Introduction: Association between Cyclin D1 (CCND1) single nucleotide polymorphism (SNP) rs9344 and cancer risk is paradoxical. Thus, we performed a meta-analysis to explore the association between CCND1 variant and overall cancer risk in Indian population. Methods: Data from 12 published studies including 3739 subjects were collected using Pubmed and Embase. RevMan (Review Manager) 5.3 was used to perform the meta-analysis. OR with 95%CI were calculated to establish the association. Results: Overall, the cumulative findings demonstrated that CCND1 polymorphism (rs9344) was not significantly associated with cancer risk in all the genetic models studied (dominant model: GG vs GA+AA: OR (95%CI) = 0.81 (0.60–1.09), P=0.17; recessive model: GG+GA vs AA: OR (95%CI) = 1.23 (0.96–1.59), P=0.11; co-dominant model: GG vs AA: OR (95%CI) = 1.35 (0.93–1.97), P=0.12; co-dominant model: (GG vs GA: OR (95%CI) = 1.16 (0.85–1.59), P=0.34; allelic model: A vs G: OR (95%CI) = 1.20 (1.14–2.85), P=0.23; allelic model: G vs A: OR (95%CI) = 0.83 (0.62–1.12), P=0.23). Subgroup analysis according to cancer types presented significant association of CCND1 polymorphism and increased breast cancer risk in dominant model (GG vs GA+AA: OR = 2.75, 95%CI = 1.54–4.90, P=0.0006) and allelic model (G vs A: OR = 1.63, 95%CI = 1.22–2.19, P=0.001). An increased esophageal cancer risk in recessive model (GG+GA vs AA: OR = 1.51, 95%CI = 1.05–2.16, P=0.03) and co-dominant model (GG vs AA: OR = 2.51, 95%CI = 1.10–5.71, P=0.03) was detected. A higher risk for colorectal cancer was detected under both the co-dominant models (GG vs AA: OR = 2.46, 95%CI = 1.34–4.51, P=0.004 and GG vs GA: OR = 1.74, 95%CI = 1.14–2.67, P=0.01). However, in case of cervical cancer risk a non-significant association was reported under the recessive model (GG+GA vs AA: OR = 1.52, 95%CI = 0.60–3.90, P=0.38) with reference to CCND1 polymorphism (rs9344). The trial sequential analysis (TSA) showed that the cumulative Z-curve neither crossed the trial sequential monitoring boundary nor reached the required information size (RIS). Thus, present meta-analysis remained inconclusive due to insufficient evidence. Conclusion:CCND1 polymorphism rs9344 may not have a role in overall cancer susceptibility in Indian population. However, this polymorphism acts as a crucial risk factor for breast, esophageal, and colorectal cancer but not for cervical cancer. Future studies with larger sample size are required to draw a reliable conclusion.

Objective This meta-analysis was conducted to investigate the relationship between the interleukin (IL)-17A rs2275913 polymorphism and rheumatoid arthritis (RA) susceptibility. Methods Eligible studies were retrieved from PubMed, Embase, and Web of Science. The fixed- or random-effects model was used to calculate the pooled odds ratios (ORs) and 95% confidence intervals (95%CIs) on the basis of heterogeneity. Results Overall, 11 studies containing 4019 RA patients and 4137 controls were included in this meta-analysis. The results suggested a significant association between the IL-17A rs2275913 polymorphism and RA susceptibility in the overall population (allelic model A vs. G: OR = 0.89, 95%CI: 0.83–0.95; heterozygote model GA vs. GG: OR = 0.87, 95%CI: 0.78–0.96; homozygote model AA vs. GG: OR = 0.82, 95%CI: 0.71–0.96; dominant model GA + AA vs. GG: OR = 0.86, 95%CI: 0.78–0.94). In the subgroup analyses, the IL-17A rs2275913 polymorphism was significantly associated with RA risk in Europeans (allelic model A vs. G: OR = 0.87, 95%CI: 0.78–0.97; heterozygote model GA vs. GG: OR = 0.79, 95%CI: 0.68–0.93; dominant model GA + AA vs. GG: OR = 0.79, 95%CI: 0.68–0.92), but not in Africans or Americans. Conclusion This study suggests that the IL-17A rs2275913 polymorphism is significantly associated with RA susceptibility in Europeans. INPLASY registration number: INPLASY202170056.

