A novel ligand with double sulfinyl groups, bis(benzylsulfinyl)methane L, was synthesized by a new method. Its novel ternary complex, has been synthesized [using L as the first ligand, and dipyridyl L' as the second ligand]. In order to study the effect of the second ligand on the fluorescence properties of rare-earth sulfoxide complex, a novel binary europium complex has been synthesized. Photoluminescent measurement showed that the first ligand L could efficiently transfer the energy to Eu (III) ions in the complex. Furthermore, the detailed luminescence analyses on the rare earth complexes indicated that the ternary Eu (III) complex manifested stronger fluorescence intensities, longer lifetimes, and higher fluorescence quantum efficiencies than the binary Eu (III) materials. The fluorescence emission intensities and fluorescence lifetimes of the ternary complex enhanced more obviously than the binary complex.

Chemical modifications have been extensively used for therapeutic oligonucleotides because they strongly enhance the stability against nucleases, binding affinity to the targets, and efficacy. We previously reported that oligonucleotides modified with an N-methylguanidine-bridged nucleic acid (GuNA[Me]) bearing the thymine (T) nucleobase show excellent biophysical properties for applications in antisense technology. In this paper, we describe the synthesis of GuNA[Me] phosphoramidites bearing other typical nucleobases including adenine (A), guanine (G), and 5-methylcytosine (mC). The phosphoramidites were successfully incorporated into oligonucleotides following the method previously developed for the GuNA[Me]-T-modified oligonucleotides. The binding affinity of the oligonucleotides modified with GuNA[Me]-A, -G, or -mC toward the complementary single-stranded DNAs or RNAs was systematically evaluated. All of the GuNA[Me]-modified oligonucleotides were found to have a strong affinity for RNAs. These data indicate that GuNA[Me] could be a useful modification for therapeutic antisense oligonucleotides.

A fluorochrome has been isolated in analytically pure form from a commercial sample of the triaryl-methane dye aniline blue. Its structure has been shown to be sodium 4,4'-[carbonylbis(benzene-4,1-diyl)bis(imino)]bisbenzenesulfonate by spectroscopic means and confirmed by synthesis. Its fluorescence emission, which is markedly solvent-dependent, is 150 times greater in butan-1-ol than in water (however, the wavelength of the emission maximum is not altered significantly). In the presence of a cationic surfactant, hexadecyltrimethylammonium bromide, the fluorescence intensity reaches a maximum at approximately the critical micelle concentration of the surfactant.

AbstractConstant (C)-region switching of heavy (H) and/or light (L) chains in antibodies (Abs) can affect their affinity and specificity, as demonstrated using mouse, human, and chimeric mouse-human (MH) Abs. However, the consequences of C-region switching between evolutionarily distinct mammalian and avian Abs remain unknown. To explore C-region switching in mouse-chicken (MC) Abs, we investigated antigen-binding parameters and thermal stability of chimeric MC-6C407 and MC-3D8 IgY Abs compared with parental mouse IgGs and chimeric MH Abs (MH-6C407 IgG and MH-3D8 IgG) bearing identical corresponding variable (V) regions. The two MC-IgYs exhibited differences in antigen-binding parameters and thermal stability from their parental mouse Abs. However, changes were similar to or less than those between chimeric MH Abs and their parental mouse Abs. The results demonstrate that mammalian and avian Abs share compatible V-C region interfaces, which may be conducive for the design and utilization of mammalian-avian chimeric Abs.

Five new meso-tetrakis(1-arylpyrazol-4-yl)porphyrins were synthesized to investigate their fluorescence properties. Preparation of these porphyrins was carried out by cyclization of tetramethoxypropane with substituted phenylhydrazine, followed by formylation to give the corresponding aldehydes, which reacted with pyrrole under the Adler reaction condition to get the target porphyrins (1a-1e). All the porphyrins were characterized by 1 H NMR, elemental analysis, UV-vis spectra and mass spectra. Red fluorescence emission of these porphyrins was observed in fluorescence spectra. Compared with meso-tetraphenylporphyrin (TPPH2), these meso-tetrakis(1-arylpyrazol-4-yl) porphyrins had a significant red shift in UV-vis and fluorescence spectra with increased fluorescence quantum yields.

Fluorescence properties of benzo[ghi]perylene (BPe) and ovalene (Ov) dissolved in 25 solvents of varying polarity are reported. Measurements indicate that emission intensities depend on solvent polarity. The BPe and Ov solvent polarity scales are defined as the ratio of the fluorescence emission intensities of bands I and III of the vibronic spectra. Benzo[ghi]perylene and ovalene solute probes enable fluorescence measurements to be made in spectral regions less prone to solvent inner filtering and other artifacts which have hampered the use of pyrene (Py) as a polarity probe molecule.
