A smart textile for live bacteria detection of antimicrobial hospital tissues is here proposed. The capacity to detect viable bacteria is based on the use of Prussian Blue (PB) as electrochromic compound, with a clear reversible change of colour from PB to Prussian White (PW) after reduction from a bacterial metabolism process. PB nanoparticles are incorporated to polyester cotton fabrics by ultrasonic deposition. After performing different tests with bacterial samples of E. coli and S. aureus, a full colour change of the textiles was observed. These smart textiles will allow to determine the self-life of the antibacterial compounds as well to improve the control of hospital infections.

Aim. Elucidation of the role of extrachromosomal elements of heredity in manifestations of toxic properties of Yersinia pestis. Materials and methods. The study was carried out in vaccine strain Y. pestis EV76 (pMTl, pCDl, pPCPl) and non-plasmid variants of vaccine EV76 (pMTl\ pCDl', pPCPl') and virulent 231 (рМТГ, pCDl’, pPCPl') strains of Y. pestis. Presence of functionally active form of lipopolysaccharide (LPS) in the incubation medium of the bacteria was evaluated via toxicity of supernatant of Y. pestis for intact animals (infection-toxic shock) and mice sensitized by D-GalN. Results. 37°C cultures of Y. pestis EV76 containing a full amount of plasmids were established to release LPS into the environment. Non-plasmid variants of both vaccine and virulent strains of Y. pestis pMTl', pCD Г, рРСР 1 do not have this ability. Separation of LPS from cell wall was detected in live bacteria of plague infectious agent. This process is assumed to be coupled with translocation of proteins coded by pMTl, pCDl, pPCPl plasmids from the cell into the environment. Conclusion. Functional interconnection between extrachromosomal elements of heredity and toxic activity of Y. pestis LPS is established for the first time.

Introduction: Alternating and pulsed electromagnetic magnetic fields (AEMF and PEMF) of different amplitudes and frequencies can induce metabolic and proliferative effects in osteoblasts, but there is no clearly directed tendency of these effects. I hypothesize that there are extremely low triggering parameters of alternating electromagnetic field (EMF) intensity, i.e., above the background magnetic field on earth but below the lowest AEMF and PEMF that have been investigated to date (above 0.07 mT and below 0.4 mT) that induce cellular response. Methods: Accordingly, human monolayer explant culture replica were exposed four times in 24-hour intervals to two minutes of 10 kHz AEMF or PEMF (10 Hz pulses at a basic 5 kHz frequency) with a maximal EMF intensity of 0.2 mT for both. Cell proliferation was estimated from microscopic cell counting and cell death by lactate dehydrogenase (LDH) specific activity in culture media (measured using a colorimetric method). The early marker of osteoblast maturation, cellular alkaline phosphatase (AP) specific activity, was measured using a colorimetric method (n=6 for all experiment conditions). Results: No difference was found in cell numbers in the culture samples exposed either to AEMF or PEMF and in the LDH’s specific activity in culture media in comparison to the unexposed controls (p>0.05, for both). The cellular AP’s specific activity increased significantly only in cell cultures exposed to the 10 kHz AEMF (p=0.011). Conclusions: The triggering for human osteoblast activation for maturation by an extremely low AEMF (10 kHz) is at least 0.2 mT, which is distinct and below the previously found triggering range of a PEMF for proliferation induction. Therefore, application of these EMF parameters in a clinical setup by a separate finetuning of osteoblast proliferation and maturation might have a therapeutic value in enhancing damaged bone regeneration.

