Effect of conception date and hind nutrition on fetal growth trajectory and gestation length of red deer (Cervus elaphus)

This study tested the hypothesis that the negative association between gestation length and conception date in red deer is mediated by nutrition. Twenty-eight pregnant red deer were randomly allocated to four groups according to a 2 × 2 factorial design, with the factors conception date (14 March, E; 28 April, L) and level of nutrition (ad libitum, H; restricted, R). Animals were housed indoors in individual pens from early winter until calving and offered daily an ad libitum pelleted ration. The daily ration was then restricted from late winter in ER (134 days post-conception) and LR (89 days post-conception) groups, so that these hinds did not experience a seasonal increase in food intake. X-Ray computed tomography scans were taken at Days 120, 150, 180 and 210 of gestation (mid–late gestation) to estimate weight of various conceptus components. Growth rate of the total fetus was significantly higher in LH than in other treatments (P < 0.01) between Days 180 and 210 of gestation. Birthweight was not significantly different (P > 0.05) between treatments or calf sex. Birthweight was associated directly with change in hind liveweight (P = 0.03) and body condition score during the third trimester of pregnancy (P = 0.01), but was not significantly associated with gestation length (P = 0.34). Gestation length was 4.4 days longer in LR than LH hinds (P = 0.03) and was negatively associated with both food intake (P = 0.03) and LW gain (P = 0.02) during the final trimester of gestation. Feeding late-conceiving hinds an ad libitum diet of high-quality food during the third trimester of gestation maximises fetal growth and shortens gestation length.

In order to determine whether pregnancy outcome was altered when women were employed outside their homes, 7,722 pregnancies were analyzed. Gestations were not shortened but newborns of women who worked in the third trimester weighed 150 to 400 gm less than newborns of mothers who remained at home. The growth retardation was greatest when women were underweight pregravid and had a low pregnancy weight gain, when they were hypertensive, or when the work required standing. The growth retardation remained after the data were stratified by race, socioeconomic status, and other maternal factors that commonly influence fetal growth. The frequency of large placental infarcts progressively increased when women continued stand-up work into late gestation. Such infarcts reached a peak of 250/1,000 births after the 37th week of gestation in stand-up workers. Low uteroplacental blood flow is a likely explanation for both the fetal growth retardation and the large placental infarcts.

Abstract Creatine is a metabolite involved in cellular energy homeostasis. In this study, we examined placental creatine content, and expression of the enzymes required for creatine synthesis, transport and the creatine kinase reaction, in pregnancies complicated by low birthweight. We studied first trimester chorionic villus biopsies (CVBs) of small for gestational age (SGA) and appropriately grown infants (AGA), along with third trimester placental samples from fetal growth restricted (FGR) and healthy gestation-matched controls. Placental creatine and creatine precursor (guanidinoacetate—GAA) levels were measured. Maternal and cord serum from control and FGR pregnancies were also analyzed for creatine concentration. mRNA expression of the creatine transporter (SLC6A8); synthesizing enzymes arginine:glycine aminotransferase (GATM) and guanidinoacetate methyltransferase (GAMT); mitochondrial (mtCK) and cytosolic (BBCK) creatine kinases; and amino acid transporters (SLC7A1 & SLC7A2) was assessed in both CVBs and placental samples. Protein levels of AGAT (arginine:glycine aminotransferase), GAMT, mtCK and BBCK were also measured in placental samples. Key findings; total creatine content of the third trimester FGR placentae was 43% higher than controls. The increased creatine content of placental tissue was not reflected in maternal or fetal serum from FGR pregnancies. Tissue concentrations of GAA were lower in the third trimester FGR placentae compared to controls, with lower GATM and GAMT mRNA expression also observed. No differences in the mRNA expression of GATM, GAMT or SLC6A8 were observed between CVBs from SGA and AGA pregnancies. These results suggest placental creatine metabolism in FGR pregnancies is altered in late gestation. The relevance of these changes on placental bioenergetics should be the focus of future investigations.

In a study of 467 gestations maternal stature had little correlation with fetal growth but mother's pregravid body weight as well as weight gain and low-calorie diets during pregnancy did have such a correlation. Maternal undernutrition before the third trimester had little or no influence on fetal body, organ, and cellular growth while such effects were pronounced in late gestation. Fetal body and organ growth improved with successive pregnancies except in the most poorly nourished mothers, whose successive neonates became more growth retarded.

To define the role of endothelial nitric oxide (NO) in developmental changes in pulmonary vascular resistance and oxygen responsiveness, we determined the ontogeny of endothelial NO production and of oxygen modulation of that process in pulmonary arteries from fetal and newborn lambs. NO production was assessed by measuring endothelium-dependent arterial guanosine 3',5'-cyclic monophosphate synthesis. Basal NO rose two-fold from late gestation to 1 wk of age and another 1.6-fold from 1 to 4 wk. Acetylcholine-stimulated NO also increased 1.6-fold from 1 to 4 wk. The maturational rise in NO was evident at high Po2 in vitro, and it was not modified by L-arginine. This suggests that the developmental increase may alternatively involve enhanced calcium-calmodulin-mediated mechanisms, increased expression of NO synthase, or greater availability of required cofactor(s). With an acute decline in Po2 in vitro from 680 to 150 or 40 mmHg, there was 50-88% attenuation of basal and acetylcholine-stimulated NO late in the third trimester and in the newborn but not early in the third trimester. Parallel studies of mesenteric endothelium revealed postnatal increases in basal and stimulated NO but no decline in NO at lower Po2. Ontogenic changes in endothelial NO production and in oxygen modulation of that process may be involved in the maturational decrease in vascular resistance and the development of oxygen responsiveness in the pulmonary circulation.

