Impact of simulated heat stress on growth, physiological adaptability, blood metabolites and endocrine responses in Malpura ewes under semiarid tropical environment

The study was conducted to develop a simulated heat stress model for sheep depicting natural environmental conditions and to observe its impact on growth, reproductive performance, physiological adaptability, blood metabolites and endocrine responses in Malpura ewes. The experiment was conducted on 16 adult Malpura ewes for a period of 35 days. The animals were randomly divided into two groups of eight animals each (GI – control, GII – heat stress). The GII ewes were exposed to different temperature at different hours of the day i.e. 38°C at 1000–1100 hours; 40°C at 1100–1200 hours; 42°C at 1200–1300 hours; 43°C at 1300–1400 hours; 44°C at 1400–1500 hours and 42°C at 1500–1600 hours in a climatic chamber. Feed intake (P < 0.01), bodyweight (P < 0.05) and body condition score (P < 0.01) reduced significantly in GII as compared with GI ewes while water intake increased significantly (P < 0.01). All physiological responses also showed significant variation between the groups for treatment. In addition, all endocrine parameters showed highly significant (P < 0.01) variation for the treatment. Plasma cortisol increased significantly (P < 0.01) while T3 (P < 0.01) and T4 (P < 0.01) decreased significantly in GII as compared with GI ewes. Further, plasma oestradiol decreased significantly (P < 0.01) while plasma progesterone increased significantly (P < 0.01) in GII as compared with GI. Among the biochemical parameters, hemoglobin, packed cell volume, plasma glucose, total cholesterol, urea and blood urea nitrogen differed significantly between the groups. In addition, oestrus duration also significantly (P < 0.05) reduced in GII as compared with GI. Based on the results obtained from the study it can be concluded that simulated heat stress influenced the adaptive behaviour of sheep in terms of changes in physiological, blood biochemical and endocrine responses. Ultimately, their growth and reproductive performance are compromised during the process of adaptation to the simulated heat stress.

The aim of the current study was to investigate the relationship between prepartum body condition score (BCS), blood metabolites (Glucose, beta-hydroxybutyrate; BHB), oxidative stress (Malondialdehyde; MDA, Glutathione peroxidase; GSH-Px) in transition period and some reproductive parameters in Holstein dairy cows. Fifty cows were divided into two groups [low-BCS = 2.75-3.0 (n=25); adequate-BCS = 3.25-3.75 (n=25)] according to BCS on the 21st day before expected calving. BCS was also recorded at 0 and +21 days after calving. Blood samples collected at -21, 0 and +21 days from calving for MDA and GSH-Px analysis as well as at -21, +14 and +21 days of postpartum for glucose and BHB. Data on reproductive parameters obtained from farm records were also collected. MDA concentrations were higher in group of low-BCS compared to group of adequate-BCS at -21 and 0 d related to calving (p<0.05). GSH-Px activity was lower in group of low-BCS than adequate one during the transition period (p<0.05). Low-BCS group also showed higher concentrations of BHB at +14 and +21 days after calving. During the transition period, low-BCS group had lower glucose concentrations compared to adequate one (p<0.05). Adequate-BCS group of cows showed shorter intervals of calving to first estrus (p<0.01) and calving to conception (p<0.01). In conclusion, cows with lower BCS at the prepartum period had worse metabolic and oxidative balance during the transition period. This situation also was associated with worse reproductive performance in cows.

The aim of the present work was to examine the effects of anionic salts (MgCl2 or MgSO4) supplementation on metabolic profile, peripartum diseases and pregnancy rate of dairy cows during the prepartum period. Fourty five Holstein dairy cows were placed on 1 of 3 prepartum treatments. Treatment groups were balanced body condition score as far as similar. Treatments were total mixed ration (TMR) without anionic salt (control), TMR supplemented with 30 g/d/cow MgCl2 or TMR supplemented with 30 g/d/cow MgSO4. After parturation, all groups were fed with same TMR. The presynch-ovsynch protocol was used and fixed time artificial insemination was done after 30 days postpartum. The experiment was lasted for 8 weeks of postpartum. Cows fed MgCl2 supplemented TMR increased milk yield compared with control and MgSO4 supplemented TMR. Supplementing anionic salts reduced serum AST and ALT concentrations, increased serum glucose, HDL, IgG, Ca and Mg concentrations. Health and reproductive performance were not different between treatments. These data suggets that anionic salts supplementation in TMR during prepartum can alter milk yield and blood biochemical parameters.

