Arcelin-1 is a lectin-like protein found in the seeds of wild varieties of the kidney bean (Phaseolus vulgaris). This protein displays insecticidal properties, but the mechanism of action is as yet unknown. In the present study we investigated the biochemical and biophysical properties of arcelin-1 from Phaseolus vulgaris cv. RAZ-2. Native arcelin-1 is a dimeric glycoprotein of 60 kDa, built from the non-covalent association of two identical monomers. This dimer resists dissociation by chaotropic agents and is highly resistant to proteolytic enzymes. Each subunit contains 10% (w/w) neutral sugars which belong to the high-mannose and complex-type glycans attached to three glycosylation sites. No interaction of the protein with simple sugars could be detected, but arcelin-1 displays an intrinsic specificity in binding complex glycans. Arcelin-1 therefore differs from the closely related phytohaemagglutinin lectins and α-amylase inhibitor in several respects: oligomerization states, sugar-binding affinities and the type and number of glycan chains. These features may be related to the toxicity of arcelin-1.

Определено содержание ингибиторов протеолитических ферментов в фасолевой муке, в пюре из зерновой фасоли, сваренной в воде, и в пюре из зерновой фасоли, сваренной после замачивания в воде в молочной сыворотке. Обосновано использование молочной сыворотки в качестве жидкой среды для варки зерновой фасоли после замачивания. Разработана технология полуфабриката в виде фасолевого пюре, предусматривающая замачивание фасоли в воде, варку в молочной сыворотке, грубое измельчение доведенной до готовности фасоли, последующее тонкое измельчение и охлаждение. Установлено, что в фасолевом пюре активность ингибиторов трипсина значительно ниже, чем в муке из фасоли, что делает пюре более предпочтительным полуфабрикатом для производства широкого ассортимента кулинарной продукции. The content of inhibitors of proteolytic enzymes in bean flour, in puree from beans harvested in water and in puree from cereal beans welded in milk whey after soaking in water is determined. The use of whey as a liquid medium for cooking grain beans after soaking is substantiated. The technology of semi-finished product in the form of bean puree, providing for soaking beans in water, cooking in milk whey, coarse grinding of the bean brought to the ready, subsequent fine grinding and cooling is developed. It has been found that the activity of trypsin inhibitors in bean puree is much lower than in bean flour, which makes mashed potatoes a more preferred semi-finished product for the production of a wide range of culinary products.

The highly conserved HOX homeodomain (HD) transcription factors (TFs) establish the identity of different body parts along the antero–posterior axis of bilaterian animals. Segment diversification and the morphogenesis of different structures is achieved by generating precise patterns of HOX expression along the antero–posterior axis and by the ability of different HOX TFs to instruct unique and specific transcriptional programs. However, HOX binding properties in vitro, characterised by the recognition of similar AT-rich binding sequences, do not account for the ability of different HOX to instruct segment-specific transcriptional programs. To address this problem, we previously compared HOXA2 and HOXA3 binding in vivo. Here, we explore if sequence motif enrichments observed in vivo are explained by binding affinities in vitro. Unexpectedly, we found that the highest enriched motif in HOXA2 peaks was not recognised by HOXA2 in vitro, highlighting the importance of investigating HOX binding in its physiological context. We also report the ability of HOXA2 and HOXA3 to heterodimerise, which may have functional consequences for the HOX patterning function in vivo.

Allelopathy, an important mechanism in invasive species stems from the lack of coevolved tolerance of indigenous species to novel chemicals released by the invader species. These chemicals are key to successful invasive of species of natural plant communities and ecosystems. This study analyzed the allelopathic effects of the invasive plant, Parthenium hysterophorous on the growth of kidney beans (Phaseolus vulgaris). One hundred and fifty viable kidney bean seeds were washed in distilled water, and transferred in triplicates to pots with wet sterile sand. Ten grams of freshly collected Parthenium leaves and roots were weighed, grounded separately using a mortar and a pestle, mixed with 100 ml of distilled water in a beaker and left for 24 hours in a dark at a room temperature (250C). The aqueous extract filtrates were used in experimental treatments. While untreated (control) seeds germinated on day 4 seeds treated with leaf extract germinated on day 14 and day 9 for the root extract. Leaf extract inhibited germination and sprouting of new leaves more effectively than root extract. Leaves in control seeds appeared on day 1 after germination. In treated seeds, leaves appeared on the 4th day then stagnated until day 15 when more leaves sprouted. Root extract significantly depressed seed germination. The number of new leaves increased significantly in control seedlings compared to the treated ones. Combined root and leaf extracts highly significantly inhibited of seed germination and the number of new leaves. Additionally, combined treatment with root and leaf extracts significantly inhibited shoot growth, and the increase in leaf length and width. A comparison of the effectiveness of the root and leaf extracts showed that leaf extract was more effective in inhibiting shoot growth and leaf width than the root extract.
