Impact of shearing sheep on feeding and behaviour during the pre-embarkment feedlot phase of live export

Context Shearing is a management procedure that can induce stress, and is one that sheep often undergo on-farm and/or at registered premises during the pre-embarkation phase of live export. Aim As inanition is one of the largest causes of death during sea transport and can result from sheep experiencing stress, whether shearing results in behavioural changes and stress before boarding a vessel needs to be determined. Methods A total of 600 merino wethers sourced from one farm were housed at registered pre-embarkment premises, where they were split between two adjacent pens. The wethers were further split into the six treatment groups (unshorn, and sheep shorn on Days 1, 2, 3, 4 or 5) with 50 sheep per treatment, per pen. All wethers were fitted with radio frequency identification tags that were used to record animal presence at feed and water troughs, and coloured ear tags to visually identify their treatment group. Temperature and humidity sensors were placed on the outside of each pen to record environmental conditions throughout the 13-day study. Body condition scores for each wether were recorded at the beginning and end of the study. Four portable cameras were placed at a corner of each pen, allowing behaviour states (standing, lying or locomotion) of the wethers to be recorded on Days 1, 2, 3, 4, 5 and 13 of the study. Key results Shorn treatment groups were observed to spend more time standing (F5,5 = 6.63, P = 0.029) and less time lying (F23,23 = 2.48, P = 0.017). The control group spent more time at water troughs from Day 4 to 12 (F5,55 = 3.98, P < 0.01), whereas all groups were recorded as spending more time at the feed trough on Day 5 (F5,55 = 2.78, P < 0.001). Conclusions A lack of variation in behavioural states between the treatments and no change in time spent feeding throughout the study suggest that the procedure of shearing within the first 5 days at the pre-embarkation facility had no impact on behaviour of these sheep. Implications These results assist the industry in ensuring best management practices are implemented.

Abstract Experiential learning is a feature common to post-secondary agricultural education. Similar to experiential learning opportunities, incorporation of service learning into post-secondary coursework is being encouraged more often. Our objective was to determine how community engagement and introduction of real-world problems in a species-specific production course would affect student performance and outlooks on community service. This experiment was conducted as a completely randomized design within the beef cattle production class during the spring 2019 semester at Tarleton State University. Thirty-eight students were enrolled in the course across two laboratory sections. Students in the first laboratory section were used as a control group. In the control, students were asked to draw from a selection of possibilities a description of herd size, sire and dam breed type, and county. Groups were asked to develop a management plan for a commercial cow-calf operation. Students in the second laboratory section were asked to brainstorm the most prevalent barriers to implementation of best management practices in cow-calf production. Groups were randomly assigned to each topic and were tasked with developing a tri-fold brochure addressing the barrier. They were instructed that this was being done as a service to the greater beef community and would be evaluated by industry representatives. Students in CON had more (P < 0.05) students that reported performing community service weekly or monthly prior to the study than TRT. Groups were similar (P ≥ 0.41) in their assessment of community needs and the role of college students in community service. Likewise, CON and TRT had similar (P ≥ 0.41) responses in having less time available for coursework and their service contributing to the betterment of the overall community. Results are interpreted to mean that incorporation of service learning opportunities does not influence student views on community service.

Цель исследования - изучение перепрограммирования мононуклеарных лейкоцитов на модели системного ювенильного идиопатического артрита (сЮИА), воспроизводимой у крыс Wistar с использованием полного адъюванта Фрейнда и липополисахарида. Методика. сЮИА воспроизведен у 6-месячных крыс-самцов Wistar. На 40-е сут. эксперимента животные были разделены на 3 группы: 1-я группа - контроль; 2-я - группа доксициклина; 3-я - группа дексаметазона. Взятие проб крови у животных проводили на нулевые, 41-е и 55-е сут. Мононуклеарные клетки периферической крови выделяли гравиметрически, после чего окрашивали их на маркеры и внутриклеточные цитокины. Дифференцировали моноциты (CD3-CD4+) и Т-хелперы (CD3+CD4+). Анализировали динамику внутриклеточной экспрессии интерлейкина IL-4 (рассматривали как маркер про-М2 фенотипа, так как в случае выделения из клетки ИЛ-4 служит стимулятором М2 поляризации макрофагов) и IFN-g (как маркер про-М1 фенотипа) по данным проточной цитофлуориметрии. Применяли непараметрический статистический тест Mann-Whitney-Wilcoxon в программе R для статистической обработки данных. Результаты и заключение. При моделировании сЮИА выявлено закономерное изменение фенотипа моноцитов. Применение же доксициклина и дексаметазона приводило к более ранней поляризации их по про-М2-пути в отношении моноцитов (на 41-е сут.) в сравнении с контролем. Про-М1 эффект (на 55-е сут., в сравнении с контролем) выявлен также в группах доксициклина и дексаметазона. У животных разных групп обнаружены характерные динамические изменения внутриклеточной экспрессии цитокинов. Важно, что различная направленность поляризации фенотипа при сЮИА и применении препаратов наблюдается не только у моноцитов, но и у Т-хелперов. The study objective was to evaluate targeted reprogramming of peripheral blood mononuclear cells in systemic juvenile idiopathic arthritis (sJIA) modeled in 6-month-old male Wistar rats by co-administration of complete Freund’s adjuvant and lipopolysaccharide. Methods. On day 40 of the experiment, rats were divided into three groups: control, doxycycline, and dexamethasone groups. Blood samples were collected on days 0, 41, and 55. Peripheral blood mononuclear cells were isolated gravimetrically and stained for markers and cytokines. Monocytes (CD3-CD4+) and T-helpers (CD3+CD4+) were differentiated as target cells. IL-4 was considered a marker for the pro-M2 phenotype since IL-4 can activate M2 macrophage polarization; IFN-g was considered a marker for the pro-M1 phenotype. Time-related changes in the intracellular expression of IL-4 and IFN-g were studied using flow cytometry. Data were analyzed using nonparametric statistical tests (Mann-Whitney-Wilcoxon) in the R environment for statistical computing. Results and conclusions. Monocytes (like macrophages) underwent reprogramming during the development of modeled sJIA disease. In monocytes of doxycycline and dexamethasone treatment groups, pro-M2 effects were observed earlier (day 41) than in the control group. Pro-M1 effects were observed in monocytes of doxycycline and dexamethasone groups on day 55, as compared with the control group. Characteristic time-related changes of intracellular cytokine expression were described for different groups. Importantly, the differently directed phenotype polarization was observed in sJIA and treatment groups for both monocytes and T-helpers.
