Despite the development of powerful molecular biological techniques and technologies, studies involving research animals remain a key component of discovery biology, and in the discovery and development of new medicines. In 1959, The Principles of Humane Experimental Technique, the 3Rs (Replacement, Reduction and Refinement) were developed to provide a framework to ensure animal research was undertaken as humanely as possible. Sixty years since their inception, the extent to which the 3Rs have been adopted and implemented by the global scientific and medical research communities has unfortunately been slow and patchy. However, this situation is changing rapidly as awareness increases, not only of the 3Rs themselves, but of the impact of animal welfare on the reproducibility, reliability and translatability of data from animal studies.

The concerns about reproducibility and validity of animal studies are partly related to poor experimental design and reporting. Here, we undertook a scoping review of the literature to determine the extent and quality of reporting of animal studies on peritoneal dialysis (PD). Online databases were searched to identify 567 relevant original articles published between 1979 and 2018. These were analyzed with respect to bibliographic parameters and general aspects of animal experimentation. A subgroup of 120 studies was analyzed in detail in terms of the impact on the reporting quality of the Animal Research: Reporting of In Vivo Experiments (ARRIVE) guidelines for animal studies. The number of animal studies on PD increased continuously over the years with a thematic shift toward long-term preservation of the peritoneum as a dialyzing organ. There were significant deficiencies in research design with the lack of sample size estimation, randomization, and blinding being the commonest shortcomings. The description of animal numbers, housing conditions, use of medication, and statistical analysis was incomplete. The introduction in 2010 of the ARRIVE guidelines produced very little improvement in the completeness of reporting regardless of journal impact factor. The animal studies on PD suffer from deficits in experimental protocols and transparent reporting. These drawbacks need to be corrected to ensure high-quality and much-needed animal research in PD.

Abstract Public acceptance of genetic modification (GM) technologies may be essential to their continued development, yet few studies have investigated the manner in which demographic and educational factors predict support for GM research. The current study examined attitudes toward animal research and GM in ~400 university undergraduates enrolled in introductory or upper-level psychology courses with material on animal experimentation. Results revealed that men were more accepting of animal and GM research than were women. Enrollment in upper-level psychology classes that addressed specific topics in animal research did not directly predict support for GM research, but such enrollment was associated with increased endorsement of the validity of animal research, which then contributed to acceptance of GM scenarios. The current findings highlight the impact of educational variables on support for animal research, which may then influence attitudes toward GM research.

The current, widely established 3R framework for the ethical use of animals in research consists of three guiding principles, that is, Replacement, Reduction and Refinement, all aiming to safeguard the overarching ethical principle of animal welfare. However, animal welfare alone does not suffice to make animal research ethical if the research does not have sufficient scientific value. The scientific value of animal studies strongly decreases if they are not sufficiently robust, if their questions have already been sufficiently addressed or if the results are selectively reported. Against this background, we argue that three guiding principles are missing, that is, Robustness, Registration and Reporting, all of which aim to safeguard and increase the scientific value of animal research. To establish a new 6R framework, we need a multistakeholder discourse to conceptualise the specific requirements of robustness, registration and reporting and to clarify responsibilities, competencies and legislation for auditing 6R compliance.

SUMMARY—Systematic research evaluating the separate and interacting impacts of neurotoxicants, micronutrients, and social environments on children's cognition and behavior has only recently been initiated. Years of extensive human epidemiologic and animal experimental research document the deleterious impact of lead and other metals on the nervous system. However, discrepancies among human studies and between animal and human studies underscore the importance of variations in child nutrition as well as social and behavioral aspects of children's environments that mitigate or exacerbate the effects of neurotoxicants. In this monograph, we review existing research on the impact of neurotoxic metals, nutrients, and social environments and interactions across the three domains. We examine the literature on lead, mercury, manganese, and cadmium in terms of dispersal, epidemiology, experimental animal studies, effects of social environments, and effects of nutrition. Research documenting the negative impact of lead on cognition and behavior influenced reductions by the Center for Disease Control in child lead-screening guidelines from 30 micrograms per deciliter (μg/dL) in 1975 to 25 μg/dL in 1985 and to 10 μg/dL in 1991. A further reduction is currently being considered. Experimental animal research documents lead's alteration of glutamate-neurotransmitter (particularly N-methyl-D-aspartate) activity vital to learning and memory. In addition, lead induces changes in cholinergic and dopaminergic activity. Elevated lead concentrations in the blood are more common among children living in poverty and there is some evidence that socioeconomic status influences associations between lead and child outcomes. Micronutrients that influence the effects of lead include iron and zinc. Research documenting the negative impact of mercury on children (as well as adults) has resulted in a reference dose (RfD) of 0.1 microgram per kilogram of body weight per day (μg/kg/day). In animal studies, mercury interferes with glutamatergic, cholinergic, and dopaminergic activity. Although evidence for interactions of mercury with children's social contexts is minimal, researchers are examining interactions of mercury with several nutrients. Research on the effects of cadmium and manganese on child cognition and behavior is just beginning. Experimental animal research links cadmium to learning deficits, manganese to behaviors characteristic of Parkinson's disease, and both to altered dopaminergic functioning. We close our review with a discussion of policy implications, and we recommend interdisciplinary research that will enable us to bridge gaps within and across domains.

