Dose-response effect of nitrate on hydrogen distribution between rumen fermentation end products: an in vitro approach

The objective of this work was to study the in vitro dose-response effect of nitrate (0, 1, 2, 4 and 6 mM) on metabolic hydrogen distribution between rumen fermentation end products. Three 48-h incubations were conducted using bovine rumen contents as an inoculum, and a mixture of hay and concentrate (50 : 50) as a substrate. Total gas production and composition (methane and hydrogen) were automatically analysed throughout the incubations. Volatile fatty acid and ammonium concentrations were analysed from samples taken after 48 h of incubation. Total gas production was decreased with the highest dose of nitrate (P = 0.002). Methane emissions linearly decreased as the nitrate dose increased (P = 0.005). Kinetics of methane emissions showed that metabolic hydrogen removal via nitrate reduction occurred mainly during the first 10 h of incubation. Gaseous hydrogen production was similar among treatments, despite higher hydrogen emissions for nitrate concentrations >4 mM. Concentrations and proportions of volatile fatty acids were not affected by treatments. The proportion of unaccounted metabolic hydrogen was positive for all treatments, and tended to linearly increase as the nitrate dose increased. In this in vitro work, we confirmed that nitrate is an efficient methane-mitigating compound in the rumen. We also suggest that nitrate or its reduced forms have a direct inhibiting effect towards methanogens, as indicated by the release of gaseous hydrogen and the high efficiency of methane mitigation. However, high nitrate concentrations also decrease overall fermentation.

Alternative feed sources can be utilized to reduce enteric methane (CH4) emissions, a major greenhouse gas that contributes to global warming. This study aimed to evaluate the potential use of tropical plants to improve digestibility, reduce protozoal populations, improve rumen fermentation, and minimize methane emissions from ruminants. The plants considered herein grow in tropical climates, are easily accessible in large quantities, and are directly related to human food production. Nine plants that grow naturally in tropical climates were assessed. Plant supplementation substantially enhanced accumulative gas production at 24 h (p < 0.05). The apparent organic matter digestibility (AOMDvt) of the diet was not affected by five of the nine plants. With the addition of the plant material, ammonia nitrogen concentrations were reduced by up to 47% and methane concentrations were reduced by 54%. Five of the nine plant materials reduced methane production in terms of CH4/dry matter and CH4/digestibility of the organic matter by 15–35% and 8–24%, respectively. In conclusion, supplementation with plants with high tannin contents was shown to be a viable strategy for improving rumen fermentation, reducing protozoal populations, and limiting methane emissions. In this regard, the leaves of Piper sarmentosum, Acmella oleracea, Careya arborea, and Anacardium occidentale were especially promising.

Actinobacteria have been researched as a source that produces crude extracts, which contain bioactive compounds able to act as antimicrobial agents. The present investigation evaluated the dose-response effect of two crude extracts, obtained at Caatinga rhizosphere (Caat) and Rhizophora mangle (AMC), on in vitro ruminal fermentation by: cumulative gas production, digestibility of dry (IVDMD) and organic matter (IVOMD), and short-chain fatty acids concentration (SCFA). Three multiparous Holstein dairy cows with ruminal fistula were used as the inoculum donors and fed a basal diet consisting of corn silage, soybean meal, urea, ground corn and mineral supplement. Ruminal fluid samples were incubated in glass bottles with 1 g of the dried and milled diet, a buffer solution, and the crude extracts evaluated in four doses (0.3, 0.6, 0.9 and 1.20 mg/10 mL inoculum) in a randomized block design, and the donators were considered as blocks with random effects. Additionally, negative controls were used. The results were expressed as average values based on triplicate analyses. Decreased cumulative gas production was observed according to linear dose response at 24, 48 and 72 h of incubation with the addition of Caat extract. The IVOMD showed a linear decrease at 72 h of incubation with dose Caat inclusion. Furthermore, the inclusion of Caat extract linearly reduced butyric and isovaleric acid concentrations, as well as acetate:propionate ratio. Finally, the Caat inclusion increased the propionic acid concentration in comparison to AMC extract. However, the inclusion of AMC extract did not affect any of the analyzed variables at the used doses. The Caat extract could be used as a modulator of in vitro ruminal fermentation, since it reduced acetate:propionate ratio and cumulative gas production.

