AbstractProper hydration of the stratum corneum, the skin’s outermost layer, is essential for healthy skin. Water-soluble substances called natural moisturizing factors (NMF) are responsible for maintaining adequate moisture in the skin and are closely associated with a variety of the skin’s functions. Therefore, quantitative analysis methods for NMF are indispensable when attempting to clarify one of the mechanisms of hydration and its effect on the skin. This study sought to develop a quick and simple analytical technique, which can quantify NMF from the skin without the need for extraction or separation, using direct analysis in real time-mass spectrometry (DART-MS). The goal was to deliver a high quantitative capability, so a unique inkjet printing technique was employed to evenly coat a stable isotope-labeled internal standard (SIL-IS) on tape-stripped skin. This technique allowed for the quantification of 26 NMF with established calibration curves and comparatively high linear correlations. The speed of measurement was found to be advantageous as 100 strips of tape can be measured in roughly 2 hours. The effectiveness of the inkjet coating was also verified by comparing its precision with that of conventional pipetting. This new technique can be an alternative method to quantify NMF rapidly and perhaps allow for a clearer elucidation of their function in skin.

Determination of T-2 and HT-2 toxins from maize by direct analysis in real time mass spectrometry

Direct analysis in real time (DART) ionisation coupled to mass spectrometry (MS) was used for the rapid quantitative analysis of T-2 toxin (T-2) and the related HT-2 toxin (HT-2), extracted from maize. Sample preparation procedures and instrument parameters were optimised to obtain sensitive and accurate determination of the toxins. The lowest calibration levels were 50 μg/kg for T-2 and 300 μg/kg for HT-2. Quantitative analysis was performed with the use of matrix-matched standards employing the 13C-labelled internal standard for T-2. DART-MS of maize extracts spiked with T-2 gave a linear response over the range of 50-1000 μg/kg. With the isotope dilution technique, good recoveries (99-110%) and repeatabilities (relative standard deviaiton 7.4-11.6%) were obtained at T-2 spiking levels of 100 and 1000 μg/kg. Adaptability of the developed method was demonstrated by analysis of T-2 and HT-2 from an oat flour quality control material. The results here further indicate the potential for application of ambient ionisation mass spectrometry to provide accurate, convenient quantitation of mycotoxins from grains.

A series of poly(ethylene-co-vinyl alcohol)/poly(ε-caprolactone) blends with different compositions were prepared using solvent casting. The miscibility of this pair of polymers was investigated using differential scanning calorimetry (DSC), and proved by a negative Flory interaction parameter value calculated from the Nishi–Wang equation. The miscibility of this blend was also confirmed by scanning electronic microscopy (SEM). The thermal behaviors of the obtained materials were investigated by DSC, thermogravimetric analysis, and direct analysis in real-time–time-of-flight mass spectrometry and the results obtained were very relevant. Furthermore, the crystalline properties of the obtained materials were studied by DSC and X-ray diffraction where the Ozawa approach was adopted to investigate the non-isothermal crystallization kinetics. The results obtained revealed that this approach described the crystallization process well.