Abstract BackgroundStem cells from the apical papilla (SCAPs) are important for tooth root development and regeneration of root dentin. Here, we examined the expression of programmed cell death protein-1 (PD-1) in SCAPs and investigated the effect of PD-1 on odontogenic and osteogenic differentiation and the relationship between PD-1 and cell differentiation and SHP2/NF-κB signals.MethodsSCAPs were obtained culture in the related medium. The proliferation ability was evaluated by cell counting kit 8 and 5‐ethynyl‐20‐deoxyuridine (EdU) assay. Alkaline phosphatase (ALP) activity assay, ALP staining, western blot, real-time RT-PCR, Alizarin Red S staining, and immunofluorescence staining were performed to explore the osteo/odontogenic potential and the involvement of SHP2/NF-κB pathways. Besides, we transplanted SCAPs component into mouse calvaria defects to evaluate osteogenesis in vivo. ResultsWe found that human SCAPs expressed PD-1 for the first time. PD-1 knockdown enhanced the osteo/odontogenic differentiation of SCAPs by suppressing SHP2 pathway and activating NF-κB pathway. Overexpression of PD-1 inhibited the osteogenesis and odontogenesis of SCAPs via activation of SHP2 signal and inhibition of NF-κB pathway. ConclusionPD-1 activated SHP2 signal to block NF-κB signal and then played a vital role in osteo/odontogenic differentiation of SCAPs.

Principal Components and Classification Analysis (PC&CA) represents one of the most utilised multivariate chemometric techniques, having the advantage to use many measurements for a single sample in the same time, being recommended for understanding better the complexity of one phenomenon. The aim of this paper was to use PC&CA to study the effects of different types of fertilizers on polyphenols content of forages harvested in autumn from permanent grassland. Gravimetrically was established the matrix of floristic composition. The experimental field was fertilized first time in 2003, organic and/or NPK mineral. The determination of polyphenols contents was made using UV-VIS SPECORD 205 spectrophotometer, in conformity to chemical Folin and Ciocalteu colorimetric method. The highest polyphenols content was identified in forages from unfertilized variant (108 µM gallic acid/g). PC&CA can be a useful tool in describing the modification of polyphenols contents of forages under the effects of organic and/or mineral fertilisation of permanent grassland.

The synthesis of new calix[4]arenes adopting a cone stereoisomeric form bearing two or four azide fragments on the upper rim and water-soluble triazolyl amphiphilic receptors with two or four polyammonium headgroups via copper-catalyzed azide–alkyne cycloaddition reaction has been performed for the first time. It was found that the synthesized macrocycles form stable aggregates with hydrodynamic diameters between 150–200 nm and electrokinetic potentials about +40 to +60 mV in water solutions. Critical aggregation concentration (CAC) values were measured using a micelle method with pyrene and eosin Y as dye probes. The CAC values of tetraalkyl-substituted macrocycles 12a,b (5 µM for both) are significantly lower than those for dialkyl-substituted macrocycles 10a,b (790 and 160 µM, respectively). Premicellar aggregates of macrocycles 10a,b and 12a,b with the dye eosin Y were used for nucleotides sensing through a dye replacement procedure. It is unusual that disubstituted macrocycles 10a,b bind more effectively a less charged adenosine 5'-diphosphate (ADP) than adenosine 5'-triphosphate (ATP). A simple colorimetric method based on polydiacetylene vesicles decorated with 10b was elaborated for the naked-eye detection of ADP with a detection limit of 0.5 mM.

This study aimed to examine the spatial cleaning effect of ultrasonic irrigation in simulated root canal with oblong cross section in the absence of antimicrobial agent. A 7-day E. faecalis biofilm was cultivated in a rectangular, simulated canal model and subjected to passive ultrasonic irrigation (PUI) with sterile saline for 5 minutes. After that, the biofilm was examined by confocal microscopy after bacterial viability staining at 58 sites around and beyond the endosonic file. Results showed that, at the vicinity of the file, the amount of viable bacteria ranged from 13.1% (2.75 mm from the tip) to 40.5% (4.5 mm level). Lesser amounts of live bacteria were observed within 1 mm from the vibrating file, which amount increased for sites farther away. At 3 mm distance, the amount of bacteria (35.5 to 64.4%) was significantly greater than areas situated close to the vibrating file (P < 0.01). Sites next to the anti-nodes of file had less bacteria remaining than sites near the nodes (P = 0.050). Apically (0.5 mm or more), the amount of bacteria was significantly greater than that at the file tip (16.4%) (P < 0.05). It was concluded that PUI is able to dislodge a single-species biofilm, provided that they are situated in close vicinity to the vibrating file.