SummaryThe three most important factors describing the nutritive value of ruminant feeds are: feed intake, rumen fermentation, and enzymatic digestion. Animal experiments are usually expensive and time-consuming, so many in vitro techniques have been developed which are supposed to assist in the prediction of nutritive value, usually of rumen fermentation. One of these is the measurement of gas production from the substrate when incubated with a specific inoculum. The cumulative gas production technique measures gas at regular intervals in time, as an indicator of the kinetics of fermentation. The resultant cumulative gas profiles, fitted to specific equations, and in combination with other end-products (i.e. VFA and NH3) can indicate both the rate and extent of fermentability of the test substrates. This information can be used to compare substrates with each other. However, the fact that the technique has not yet been standardized, and that inoculum varies according to time and place, means that comparison both between runs and locations, can be very difficult. Also, there are a number of aspects of the technique which will always mean that its resemblance to the in vivo situation must be limited. Some of these limitations and other important considerations of the technique are described in this review.

Nutritional strategies can be employed to mitigate greenhouse emissions from ruminants. This article investigates the effects of polyphenols extracted from the involucres of Castanea mollissima Blume (PICB) on in vitro rumen fermentation. Three healthy Angus bulls (350 ± 50 kg), with permanent rumen fistula, were used as the donors of rumen fluids. A basic diet was supplemented with five doses of PICB (0%–0.5% dry matter (DM)), replicated thrice for each dose. Volatile fatty acids (VFAs), ammonia nitrogen concentration (NH3-N), and methane (CH4) yield were measured after 24 h of in vitro fermentation, and gas production was monitored for 96 h. The trial was carried out over three runs. The results showed that the addition of PICB significantly reduced NH3-N (p < 0.05) compared to control. The 0.1%–0.4% PICB significantly decreased acetic acid content (p < 0.05). Addition of 0.2% and 0.3% PICB significantly increased the propionic acid content (p < 0.05) and reduced the acetic acid/propionic acid ratio, CH4 content, and yield (p < 0.05). A highly significant quadratic response was shown, with increasing PICB levels for all the parameters abovementioned (p < 0.01). The increases in PICB concentration resulted in a highly significant linear and quadratic response by 96-h dynamic fermentation parameters (p < 0.01). Our results indicate that 0.2% PICB had the best effect on in-vitro rumen fermentation efficiency and reduced greenhouse gas production.

Methane (CH4) from ruminal feed degradation is a major pollutant from ruminant livestock, which calls for mitigation strategies. The purpose of the present 4 × 2 factorial arrangement was to investigate the dose–response relationships between four doses of the CH4 inhibitor 3-nitrooxypropanol (3-NOP) and potential synergistic effects with low (LC) or high (HC) concentrate feed proportions (CFP) on CH4 reduction as both mitigation approaches differ in their mode of action (direct 3-NOP vs. indirect CFP effects). Diet substrates and 3-NOP were incubated in a rumen simulation technique to measure the concentration and production of volatile fatty acids (VFA), fermentation gases as well as substrate disappearance. Negative side effects on fermentation regarding total VFA and gas production as well as nutrient degradability were observed for neither CFP nor 3-NOP. CH4 production decreased from 10% up to 97% in a dose-dependent manner with increasing 3-NOP inclusion rate (dose: p < 0.001) but irrespective of CFP (CFP × dose: p = 0.094). Hydrogen gas accumulated correspondingly with increased 3-NOP dose (dose: p < 0.001). In vitro pH (p = 0.019) and redox potential (p = 0.066) varied by CFP, whereas the latter fluctuated with 3-NOP dose (p = 0.01). Acetate and iso-butyrate (mol %) decreased with 3-NOP dose, whereas iso-valerate increased (dose: p < 0.001). Propionate and valerate varied inconsistently due to 3-NOP supplementation. The feed additive 3-NOP was proven to be a dose-dependent yet effective CH4 inhibitor under conditions in vitro. The observed lack of additivity of increased CFP on the CH4 inhibition potential of 3-NOP needs to be verified in future research testing further diet types both in vitro and in vivo.

