odd-chain alkanes C27, C29, C31 and C33) in diet and faeces (with or without correction for incomplete faecal recoveries) using least-squares procedures.Dietary treatment and animal species significantly affected alkane faecal recoveries, except for C24 and C36. When applying the faecal recovery corrections, there were no significant differences between measured proportions of dietary components and those estimated using all alkanes or odd-chain alkanes. In contrast, the proportions calculated without faecal recovery correction differed significantly (P<0.05) from the actual proportions and over-estimated the amount in the diet of those plant components with higher concentrations of long-chain alkanes (Erica sp. and Lolium perenne). The results indicate that alkanes are useful markers to estimate diet composition, however, it was observed that animal species and diet composition influenced the faecal recovery of alkanes. This suggests that the use of the alkane methodology for estimating the diet selection of grazing animals should be preceded by a calculation of the actual alkane faecal recoveries for each experimental condition.

Stone martens (Martes foina) are documented as generalist throughout their distributional range whose diet composition is affected by food availability. We tested if this occurs and what feeding strategies it follows in a typical Mediterranean ecosystem in Central Greece by analysing contents from 106 stomachs, seasonally collected from three different habitats during 2003–2006. Seasonal variation in diet and feeding strategies was evident and linked to seasonal nutritional requirements, but possibly imposed by strong interference competition and intraguild predation. Fleshy fruits and arthropods predominated in the diet, but also mammals and birds were frequently consumed. An overall low dietary niche breadth (BA=0.128) indicated a fruit specialization tendency. A generalised diet occurred in spring with high individual specialisation, whereas more animal-type prey was consumed than fruits. A population specialization towards fruits was indicated during summer and autumn, whereas insects were consumed occasionally by males. In those seasons it switched to more clumped food types such as fruits and insects. In winter it selectively exploited both adult and larvae insects and partially fruits overwinter on plants. The tendency to consume particular prey items seasonally reflected both the population specialist behaviour and the individual flexibility preyed on different food resources.

To control the nematode Anguina funesta (considered by some to be A. agrostis), the vector in annual ryegrass toxicity, pasture management treatments were applied before and after gall initiation. Before gall initiation, the herbicides paraquat and diclofop methyl were used to remove ryegrass. These treatments gave good control of the nematode, but reduced pasture production in winter when feed is generally scarce. Diclofop methyl had no detrimental effect on legume growth, and was more effective on actively growing ryegrass than was paraquat, which severely retarded subterranean clover. Desiccating the ryegrass heads, with mechanical topping or the desiccant herbicide paraquat, after gall initiation, but before hatching of the second-stage juveniles ( J2s ) , gave good control of the nematode population. The main limitations with mechanical topping are that the ryegrass heads must be tall enough to cut before the J2s hatch and not all paddocks are suitable for topping. With paraquat, the main limitations are that treated material was susceptible to weathering and there was less regrowth to set seed. Oxamyl, a systemic nematicide applied after gall initiation, had no effect on the nematode, nor had spraying pasture with the nematode parasitic fungus Verticillium sp.

The endangered black-tailed dusky antechinus (Antechinus arktos) was described in 2014, so most aspects of its ecology are unknown. We examined diet composition and prey selection of A. arktos and a sympatric congener, the northern form of A. stuartii, at two sites in Springbrook National Park. Overall, taxa from 25 invertebrate orders were identified in the diets from 252 scat samples. Dietary components were similar for each species, but A. arktos consumed a higher frequency and volume of dipteran larvae and Diplopoda, while A. stuartii consumed more Coleoptera, Lepidoptera, Orthoptera and Isopoda. Both species of Antechinus had a higher percentage of ‘empty’ scats (devoid of any identifiable invertebrate material) in 2014 compared with 2015. The former was a drier year overall. Lower rainfall may have reduced abundance and diversity of arthropod prey, causing both species to supplement their diet with soft-bodied prey items such as earthworms, which are rarely detected in scats. Comparison of prey in scats with invertebrate captures from pitfall traps showed both species to be dietary generalists, despite exhibiting distinct preference and avoidance of certain prey categories. The ability of an endangered generalist marsupial to switch prey may be particularly advantageous considering the anticipated effects of climate change on Gondwanan rainforests during the mid-late 21st century.

Previous approaches to the description of complex diets, based on n-alkanes and optimization techniques, have grouped the plant species to reduce the number of components. Diet estimates have been obtained with least-squares routines by minimizing the discrepancy between faecal alkane concentrations calculated from herbage concentrations and actual faecal alkane concentrations. The effect of diet selection within groups can only be assessed by using sensitivity tests or by giving subjective weights to the individual plants. In the current study, a new optimization algorithm was developed that selects weightings that lead to consistent estimates of group proportions. The diet of the wild rabbit in a southern Portuguese montado was used as a case study. Estimates of the diet composition obtained using the new algorithm were compared with those of a conventional routine. The new algorithm was shown to provide, on average, more accurate estimates of the proportions of the groups in the diet. The effect of grouping plant species according to criteria other than similarity in n-alkane pattern on the accuracy of estimates was shown to be non-significant.
