This communication describes the synthesis of a triple acid-functionalized RAFT agent and its use to prepare well-defined 3-arm star polymers of N,N-dimethylacrylamide (DMAc).

Various pathways to generate star polymers using reversible addition–fragmentation transfer (RAFT) are discussed. Similar to other polymerization techniques, star polymers can be generated using arm-first and core-first approaches. Unique to the RAFT process is the subdivision of the core-first approach into the R-group and Z-group approaches, depending on the attachment of the RAFT agent to the multifunctional core. The mechanism of the R- and Z-group approaches are discussed in detail and it is shown that both techniques have to overcome difficulties arising from termination reactions. Termination reactions were found to broaden the molecular weight. However, these side reactions can be limited by careful design of the synthesis. Considerations include RAFT and radical concentration, number of arms, type of RAFT agent and monomer. Despite obvious challenges, the RAFT process is highly versatile, allowing the synthesis of novel polymer architectures such as poly(vinyl acetate) and poly(vinyl pyrrolidone) star polymers.

Polymers with a single central point of carboxylic acid functionality were prepared by living radical polymerization methods, RAFT and ATRP. A convenient water-based synthesis of a Y-branched ATRP initiator from 3,5-dihydroxybenzoic acid and 2-bromopropionyl bromide, from which the Y-branched RAFT agent is then subsequently derived, is described. Polymerization occurred uniformly from both of the RAFT groups to give chains of equal length as shown by hydrolysis. ATRP polymerization based on an ester derivative of 3,5-bis(2-bromopropionyloxy)benzoic acid as initiator was well controlled, whereas the free carboxylic acid gave inconsistent performance. The ability to couple functional molecules to the middle of polymers would provide better protection or interaction of the functional molecule with the polymer than conventional end attachment. This would find applications such as in drug delivery where more efficient protection would allow the use of lower molecular weight polymers.

We present a new family of porphyrin-functionalized coordination star polymers prepared through combination of coordination-driven self-assembly and post-assembly polymerization. Their self-assembly behaviour in water and potential for photodynamic therapy were demonstrated.

SummaryBlood was injected into the brains of dogs to produce artificial haematomas, and paraffin injected to produce intracerebral paraffin masses. Cerebrospinal fluid (CSF) and peripheral blood samples were withdrawn at regular intervals and their fibrinolytic activities estimated by the fibrin plate method. Trans-form aminomethylcyclohexane-carboxylic acid (t-AMCHA) was administered to some individuals. Genera] relationships were found between changes in CSF fibrinolytic activity, area of tissue damage and survival time. t-AMCHA was clearly beneficial to those animals given a programme of administration. Tissue activator was extracted from the brain tissue after death or sacrifice for haematoma examination. The possible role of tissue activator in relation to haematoma development, and clinical implications of the results, are discussed.