Abstract This study evaluated the effect of heifer development system on body weight (BW), body condition score (BCS), fescue toxicosis symptoms, reproductive performance, and subsequent calf growth of fall-calving beef heifers. Angus × Simmental heifers [n = 399; 240 ± 20.0 kg initial BW; age = 252 ± 20 d] were stratified by BW and BCS and assigned to 1 of 12 groups in each of the 2 production years. The study utilized a stratified randomized design. Pens were randomly assigned to 4 treatments: drylot (DL) development (fed ad-libitum diet consisting of 90% hay and 10% DDGS on a dry matter basis), grazing endophyte-infected fescue supplemented daily (2.3 kg as-fed/heifer/d; 50:50 mix of soybean hulls and DDGS; E+/S), grazing endophyte-infected fescue and supplemented from the midpoint of treatment period until breeding (4.5 kg as-fed/heifer/d; 50:50 mix of soybean hulls and DDGS; E+/LS), and grazing novel endophyte-infected fescue with no supplement (NE+/NS). Treatments ceased on d 168 [time of artificial insemination (AI)] and heifers were commingled and managed as a group through second breeding season. Heifers in DL had greatest (P ≤ 0.05) BW and BCS from d 28 until d 254. Furthermore, E+/S heifers had greater (P ≤ 0.05) BW and BCS than both E+/LS and NE+/NS from d 28 until d 168. On d 56 and 84, E+/LS heifers had lower (P ≤ 0.05) BW and BCS compared to NE+/NS, but on d 148 treatments reranked and E+/LS remained at a greater (P ≤ 0.05) BW and BCS compared to NE+/NS through the first breeding season. Drylot heifers had greatest (P ≤ 0.05) percentage cycling and percentage of mature BW at AI (66.6%) and had greater (P ≤ 0.05) AI and overall pregnancy rates compared to E+/LS and NE+/NS. The E+/S (55%) and E+/LS (53.7%) heifers were developed to a greater (P &lt; 0.01) percentage of mature BW than NE+/NS (49.3%). A greater (P ≤ 0.02) percentage of DL and E+/S heifers were pregnant at the end of the first breeding season (89.3 and 85.1%; respectively) compared to NE+/NS (61.5%). In summary, DL heifers had the greatest BW and BCS at AI, percentage cycling, and AI pregnancy rate. However, this strategy did not result in differing overall pregnancy rates between DL, E+/S, and E+/LS and there were no differences in milk production, rebreeding reproductive performance, and calf performance between all treatments. Finally, the poorest AI and overall pregnancy rates of the NE+/NS heifers suggests this is not a viable development strategy for fall-born heifers.

