In this study, poly(∊-caprolactone) (PCL), gelatin (GEL) and nanocrystalline hydroxyapatite (HAp) was applied to fabricate novel PCL-GEL-HAp nanaocomposite scaffolds through a new fabrication method. With the aim of finding the best fabrication method, after testing different methods and solvents, the best method and solvents were found, and the nanocomposites were prepared through layer solvent casting combined with freeze-drying. Acetone and distillated water were used as the PCL and GEL solvents, respectively. The mechanical test showed that the increasing of the PCL weight through the scaffolds caused the improvement of the final nanocomposite mechanical behavior due to the increasing of the ultimate stress, stiffness and elastic modulus (8 MPa for 0% wt PCL to 23.5 MPa for 50% wt PCL). The biomineralization investigation of the scaffolds revealed the formation of bone-like apatite layers after immersion in simulated body fluid (SBF). In addition, the in vitro cytotoxity of the scaffolds using L929 mouse fibroblast cell line (ATCC) indicated no sign of toxicity. These results indicated that the fabricated scaffold possesses the prerequisites for bone tissue engineering applications.

Three-dimensional (3D) printing application is a promising method for bone tissue engineering. For enhanced bone tissue regeneration, it is essential to have printable composite materials with appealing properties such as construct porous, mechanical strength, thermal properties, controlled degradation rates, and the presence of bioactive materials. In this study, polycaprolactone (PCL), gelatin (GEL), bacterial cellulose (BC), and different hydroxyapatite (HA) concentrations were used to fabricate a novel PCL/GEL/BC/HA composite scaffold using 3D printing method for bone tissue engineering applications. Pore structure, mechanical, thermal, and chemical analyses were evaluated. 3D scaffolds with an ideal pore size (~300 µm) for use in bone tissue engineering were generated. The addition of both bacterial cellulose (BC) and hydroxyapatite (HA) into PCL/GEL scaffold increased cell proliferation and attachment. PCL/GEL/BC/HA composite scaffolds provide a potential for bone tissue engineering applications.

Abstract In tissue engineering, 3D printing is an important tool that uses biocompatible materials, cells, and supporting components to fabricate complex 3D printed constructs. This review focuses on the cytocompatibility characteristics of 3D printed constructs, made from different synthetic and natural materials. From the overview of this article, inkjet and extrusion-based 3D printing are widely used methods for fabricating 3D printed scaffolds for tissue engineering. This review highlights that scaffold prepared by both inkjet and extrusion-based 3D printing techniques showed significant impact on cell adherence, proliferation, and differentiation as evidenced by in vitro and in vivo studies. 3D printed constructs with growth factors (FGF-2, TGF-β1, or FGF-2/TGF-β1) enhance extracellular matrix (ECM), collagen I content, and high glycosaminoglycan (GAG) content for cell growth and bone formation. Similarly, the utilization of 3D printing in other tissue engineering applications cannot be belittled. In conclusion, it would be interesting to combine different 3D printing techniques to fabricate future 3D printed constructs for several tissue engineering applications.

AbstractBiocompatibility of materials used in dental and biomaterials applications is very important and depends on the components of these materials. Photopolymerized materials for dental and biomaterials applications have been progressively used since the 1970s. One of the crucial components in these materials is the photoinitiator (PI) that initiates the polymerization reaction. Synthetic PIs are the most commonly used types, but owing to their drawbacks such as cytotoxicity, insolubility in water, and high cost, research on naturally derived (bio-sourced) PIs is growing, to find an alternative to these synthetic types, especially in the growing field of three-dimensional (3D) printing and bioprinting of biomaterials for tissue engineering applications. Naturally derived PIs are biocompatible, highly water-soluble, and abundant. Naturally derived PIs have been used to prepare experimental dentine bonding agents, dentine primers, photo-crosslinked hydrogels for tissue engineering applications, antibacterial coatings, guided tissue regeneration membranes, and 3D printed biomaterials. An electronic search was done using MEDLINE/PubMed and Scopus databases using the keywords naturally derived, bio-sourced, PIs, dental, biomaterials, 3D printing, and 3D bioprinting, to review potential naturally derived PIs for dental and biomaterials applications. There are a variety of naturally derived PIs with various colors and absorption spectra to choose from, according to the intended application. Most of naturally derived PIs can be used with modern conventional dental light curing units, making them applicable for experimental studies for potential dental and biomaterials applications. Due to their biocompatibility and availability it is expected that in the upcoming years, research on naturally derived PIs and their dental and biomaterials applications will increase especially in the growing field of 3D bioprinting in which cell viability is essential; thus this review was done.