Context: Thyroid function is known to play an important role in fetal neurological development, but its role in regulating fetal growth is not well established. Overt maternal and fetal thyroid disorders are associated with reduced birth weight. We hypothesized that, even in the absence of overt thyroid dysfunction, maternal and fetal thyroid function influence fetal growth. Aim: In normal, healthy pregnancies, we aimed to assess whether fetal thyroid hormone at birth (as measured in cord blood) is associated with fetal growth. We also aimed to study whether fetal thyroid hormone at birth is associated with maternal thyroid hormone in the third trimester. Methods: In 616 healthy mother-child pairs, TSH, free T4 (FT4), and free T3 (FT3) were measured in mothers at 28 wk gestation and in umbilical cord blood at birth. Birth weight, length, head circumference, and tricep and bicep skinfold thicknesses were measured on the babies. Results: Cord FT4 was associated with birth weight (r = 0.25; P &lt; 0.001), length (r = 0.17; P &lt; 0.001), and sum of skinfolds (r = 0.19; P &lt; 0.001). There were no associations between birth measurements and either cord TSH or cord FT3. Maternal FT4 and cord FT4 were correlated (r = 0.14; P = 0.0004), and there were weaker negative associations between maternal TSH and cord FT4 (r = −0.08; P = 0.04) and FT3 (r = −0.10; P = 0.01). Conclusion: Associations between cord FT4 and birth size suggest that fetal thyroid function may be important in regulating fetal growth, both of skeletal size and fat. The correlation between third-trimester maternal FT4 and cord FT4 supports the belief that maternal T4 crosses the placenta even in late gestation.

SummaryMilk yield of 2-year-old Scottish Blackface and of East Friesland × Scottish Blackface ewes rearing single or twin lambs was estimated by the oxytocin method on 1 day each week up to the 14th week of lactation. Both groups of ewes had been reared from birth under good nutritional conditions. They were housed in individual pens from mid-pregnancy and offered a liberal ration of a pelleted concentrate food until parturition. Similar food was offered ad libitum during lactation.The cross-bred ewes rearing singles or twins produced significantly more milk in the first 12 weeks (181 and 258 kg respectively) than did pure Blackface ewes in the same period (144 and 208 kg respectively). The pattern of lactation differed between breeds. Cross-bred ewes attained higher maximum yields between the second and sixth weeks (2·37 and 3·32 kg/day, respectively, for single- and twin-suckled ewes) than did the Blackface ewes (1·97 and 2·88 kg/day). The yield from the cross-breds was sustained at a higher level throughout lactation. In the third 4-week period cross-bred ewes rearing single and twin lambs produced 116 and 92%, respectively, of their yield in the first 4 weeks whilst Blackface ewes produced 87 and 65%, respectively.In the sixth week of lactation the milk produced by the two breed groups was similar in quality (mean value of solids-not-fat 11·04% and fat 5·48%) but by the 11th week the fat percentage of milk produced by Blackface ewes had increased significantly to 7·34% whereas that of the cross-breds had remained almost unchanged.Single-suckled ewes gained weight throughout lactation but twin-suckled ewes remained relatively constant. Lambs reared as singles by cross-bred or Blackface ewes reached a mean live weight of 36·2 and 35·0 kg at 102 days of age, respectively, compared with 33·6 and 30·1 kg for twin lambs. Food intake of all ewes increased until the fifth or sixth week of lactation.

Glucocorticoids play a fundamental role in the endocrinology of pregnancy but excess glucocorticoids in utero may lead to abnormalities of fetal growth. Protection against fetal exposure to cortisol is provided by the enzyme 11β-hydroxysteroid dehydrogenase 2 (11β-HSD2) located in the human placental trophoblast. By contrast, relatively little is known concerning the function of glucocorticoid-activating 11β-HSD1, which is strongly expressed within human maternal decidua. To address this we have assessed: i) changes in decidual 11β-HSD1 expression across gestation and ii) the functional role of glucocorticoids in decidua. Human decidua was collected from women undergoing surgical termination of pregnancy in first (n = 32) and second (n = 10) trimesters, and elective caesarean sections in the third trimester (n = 9). Analysis of mRNA for 11β-HSD1 by real-time RT-PCR showed increased expression in second (9.3-fold, P < 0.01) and third (210-fold, P < 0.001) trimesters. Studies using primary cultures of decidual cells also revealed higher levels of cortisol generation in the third trimester. Changes in decidual 11β-HSD1 with gestation were paralleled by increased expression of the apoptosis markers caspase-3 and annexin-V, particularly in cluster designation (CD)10−VE non-stromal cells (20-fold in third trimester relative to first trimester). Apoptosis was also readily induced in primary cultures of third trimester decidual cells when treated with cortisol, cortisone, or dexamethasone (all 100 nM for 24 h). The effect of cortisone but not cortisol or dexamethasone was blocked by an 11β-HSD inhibitor confirming the functional significance of endogenous cortisol generation. These data show that autocrine metabolism of glucocorticoids is an important facet of the feto-placental unit in late gestation and we propose that a possible effect of this is to stimulate programmed cell death in human decidua.

