Background: Morin has many pharmacological functions including antioxidant, anticancer, anti-inflammatory, and antibacterial effects. It is commonly used in the treatment of antiviral infection, gastropathy, coronary heart disease and hepatitis B in clinic. However, researches have shown that morin is likely to show prooxidative effects on the cells when the amount of treatment is at high dose, leading to the decrease of intracellular ATP levels and the increase of necrosis process. Therefore, it is necessary to determine the concentration of morin in biologic samples. Method: Novel water-soluble and green nitrogen and sulfur co-doped carbon dots (NSCDs) were prepared by a microwave heating process with citric acid and L-cysteine. The fluorescence spectra were collected at an excitation wavelength of 350 nm when solutions of NSCDs were mixed with various concentrations of morin. Results: The as-prepared NSCDs were characterized by transmission electron microscopy, X-ray diffraction and X-ray photoelectron spectroscopy. The fluorescence intensity of NSCDs decreased significantly with the increase of morin concentration. The fluorescence intensity of NSCDs displayed a linear response to morin in the concentration 0.10-30 μM with a low detection limit of 56 nM. The proposed fluorescent probe was applied to analysis of morin in human body fluids with recoveries of 98.0-102%. Conclusion: NSCDs were prepared by a microwave heating process. The present analytical method is sensitive to morin. The quenching process between NSCDs and morin is attributed to the static quenching. In addition, the cellular toxicity on HeLa cells indicated that the as-prepared NSCDs fluorescent probe does not show obvious cytotoxicity in cell imaging. Our proposed method possibly opens up a rapid and nontoxic way for preparing heteroatom doped carbon dots with a broad application prospect.

Angelica polymorpha Maxim. (APM) is used in traditional medicine to treat chronic gastritis, rheumatic pain, and duodenal bulbar ulcers. However, it is not known whether APM has epidermis-associated biological activities. Here, we investigated the effects of APM flower absolute (APMFAb) on responses associated with skin wound healing and whitening using epidermal cells. APMFAb was obtained by solvent extraction and its composition was analyzed by GC/MS. Water-soluble tetrazolium salt, 5-bromo-2′-deoxyuridine incorporation, Boyden chamber, sprouting, and enzyme-linked immunosorbent assays and immunoblotting were used to examine the effects of APMFAb on HaCaT keratinocytes and B16BL6 melanoma cells. APMFAb contained five compounds and induced keratinocyte migration, proliferation, and type IV collagen synthesis. APMFAb also induced the phosphorylations of ERK1/2, JNK, p38 mitogen-activated protein kinase, and AKT in keratinocytes. In addition, APMFAb decreased serum-induced B16BL6 cell proliferation and inhibited tyrosinase expression, melanin contents, and microphthalmia-associated transcription factor expression in α-melanocyte-stimulating hormone-stimulated B16BL6 cells. These findings demonstrate that APMFAb has beneficial effects on skin wound healing by promoting the proliferation, migration, and collagen synthesis of keratinocytes and on skin whitening by inhibiting melanin synthesis in melanoma cells. Therefore, we suggest that APMFAb has potential use as a wound healing and skin whitening agent.

Peroxynitrite (ONOO−) is one of the sources of oxidation stress involved in many biological signaling pathways. The role of ONOO− being a double-edged sword in biological systems drives the development of effective detection tools. In this work, a boronate-based polymeric fluorescent probe PB-PVA was synthesized and the probe performance was evaluated. The probe exhibits ratiometric sensing of ONOO− in a range of 0–6 µM. There is good linear relationship between the probe fluorescence intensity ratio and ONOO− concentration. The probe also displays moderate selectivity towards ONOO− over other ROS. Moreover, it is water-soluble and possesses good biocompatibility which aids the imaging of ONOO− in living cells. These properties could make the probe a promising tool in in vitro study related to ONOO−.

We report a new fluorescent probe (Myco-Cys) for live cell cysteine imaging. Myco-Cys contains a methacrylate cleavable pendant group that preferentially reacts with cysteine over homocysteine and glutathione to afford a “turn-on” response.
