Abstract Morphine antibody purified by affinity chromatography was used to develop a solid-phase radioimmunoassay for morphine in polystyrene tubes. The tubes are coated with an appropriate concentration of the purified antibody, rinsed three times with buffered saline, and stored at -15 degrees C. Using tritiated dihydromorphine, we determined competitive morphine binding by difference when the radioactivity in the assay supernates was measured after incubation (1 h, 37 degrees C). Five standard curves, with use of serum equivalents of morphine ranging from 0 to 6 mug/liter, were linear and had a mean correlation coefficient of 0.98. Uncer conditions of the assay, levorphanol was comparable to morphine in its inhibitory effect on binding of labeled dihydromorphine, whereas dextrorphan was essentially inactive. Morphine-3-glucuronide, a major metabolite, is 55-fold less inhibitory in terms of its capacity to displace the radiolabel. We believe that the sensitivity of the technique, coupled with the simplicity of nonseparatory sampling, renders the system suitable for rapid determination of morphine and related compounds in biological fluids.

Abstract Domoic acid is the toxin responsible for incidents of amnesic shellfish poisoning. A rapid extraction and cleanup for the liquid chromatographic determination of domoic acid in unsalted seafood is reported. The method uses a single-step extraction with 50% aqueous methanol and a selective cleanup and preconcentration with strong-anion exchange, solid-phase extraction. Determination is performed by liquid chromatography with ultraviolet absorbance detection. The detection limit was 20–30 ng/g. Recoveries of 93% were achieved from 0.2 to 20 μg/g in mussel tissues. The method gave a precision of less than 3% for concentrations greater than 2 μg/g and less than 6% at 0.2 μg/g. A linear dynamic range of 104 can be achieved. The method was successfully applied to a variety of seafood products, including mussels, razor clams, crabs, and anchovies.

Abstract Methylmalonic acid concentrations are increased in serum in vitamin B12 (cobalamin) deficiency. Here I demonstrate the successful use of anion-exchange extraction for improving newly developed analytical procedures and describe well-documented, reliable performance of this method for rapid determination of methylmalonic acid. The sorbent counter ion is formate, and the elution solvent is formic acid. The dicyclohexyl derivative is measured by selected ion monitoring. For serum, the assay curve is linear from 0.026 to 200 mumol/L. The normal reference interval is 0.08 to 0.56 mumol/L. Added methylmalonic acid is accurately quantified. The sensitivity and the precision exceed those of the current method by three orders of magnitude. The total and within-day CVs are 4.6% to 7.9% and 2.6% to 4.7%, respectively. Similar figures were obtained for urine. This convenient method is useful for evaluation of cobalamin deficiency, especially in patients with normal or moderately depressed cobalamin concentrations in serum.