AbstractLow winter growth is a characteristic of male red deer and is caused, in part by a combination of reduced appetite and higher energy expenditure due to cold weather. This study aimed to determine whether housing during winter would reduce energy expenditure and increase the growth rate of male red deer calves. An additional aim was to investigate whether food restriction in winter would be compensated for by increased spring growth. In each of two consecutive years, 80 calves were randomly allocated to eight groups (no. = 10) comprising two replicates of four treatments during winter. Groups were housed inside (I) or outside (O) and given food either ad libitum (AL) or restricted (R) to maintain live weight. Winter treatments (southern hemisphere) ran from 22 May to 25 August (year 1) and from 5 June to 5 September (year 2). During these periods, animals were weighed weekly and group food intake recorded daily. At the end of winter animals were moved outside onto pasture and weighed monthly until the end of spring (27 November, year 1 and 7 December, year 2). In year 2 weighing continued during summer, until 4 April. The animals were slaughtered on 28 November and 18 January (year 1) and 5 April (year 2). The effect of housing on live-weight gain (LWG) and dry-matter intake (DM1) in AL groups was not significant in either year. However in R groups, O had a higher DMI than I in both years (P < 0·05) and a higher LWG than I in year 1 (P < 0·05). LWG was loiver in R than in AL groups in winter in year 1 (P < 0·05) and year 2 (P < 0·001) and live weight was lower in R than in AL groups at the end of winter in both years. Live weight was still lower in R than in AL groups at the end of spring in both years (P < 0·01). In year 2, this live-weight difference was not significant by the end of summer. Hot carcass weight (HCW) was greater in AL animals than R animals (P < 0·05) and dressing proportion was higher in R than in AL (P < 0·05) in year 1. GR (an index of body fatness) was greater (P < 0·05) in O than I in year 1 and was greater (P < 0·05) in AL than in R animals in year 2. Differences in GR between treatments were not significant in either year, with HCW as a covariate.In conclusion, housing calves given food ad libitum during winter did not reduce DMI or increase growth rate. When normal growth rates were prevented by restricting food intake, housing lowered DMI requirement, although such a situation is unlikely to be a useful farm management practice as recovery from the growth check was slow. Annual variations in climate may determine both the food savings made by housing and the extent of compensatory growth of food-restricted animals in spring.

This study set out to determine the stage of gestation at which maternal constraint on fetal growth occurs and whether pregnancy nutritional level could alleviate that constraint. One-hundred and thirty-eight Cheviot (C) and 114 Suffolk (S) ewes were split into two groups and bred with either 12 S or 12 C rams to generate four ewe/fetal groups CC (C dam and C sire), CSinC (crossbred fetus in C ewe), CSinS, and SS. At Day 21 of pregnancy (P21), half of the ewes in each of the four groups were randomly allocated to either a maintenance (M) or ad libitum (A) nutritional treatment, under pastoral grazing conditions. At P100, a subgroup of singleton-bearing ewes including ewes from all four groups (n = 55 in total) were euthanised (Study 1). Maternal, placental and fetal weights and sizes were recorded. The remaining ewes were fed to appetite from P140 and were allowed to lamb (n = 114 in total, Study 2) and lamb liveweights were recorded within 12 h of birth and at average days 30 and 100 (L30, L100) of lactation. In both studies, M ewes were lighter (P < 0.05) than A ewes, and CC and CSinC ewes were lighter (P < 0.05) than CSinS and SS ewes. In Study 1, maternal nutritional treatment had no effect (P > 0.05) on fetal bodyweight although fetuses from A ewes had heavier (P < 0.05) livers, spleens and thyroids than fetuses from M ewes. CC and CSinC fetuses were lighter (P < 0.01) than both CSinS and SS fetuses. In Study 2, lambs born to M ewes were lighter (P < 0.05) at birth and at L100 than lambs born to A ewes. CC lambs were lighter (P < 0.01) than CSinC, CSinS and SS lambs at birth. At L30 and L100, CC lambs were lighter (P < 0.05) than CSinC lambs, which, in turn, were lighter (P < 0.05) than both CSinS and SS lambs, which did not differ (P > 0.05). Combined, these studies indicate that maternal nutrition may have little impact on singleton-offspring development until late gestation while, in contrast, dam size affected fetal size by the end of the second trimester. These data suggest that the C ewe constrains the growth of the crossbred fetus well before a ‘physical’ constraint would be expected.
