The objective of this study was to investigate the adsorption/desorption behavior of oleuropein on different types of silk fibroin matrices including silk fibroin microfibers (MF), regenerated silk fibroin (RSF), and silk fibroin nanofibers (NF). Nanofibers with an average diameter of ranging between 24 and 326 nm were successfully prepared using the electrospinning technique. The effects of the silk fibroin concentration, the voltage applied and the distance between needle tip and collector plate on the morphology of the NF were investigated. The adsorption capacities of MF, RSF and NF were determined as 104.92, 163.07 and 228.34 mg oleuropein per gram of material, respectively. The percentage of initially adsorbed oleuropein that was desorbed was 86.08, 91.29 and 96.67% for MF, RSF and NF, respectively.NF and RSF discs loaded with oleuropein were subjected to disc diffusion assays to determine their antibacterial activity against test microorganisms Staphylococcus epidermidis (Gram +) and Escherichia coli (Gram – ). The results showed that both biomaterials possessed antibacterial properties after loading with oleuropein. Wound scratch assays using oleuropein released from NF revealed an enhancement of cell migration, indicating a wound healing property of the material.In conclusion, the NF can be utilized as a biofunctional polymeric material with better performance for the adsorption and desorption of oleuropein compared with MF and RSF.

Nonwoven nanofiber matrices were prepared by electrospinning a solution of silk fibroin (SF) dissolved either in formic acid or in 1,1,1,3,3,3-hexafluoro-2-isopropyl alcohol (HFIP). The mean diameter of the electrospun nanofibers prepared from SF dissolved in formic acid was 80 nm with a unimodal size distribution, which was smaller than those prepared from HFIP (380 nm). SF nanofibers were then treated with an aqueous methanol solution, and structural changes due to solvent-induced crystallization of SF were investigated using IR and 13C solid-state CP/MAS NMR spectroscopy. SF nanofibers prepared from formic acid were found to have a higher proportion of β-sheet conformations than those prepared from HFIP. Methanol treatment provided a fast and effective means to alter the secondary structure of both types of SF nanofibers from a random coil form to a β-sheet form. As demonstrated in the present study, this approach to controlling the dimensions and secondary structure of proteins using various solvents may be useful for the design and tailoring of materials for biomedical applications, especially for tissue engineering applications.

Regenerated silk fibroin (SF) mats were fabricated using electrospinning technique, followed by mild water vapor annealing to effectively tune the structures and improve the mechanical properties of the mats at different annealing times and temperatures. The breaking strength and the breaking energy of the mats treated with water vapor at 65°C for 12 h reached 6.0 MPa and 171.7 J/kg, respectively. The conformational transition of the SF mats was significantly influenced by the treating temperature, while the influence of time was comparatively limited. The influence is consistent with the time-temperature equivalent principle and would be helpful for the preparation of water-vapor-annealed silk-based biomaterials for various applications.

In recent years, electrospinning has received much attention to prepare porous tubular scaffold due to electrospun nanofibers mat, which can mimic the structure characteristics of the extracellular matrix (ECM). In the present paper, silk fibroin (SF) tubular scaffolds with small-diameter were prepared by electrospinning from regenerated silk fibroin (RSF) aqueous solutions with rotating mandrel collector. The morphology and fiber diameter distribution of SF tubular scaffolds were influenced much by the concentration of regenerated silk fibroin (RSF) solution, collecting distance and mandrel diameter. The results showed that SF tubular scaffold, which was in flat surface with smaller fiber diameter and uniform distributed, could be obtained at the concentration of 22%, collection distance of 10 cm and mandrel diameter of 4 mm. The structure of SF tubular scaffolds before and after ethanol treatment was characterized by FTIR and XRD. After ethanol treatment, the SF tubular scaffolds had a conformation transition from random-coil conformation to Silk II conformation.

AbstractElectrospinning is a common and versatile process to produce nanofibers and deposit them on a collector as a two-dimensional nanofiber mat or a three-dimensional (3D) macroscopic arrangement. However, 3D electroconductive collectors with complex geometries, including protruded, curved, and recessed regions, generally caused hampering of a conformal deposition and incomplete covering of electrospun nanofibers. In this study, we suggested a conformal fabrication of an electrospun nanofiber mat on a 3D ear cartilage-shaped hydrogel collector based on hydrogel-assisted electrospinning. To relieve the influence of the complex geometries, we flattened the protruded parts of the 3D ear cartilage-shaped hydrogel collector by exploiting the flexibility of the hydrogel. We found that the suggested fabrication technique could significantly decrease an unevenly focused electric field, caused by the complex geometries of the 3D collector, by alleviating the standard deviation by more than 70% through numerical simulation. Furthermore, it was experimentally confirmed that an electrospun nanofiber mat conformally covered the flattened hydrogel collector with a uniform thickness, which was not achieved with the original hydrogel collector. Given that this study established the conformal electrospinning technique on 3D electroconductive collectors, it will contribute to various studies related to electrospinning, including tissue engineering, drug/cell delivery, environmental filter, and clothing.