This article describes, from a personal viewpoint, some of the major advances in refinement in animal experimentation over the past 25–35 years. It is written mainly from a UK perspective. Aspects of public perception and policy, education and training, and the competence of those involved in animal research, are commented upon. Recognition of adverse effects on animals is seen as the key to the refinement of animal experimentation. Elaboration on, and examples of, this theme are given by referring to refinements in animal husbandry and experimental technique. A brief look forward to the changes that may occur in the next 25 years is also included.

AbstractAlone among Western nations, the United States has a two-tier system for welfare protections for vertebrate animals in research. Because its Animal Welfare Act (AWA) excludes laboratory rats and mice (RM), government veterinarians do not inspect RM laboratories and RM numbers are only partially reported to government agencies1. Without transparent statistics, it is impossible to track efforts to reduce or replace these sentient animals’ use or to project government resources needed if AWA coverage were expanded to include them. I obtained annual RM usage data from 16 large American institutions and compared RM numbers to institutions’ legally-required reports of their AWA-covered mammals. RM comprised approximately 99.3% of mammals at these representative institutions. Extrapolating from 780,070 AWA-covered mammals in 2017–18, I estimate that 111.5 million rats and mice were used per year in this period. If the same proportion of RM undergo painful procedures as are publicly reported for AWA-covered animals, then some 44.5 million mice and rats underwent potentially painful experiments. These data inform the questions of whether the AWA needs an update to cover RM, or whether the NIH should increase transparency of funded animal research. These figures can benchmark progress in reducing animal numbers in general and more specifically, in painful experiments. This estimate is higher than any others available, reflecting the challenges of obtaining statistics without consistent and transparent institutional reports.

Abstract Institutions that engage in animal research and production must ensure that high standards of animal care and use meet expectations of society while being ethical stewards of the animals they use in research. In order to achieve engagement in best practices, the Ag Guide is the most appropriate standard for assessing agricultural animals used in research and teaching. The Ag Guide minimizes the potential to overuse performance standards while enhancing the ability to appropriately address specific performance-derived exceptions to situations for which they have been validated. The primary objectives of the standards established in the Ag Guide are well-aligned with the goals of the AAALAC International accreditation program. The Ag Guide provides scientifically-sound, performance-based approaches to animal care and housing, which meet the expectations of AAALAC’s accreditation program. AAALAC provides a third-party peer review of all facets of the animal care and use program that serves as an effective mechanism to ensure institutions meet the standards of the Ag Guide. The process is designed to help identify the strengths and weaknesses of the program to ensure high-quality scientific outcomes and a high level of animal welfare. AAALAC accreditation program for agricultural animal research program is built on the cornerstone of the Ag Guide standards and connects science and responsible animal care. AAALAC accreditation promotes a comprehensive, institutionally supported program with a commitment to continuous improvement, humane and ethical animal care resulting in high-quality animal welfare, and scientific validity. AAALAC takes the position that, in accredited programs, the housing and care for agricultural animals should meet the standards that prevail on a high-quality, well-managed farm and the Ag Guide serves as this foundation. Therefore, the use of the Ag Guide for agricultural animal programs ensures a review that is based on science, professional judgment, and the best interests of the animal.

The ideal animal model would contribute no confounding variables in experimental science. Variables affect experimental design resulting in increased animal use or repeated studies. We demonstrated a simple refinement which may reduce the number of animals used experimentally while simultaneously improving animal welfare. The objective of this study was to determine if the presence of a hut was an impact on physiological stress levels, as determined by faecal cortisol concentration, during a routine four-day acclimatization period of newly received male Hartley-Outbred guineapigs. We hypothesized that those animals provided with huts would have decreased physiological stress compared with animals not provided with huts. We examined this effect within both paired and single-housed animals. A between-subjects one-way analysis of variance revealed that pair-housed animals with a hut had significantly lower faecal cortisol concentration than pair-housed animals without a hut and the presence and absence of a hut had no significant impact on faecal cortisol concentration in single-housed animals. These findings show that presence of a hut is beneficial in reducing physiological stress when pair housing male guineapigs and does not appear to have an impact when single housing male guineapigs. In addition, we have shown that faecal cortisol, and therefore physiological stress, is still increasing on study day 4 suggesting a longer acclimatization period is necessary. A simple refinement in housing environment and acclimatization time can both reduce the number of animals used experimentally and improve animal welfare.

Animal experimentation has been subject to legislative control in the United Kingdom since 1876. This paper reviews the impact of that legislation, which was replaced in 1986, on the teaching of practical physiology to undergraduate students. Highlights and case studies are also presented, drawing on Government reports and statistics, published books and papers, and unpublished archival data.