A Inseminação artificial em tempo fixo (IATF) está se difundindo no Brasil, pois contribui para o aumento da produtividade. O objetivo neste trabalho foi avaliar o uso do GnRH em protocolos de IATF de vacas criadas no pantanal Sul-mato-grossense. Participaram da pesquisa 531 vacas paridas da raça Nelore, que foram avaliadas de acordo com o escore de condição corporal (ECC), submetidas a três tipos de protocolos de IATF (lote 1, 2 e 3). Após 40 dias da inseminação artificial foi realizado diagnóstico de gestação. As matrizes que não receberam GnRH no lote 1 (estro aparente), lote 2 (estro parcial) e lote 3 (sem apresentação de estro) apresentaram, respectivamente 55,4%, 19,2% e 25,9% de prenhez, enquanto as que receberam o fármaco apresentaram 48,6%, 37,2% e 38,8%. Relacionado ao ECC, pode-se observar que a maior taxa de prenhez ocorreu em animais com ECC 3. Primíparas apresentaram melhor taxa de prenhez quando receberam GnRH independente do ECC. Concluiu-se que o uso de GnRH mostrou-se eficiente para melhoria da taxa de prenhez principalmente em primíparas, assim como nos animais que não apresentaram estro ou apresentaram estro parcial, e que matrizes com ECC 3 apresentaram melhores taxas de prenhez que vacas com escore inferior. Palavras-chave: bovino de corte; inseminação artificial em tempo fixo; produtividade; GnRH; Nelore.   Evaluation of cow reproductive performance in the region of Mato Grosso do Sul swamp submitted to TAI with GnRH application   ABSTRACT: The technique of timed artificial insemination (TAI) is spreading in Brazil, as it contributes to the increase of productivity. The objective of this work was to evaluate the use of GnRH in TAI protocols of cows raised in the swamp of Mato Grosso do Sul. 531 Nelore breed calved cows participate in the survey, which were evaluated according to the body condition score (BCS), and then performed three types of TAI protocols (batch 1, 2 and 3). After 40 days of the insemination, a pregnancy diagnosis was made. The matrices that did not receive GnRH in batch 1 (apparent oestrus), batch 2 (partial oestrus) and batch 3 (no oestrus presentation) presented respectively 55.4%, 19.2% and 25.9% of pregnancy, while those receiving the drug showed 48.6%, 37.2% and 38.8%. Related to BCS, it can be observed that the highest pregnancy rate occurred in animals with BCS 3. Primiparous had a better pregnancy rate when receiving GnRH regardless of BCS. It was concluded that the use of GnRH was efficient to improve the pregnancy rate mainly in primiparous as well as animals that showed no oestrus or partial oestrus, and matrices with BCS 3 had better pregnancy rates than cows with lower scores. ABSTRACTData from a large controlled experiment to investigate the effect of level of nutrition on reproductive performance were used to assess the value of production and blood measures as indicators of energy status in lactating dairy heifers. Live-weight change showed the strongest and most consistent relationship to mean energy balance (the difference between metabolizable energy intake and that used for milk production and maintenance) (P < 0·01 to P < 0·001). Body-condition score at a particular time was more closely related to mean energy balance in the preceding 4-week period (P < 0·05) than current energy balance. There was also a lag in the relationship between energy balance and live-weight change and mean body-condition score. Ultrasonic back fat measurements were significantly correlated with both loin and tailhead body-condition score (P < 0·001) but were more strongly related to mean energy balance in the preceding period (P < 0·05 to P < 0·001) than the body-condition scores.Blood samples were taken from all heifers 2 weeks before calving and 1, 5, 9, 13 and 18 weeks after calving and were analysed for 13 constituents. Concentrations of blood metabolites did not show consistently strong correlations with mean energy balance. The only blood metabolite to be measurably affected by the nutritional treatments applied in lactation was β-hydroxybutyrate.At best, combinations of production measures and blood metabolites were only able to predict the mean daily energy balance with a 95% confidence interval of ±20 MJ for an individual animal although this confidence interval was reduced to ±3 MJ for 100 animals.

ABSTRACTThe effects of high-low (HL) and low-high (LH) patterns of nutrition during the 5 weeks prior to a synchronized mating were studied on the reproductive performance of 115 Welsh Mountain and 114 Brecknock Cheviot ewes in a range of body condition at the start of treatment. For 2 months prior to treatment imposition, ewes were grazed in a systematic way at different stocking rates on different sward heights to obtain a range of body condition scores. Target score groupings were 2·00 to 2·25, 2·50 to 2·75 and 3·00 to 3·25 and although scores of 1·50 to 3·25 were obtained, most lay in the 2·00 to 2·75 range. Two treatment groups, balanced for live weight and body condition score, received high and low levels of nutrition for 16 days from mid October, achieved, respectively, by a low stocking rate on a sward with a high surface height plus ad libitum concentrate and a high stocking rate on a sward with a low surface height plus 200 g hay per head per day. Treatments were then reversed for the 17 days prior to mating. Live-weight and body condition-score changes were recorded and reproductive performance at first mating was measured from counts of corpora lutea and viable embryos at slaughter either at return to service or at 4 weeks after first mating. There were no differences due to nutritional pattern in live weight or body condition score at mating. The LH treatment significantly increased the rates of ovulation, conception, multiple ova survival and potential lambing per ewe pregnant and per ewe mated in the Welsh Mountain breed but significantly increased only the rates of ovulation and potential lambing per ewe pregnant in the Brecknock Cheviot breed compared with the HL treatment. The relatively poorer response in the latter breed was partially due to a lower ovulation rate potential coupled with non-significantly lower rates of conception and survival of single-shed ova associated with the LH treatment. Much of this relatively poorer reproductive performance in the Brecknock Cheviot breed was in ewes with body condition scores 3= 2·75. Reproductive performance increased with increasing body condition at the start of treatment over the range of scores =≤ 2·25 to 2·50 in both breeds and, while continuing to improve in scores above 2·50 in the Welsh Mountain breed, in the Brecknock Cheviot breed it started to decline. Ewes with a body condition score of 2·50 in both breeds showed the greatest response in potential lambing rate to the LH treatment.