Abstract Vistore® minerals are hydroxychloride minerals that feature high metal content and improved bioavailability. This study was conducted to compare different sources of zinc (Zn) on in vitro rumen fermentation parameters. Three ruminally-cannulated Jersey heifers were adapted to a lactation diet for two weeks before used as donors. Three sources of Zn were tested at 20 ppm: No supplemental Zn (CON), ZnSO4, Vistore Zn, and another Zn hydroxychloride (Vistore-competitor). The concentration of Zn in this study was selected from a titration study (0 to 40 ppm ZnSO4) to identify the minimum concentration of ZnSo4 affecting rumen fermentation. The lactation diet (TMR) was dried and ground to 1mm and used as substrate. Rumen fluid was collected two hours after feeding. Substrate (0.5 g) was inoculated with 100 mL of 3:1 McDougall’s buffer: ruminal flued mixture at 39ºC for 24 h. Each treatment was run in triplicate and in three runs. Data were analyzed with R 3.0. The model included fixed effect of treatment and random effect of run. ZnSO4 reduced (P &lt; 0.05) maximum gas production, DMD (54 vs. 55.9%) and cellulose (27.5 and 40.7%) digestibility. acetate to propionate ration (2.20 vs. 2.24) and NH3-N concentration (6.0 vs. 7.0 mg/dL), increased (P &lt; 0.05) propionate % (27.2 vs 26.7%) compared to control. Vistore had higher pH than control (6.44 vs. 6.40, P = 0.02) but did not affect other parameters compared to CON. Vistore-competitor reduced total VFA production compared to control, ZnSO4, and Vistore (94 vs. 102, 106 and 107 mM, respectively, P = 0.01) but did not affect other parameters. In general, Vistore Zn maintained in vitro ruminal fermentation and digestibility, while ZnSO4 had negative effects on both fermentation and digestibility and Vistore-competitor reduced total VFAs. Results indicate hydroxychloride minerals may stabilize rumen parameters versus sulfate sources but different hydroxychloride sources appear to influence rumen parameters differently.

Abstract The aim of the research was to evaluate the effect of three Eucalyptus globulus extracts rich in phenolic compounds, especially flavonoids, on rumen fermentation, methane (CH4) production, organic matter degradability and protozoa population using an in vitro gas production technique. Four concentrations (0, 50, 75 and 100 mg) of three Eucalyptus extracts (ethyl acetate, n-butanol and aqueous) were added to a diet of ruminants (forage: concentrate ratio 60:40) and incubated at 39°C under anaerobiosis with buffered rumen fluid. After 24 h, the fermentation fluid was analysed for ammonia-N and volatile fatty acids (VFA). Organic matter degradability (OMD) and protozoa were also determined; in vitro gas production was also recorded and CH4 concentration was measured. Compared to the control, CH4 production was significantly lower for ethyl acetate extract (P<0.05), but higher for n-butanol and aqueous extracts. Production of ammonia- N was lower in all Eucalyptus extracts (P<0.05). Propionate production (P<0.05) increased for ethyl acetate and n-butanol extracts, whereas no effect was registered for VFA, for all Eucalyptus extracts. Ethyl acetate extract decreased in vitro OMD (P<0.05), whereas n-butanol and aqueous extracts were comparable to the control. Protozoa population decreased (P<0.05) for all extracts in comparison with the control. Eucalyptus ethyl acetate extract might be promising to be used as a potent anti-methanogenic additive. Moreover, the assessment of the right dosage seems to be important to decrease methane production, without reducing feed nutritional value.

Abstract Background A need for research searching for alternative rumen enhancers warrants immediate attention. The in vitro fermentation experiment was conducted using factorial arrangement of two factors of roughage to concentrate and seven level of red amaranth leaf powder percentage of total substrate in a Completely randomized design (CRD). Two factors, namely Factor A was two ratio of roughage (R) to concentrate (C) at 60:40 and 40:60 and Factor B was level of red amaranth (Amaranthus cruentus, L) leaf powder (RALP) supplementation at 0, 2, 4, 6, 8, 10, and 12% of total dietary substrate. Results Red amaranth leaf powder (RALP) contained phytonutrients both condensed tannins and saponins in addition with high macro minerals (Ca, K, and Mg). This experiment revealed innovations of the RALP supplementation by enhancing rumen propionate (C3) production, reducing acetate (C2) to (C3) ratio, reducing protozoal population and mitigating methane (CH4) production. Furthermore, rumen dry matter degradation percentages were remarkably enhanced (P < 0.001) by increasing RALP supplementation. Conclusion Plants rich in phytonutrients and minerals such as red amaranth leaf powder (RALP) have a vital and promising role in modulating rumen fermentation, mitigating methane production, as well as increasing substrate degradability.
