Cell extracts from Bacillus brevis (A.T.C.C. 10068), grown with various media, incorporated certain 14C-labelled amino acids that are normally components of tyrothricin into material that was extracted by ethanol from the precipitate formed by adding acid. When this material was separated by paper and silica-gel thin-layer chromatography and paper electrophoresis 14C was located in those regions that also contained gramicidin and tyrocidine. From a study of the properties of the system responsible for the incorporation it was deduced that non-tyrothricin materials were present. It was shown that the methods normally used to characterize tyrothricin do not adequately distinguish between tyrothricin and non-tyrothricin materials. However, a method for separating these materials was devised. This involved elution with ethanol from columns of acid alumina followed by gel filtration on Sephadex LH-20 with dimethylformamide–water solvent. The behaviour of gramicidin and tyrocidine on the Sephadex LH-20 column was examined, and it was concluded that the separation was not caused simply by gel filtration of unassociated molecules. Also, tyrocidine molecules with different amino acid compositions seemed to have different affinities for the Sephadex LH-20 column.

Tryptensin, a vasopressor substance generated from human plasma protein fraction IV-4 by trypsin, has been isolated and the amino acid composition analysed. The procedures used for the isolation were: (a) adsorption of the formed tryptensin on Dowex 50W (X2; NH4+ form); (b) gel filtration through Sephadex G-25; (c) cation-exchange chromatography on CM-cellulose; (d) anion-exchange chromatography on DEAE-cellulose; (e) re-chromatography on CM-cellulose; (f) gel filtration on Bio-Gel P-2; (g) partition chromatography on high-pressure liquid chromatography. The homogeneity of the isolated tryptensin was confirmed by thin-layer chromatography and thin-layer electrophoresis. The amino acid analysis of the hydrolysate suggested the following proportional composition: Asp, 1; Val, 1; Ile, 1; Tyr, 1; Phe, 1; His, 1; Arg, 1; Pro, 1. This composition is identical with that of human angiotensin.

An isozyme of S-adenosylmethionine synthetase has been purified to homogeneity by ammonium sulfate fractionation, DEAE-cellulose column chromatography, and gel filtration on a Sephadex G-200 column. The purified enzyme is very unstable and has a molecular weight of 120 000 consisting of two identical subunits. Amino acid analysis on the purified enzyme showed glycine, glutamate, and aspartate to be the most abundant and the aromatic amino acids to be the least abundant. It possesses tripolyphosphatase activity which can be stimulated five to six times by S-adenosylmethionine (20–40 μM). The findings support the conclusion that an enzyme-bound tripolyphosphate is an obligatory intermediate in the enzymatic synthesis of S-adenosylmethionine from ATP and methionine.

AbstractThe effects of 5, 20, and 40 miracidia dose exposures of Echinostoma caproni on the amino acid contents of Biomphalaria glabrata were studied using high performance thin-layer chromatography-densitometry. Amino acids were identified and quantified in whole bodies of exposed snails and in the uninfected matched controls at 2 and 4 weeks post-exposure. Using cellulose layers with the mobile phase 2-butanol-pyridine-glacial acetic acid-deionized water (39:34:10:26) and ninhydrin detection reagent [2% ninhydrin in acetone-n-butanol (1:1)], five amino acids were identified, i.e., leucine/isoleucine, valine, alanine, glycine, and ornithine, by hRF value comparison and color differentiation. Quantitatively, there was a marked elevation in the amounts of four of these five amino acids (isoleucine/leucine, valine, alanine, and ornithine) across dose levels at 4 weeks post-infection (P<0.05). Elevation of the amino acid content in the high dose snail group suggested that some changes occurred in the amino acid metabolism of the snails in that group as a function of miracidia dose.

AbstractSeveral methods to determine amino acids and vitamins in biological and pharmaceutical samples have been reported. Thin layer chromatography (TLC) finds its place when the relatively costly equipment required by other methods is unavailable. This review covers the 1991–2010 literature on TLC/HPTLC (high performance thin layer chromatography) amino acid and vitamin determinations. It gives an overview of the special features as well as the problems in TLC/HPTLC determinations of amino acids and vitamins. Various chromatographic systems useful in amino acid and vitamin identification, separation and quantitation of are presented in tabular form. Future prospects of TLC/HPTLC for amino acid and vitamin determinations are also discussed.

Quantitative investigation of Sceliphron caementarium venom, obtained by electrical excitation of the wasp, has shown an unexpectedly small protein fraction of the dried venom. A few percent of the venom consists of the free amino acids histidine, methionine, and pipecolic acid. Three other components have been isolated by paper and thin-layer chromatography and are found to give weak ninhydrin reactions, in addition to certain color tests characteristic of alkaloids.

The stability of disodium sulfobenzylpenicillin (SB-PC) in several large-volume parenteral solutions was studied. A study was also made of the influences of other additive drugs upon the stability of SB-PC in SB-PC IV fluid admixtures. Quantitative determinations were made by iodometry of the residual rate over time of SB-PC. The residual rate of SB-PC in 5-fluorouracil (5-FU injection)-10 percent glucose admixture was 67 percent after 24 hours at 25 °C, (77 °F) and in aminophylline (aminophylline injection J.P.)-10 percent glucose admixture it was 54 percent after 24 hours. It was found that the decomposition of SB-PC was due to amines which were added to these injections. In an admixture of amino acid and SB-PC, the residual rate of SB-PC decreased linearly with the increase in amino acid concentration. In the 12 kinds of amino acids used in this study, the greatest decrease was observed in the admixture with lysine, the residual rate being 76.7 percent. For the study of degradation products of SB-PC, thin layer chromatography was employed and a spot of the degradation product was detected at an Rf value of 0.2.

The distribution of individual amino acids and monosaccharides in fulvic acid and its fractions separated by polyamide chromatography was investigated in five different Italian soils. Although little differences were generally found in the two polyamide fractions (FI and FII), the highest percentage content of acidic amino acids and the lowest percentage content of neutral amino acids have been found in the second one (FII); monosaccharides composition was more irregular, but generally FII contained more pentoses. Both chromatographic fractions (FI and FII) have been chromatographed on Sephadex G-25. The composition in carbohydrate and amino acid components of the further different fractions resolved by gel filtration showed great differences depending on the molecular weight distribution.