Objective To investigate the potential correlation between the Arg188His (rs3218536) polymorphism of X-ray repair cross-complementing 2 ( XRCC2) and colorectal cancer (CRC) risk, as the association remains unclear. Methods The CNKI, PubMed, EMBASE and Cochrane library databases were systematically searched for relevant studies published up to July 2021. Data were extracted from included studies, and analysed for pooled or subgroup odds ratios (ORs) with 95% confidence intervals (CIs) using STATA 12.0 software. Results Seven published studies were included. Pooled analysis revealed that the XRCC2 Arg188His polymorphism was associated with increased CRC risk (His versus Arg: OR 1.14, 95% CI 1.01, 1.29). Trial Sequential Analysis to test the power of the results showed that they were unreliable and the meta-analysis required additional studies. Conclusion The current meta-analysis suggests that the XRCC2 Arg188His polymorphism may be a risk factor for CRC.

Objective: The renin–angiotensin system gene has been implicated in the progression of colorectal cancer. Nevertheless, the details of that role remain controversial. We performed a meta-analysis to investigate the correlation between renin–angiotensin system gene polymorphisms and colorectal cancer. Methods: We retrieved relevant studies from PubMed and Embase. Subsequently, fixed or random-effects models were used to calculate pooled odds ratios (ORs) with 95% confidence intervals (CIs). Results: We identified six studies of the angiotensin-converting enzyme insertion/deletion (I/D) polymorphism, and two studies of the angiotensinogen M235T polymorphism. The angiotensin-converting enzyme I/D polymorphism did not significantly correlate with colorectal cancer risk in the total population (DD vs. II: OR 0.77, 95% CI 0.39–1.50; DI vs. II: OR 1.05, 95% CI 0.85–1.30; dominant model: OR 0.94, 95% CI 0.68–1.31; recessive model: OR 1.01, 95% CI 0.80–1.27). Similarly, the angiotensinogen M235T polymorphism was not associated with colorectal cancer risk (TT vs. MM: OR 1.38, 95% CI 0.52–3.67; TM vs. MM: OR 1.19, 95% CI 0.96–1.47; dominant model: OR 1.28, 95% CI 0.77–2.14; recessive model: OR 1.17, 95% CI 0.53–2.59). Conclusion: Our findings suggest that the angiotensin-converting enzyme I/D and angiotensinogen M235T polymorphisms are unlikely to correlate with colorectal cancer.

Background Several studies have reported correlations between BRCA1 polymorphisms rs799917 and rs1799966 with the risk of breast cancer (BC). However, this relationship remains controversial. Methods We conducted a meta-analysis of seven studies to assess the associations between BRCA1 rs799917 and rs1799966 and BC risk, with the aim of more accurately determining the potential correlation. Odds ratios (ORs) and 95% confidence intervals (CIs) were estimated to evaluate the correlation of rs799917 and rs1799966 with BC risk. Results There was no overall correlation between BRCA1 rs799917 and BC risk (TT vs CC: OR = 0.87, 95% CI = 0.66–1.16; CT vs CC: OR = 1.02, 95% CI = 0.89–1.15; dominant model: OR = 0.99, 95% CI = 0.88–1.11; recessive model: OR = 0.87, 95% CI = 0.65–1.16). Subgroup analysis by ethnicity also revealed no significant correlation between rs799917 and BC risk in either Asians or Caucasians. There was also no significant association between BRCA1 rs1799966 and BC risk (GG vs AA: OR = 0.70, 95% CI = 0.33–1.47; AG vs AA: OR = 0.68, 95% CI = 0.35–1.30; dominant model: OR = 0.76, 95% CI = 0.49–1.06; recessive model: OR = 0.82, 95% CI = 0.49–1.36). Conclusion BRCA1polymorphisms rs799917 and rs1799966 were not significantly associated with BC risk in this meta-analysis.

Background. Single nucleotide polymorphisms (SNPs) in genes encoding microRNAs may play important role in the development of gastric cancer. It has been reported that common SNPs rs2910164 in miR-146a and rs11614913 in miR-196a2 are associated with susceptibility to gastric cancer. The published results remain inconclusive or even controversial. A meta-analysis was conducted to quantitatively assess potential association between the two common SNPs and gastric cancer risk.Methods. A comprehensive literature search was performed in multiple internet-based electronic databases. Data from 12 eligible studies were extracted to estimate pooled odds ratios (ORs) and 95% confidence intervals (95% CI).Results. C allele of rs2910164 is associated with reduced gastric cancer risk in heterozygote model and dominant model whereas rs11614913 indicates no significant association. Subgroup analysis demonstrates that C allele of rs2910164 and rs11614913 may decrease susceptibility to diffuse type gastric cancer in dominant model and recessive model, respectively, while rs11614913 increased intestinal type gastric cancer in dominant model.Conclusion. SNPs rs2910164 and rs11614913 might have effect on gastric cancer risk in certain genetic models and specific types of cancer. Further well-designed studies should be considered to validate the potential effect.