The aim of this study was to compare 3 methods for synchronization of ovulation in anestrous beef cows. The hypothesis of this study was to determine whether low doses of hCG has superior efficacy to cypionate to induce ovulation in anestrous cows and provide higher pregnancy rate in oestrus-synchronization programs. Synchronization of ovulation and conception rate to timed AI (TAI) were evaluated in anestrus Bos taurus taurus suckling beef cows 45 ± 15 days postpartum and with body condition score of 2.9 (1 to 5) maintained in a native pastured system in the south of Brazil. Females were evaluated with ultrasound on the Day 0 (D0) of the protocol (Day 0), day 8 (D8), immediately before TAI (D10), and 7 days after TAI (Day 17). All cows were synchronized with an intravaginal progesterone-releasing device (IPRD; 0.75 g of progesterone, Prociclar®, Hertape Calier Animal Health, Juatuba, Brazil) and 2 mg IM of oestradiol benzoate (EB; Benzoato HC®) on D0. On Day 8, the IPRD was removed and 150 μg of D (+) cloprostenol (Veteglan Luteolytic®), and 25 IU IM FSH/LH (Pluset®) were administered. Females of the EC (n = 84) group received 1 mg IM of oestradiol cypionate (EC; Cipionato HC®). Females on D8 of the hCG (n = 81) group received 500 IU IM of hCG (Vetecor®, Hertape Calier) at the time of TAI. The females of the EC + hCG group (n = 83) received both treatments. All cows were submitted to TAI 54 h after withdrawal of IPRD. A part of the cows (n = 102) had the ovulation evaluated every 12 h from the withdrawal of IPRD [EC (n = 34), hCG (n = 34), and hCG + EC (n = 33)]. Statistical analysis was performed using SAS PROC GLIMMIX. The dominant follicle diameter (FD) on Day 8 (8.7 ± 0.2, 8.8 ± 0.2, 8.6 ± 0.2) did not differ between treatments EC, EC + hCG, or hCG (P = 0.79). However, the FD on D10 was higher (P = 0.001) for cows treated with hCG (12.9 ± 0.3) compared with cows from the EC (11.3 ± 0.2) or EC + hCG group (11.8 ± 0.2). The interval (h) between the withdrawal of IPRD and ovulation was lower (P = 0.01) for the hCG group, (71.2 ± 1.7) compared with the groups treated with EC or EC + hCG (76.6 ± 2.18 and 74.2 ± 1.65), respectively. The ovulation rate did not differ (P = 0.61) among the EC (85.2%, 29/34), hCG (91.1%, 31/34), or EC + hCG groups (90.9%, 30/33). Corpus luteum diameter (mm) was higher (P = 0.04) on D17 for the hCG-treated group (21.4 ± 0.3) compared with others treatments (EC = 19.1 ± 0.8 or EC + hCG = 20.4 ± 0.8). However, the plasma progesterone levels on D17 were EC = 2.0 ± 0.1, hCG = 2.4 ± 0.1, and EC + hCG = 2.3 ± 0.1 ng mL–1 (P = 0.19), and the conception rate on the 28th day after TAI (EC = 43.0%; hCG = 47.0%, and EC + hCG = 48.8%; P = 0.76) was also similar. The hCG determined smallest ovulation interval, but similar rates of pregnancy were observed with both treatments.