Abstract Prostate cancer is one of the most common noncutaneous malignancies in Western countries. Because there has been a debate regarding the relationship between the XRCC1-Arg399Gln and Arg280His polymorphisms and prostate cancer risk, we therefore performed this meta-analysis. The electronic databases PubMed, EMBASE and Medline were searched prior to October 1, 2014. An odds ratio and 95% confidence interval were used to calculate association. Heterogeneity was tested by both a chi-square test and I2statistic. Funnel plots and Egger’s test were used to assess publication bias. All statistical analyses were performed using STATA 12.0 software. A significant association between the XRCC1-Arg399Gln polymorphism and prostate cancer risk was found under a homozygote model and a recessive model. A significant association between XRCC1-Arg280His and prostate cancer risk was found under a heterozygote model and a dominant model. Overall, the results of this meta-analysis show that the XRCC1-Arg399Gln polymorphism may be associated with an increased risk for prostate cancer under the homozygote model and the recessive model. And XRCC1-Arg280His polymorphism is likely to be related with prostate cancer risk under the heterozygote model and the dominant model. Additional larger well-designed studies are needed to validate our results.

Abstract Background: To explore the generic association between miR-499 rs3746444 polymorphism and ischemic stroke (IS). Methods: We performed a systematic review and meta-analysis, odds ratio (OR) and 95% confidence intervals (CIs) were used to estimate the association quantitively. Results: A total of 6 studies (involving 2569 IS cases and 2645 controls) were included. miR-499 (rs3746444) polymorphism showed a statistically significant association with IS risk in the allelic model (G allele vs A allele), the dominant model (GG+AG vs AA), the recessive model (GG vs AG+AA) and the homozygote model (GG vs AA). ORs of the above 4 models were 1.20 (95%CI: 1.02, 1.40), 1.21 (95%CI: 1.01, 1.46), 1.40 (95%CI: 1.04, 1.88), 1.48 (95%CI: 1.10, 2.00) respectively. The I square of the allelic model and the dominant model was 58% and 59%, indicating large heterogeneity among included studies. By sensitivity analysis, I square of the two models dropped to 34.5% and 38.4%, the ORs were 1.26 (95%CI: 1.13, 1.42) and 1.28 (95%CI: 1.12, 1.46), there was still a statistical association between miR-499 (rs3746444) polymorphism and IS. The heterozygote model (AG vs AA) was not statistically significant, the OR was 1.18 (95%CI: 0.99, 1.42), the I square was 54%. Notably, by sensitivity analysis, I square of the heterozygote model dropped to 34.6%, the OR was 1.25 (95%CI: 1.08, 1.43), indicating a statistically significant association between miR-499 (rs3746444) polymorphism and IS. There was no publication bias for all the models by Egger's test. Conclusion: miR-499 (rs3746444) polymorphisms is associated with the increase of IS risk.

Abstract Background: LncRNA metastasis associated with lung adenocarcinoma transcript-1 (MALAT1) was involved in pathogenesis and progress of diverse cancers. To investigate the association of MALAT1 and cancer susceptibility, this meta-analysis was appraised.Methods: 12 studies including 7007 cancer patients and 8791 controls were selected for this meta-analysis. Ratio radiation (ORS) and 95% confidence interval (CIS) were used to assess cancer susceptibility.Results: There was no significant association between rs3200401 polymorphism and the risk of cancer. However, rs3200401 was correlated with an increased risk of digestive cancer in allelic model (OR=1.15, 95%CI=1.04-1.28, P=0.009) and dominant model (OR=1.16, 95%CI=1.02-1.31, P=0.02). There was a borderline association between rs664589 and cancer susceptibility under the dominant model (OR=1.17, 95%CI=1.00-1.38, P=0.05). Rs619586 was associated with decreased cancer risk in all populations under four models (G vs A: OR=0.86, 95%CI=0.78-0.94, P=0.001; GG vs AA: OR=0.60, 95%CI=0.42-0.84, P=0.003; GG+AG vs AA: OR=0.87, 95%CI=0.78-0.97, P=0.009; GG vs AG+AA: OR=0.61, 95%CI=0.44-0.84, P=0.003). Moreover, rs1194338 was decreased associated with cancer susceptibility (A vs C: OR=0.89, 95%CI=0.80-0.98, P=0.01; AA vs CC: OR=0.77, 95%CI=0.62-0.96, P=0.02; AA+AC vs CC: OR=0.87, 95%CI=0.77-1.00, P=0.04; AA vs AC+CC: OR=0.82, 95%CI=0.67-1.00, P=0.05).Conclusion: Our results suggest that rs619586 and rs1194338 are associated with decreased cancer risk, while rs3200401 and rs664589 correlated with increased digestive cancer risk.
