A water soluble and biocompatible glucopyranosyl conjugate (L) has been synthesized, characterized and shown to be selective to Cys among the naturally occurring amino acids. TheLshows green fluorescence upon reaction with –SH containing molecules present in biological cells and hence provides cell imaging.

Green fluorescent, pH dependent and water soluble copper nanoclusters (3–5 nm) were synthesized by stabilizing with lysozyme and these were demonstrated for imaging in both healthy and cancer cell lines as an alternate to green fluorescent protein.

Molecular conformation as a subproblem of the geometrical shaping of the molecules is essential for the expression of biological activity. It is well known that from the series of all possible sugars, those that are most naturally occurring and usable by living organisms as a source of energy—because they can be phosphorylated by hexokinase, the first enzyme in the glycolysis pathway—are D-sugars (from the Latin dextro). Furthermore, the most naturally occurring amino acids in living cells are L-sugars (from the Latin laevo). However, a problem arises in dealing with the comparison of their conformers. One alternative way to compare sugars is via their molecular alignment. Here, a solution to the eigenproblem of molecular alignment is communicated. The Cartesian system is rotated, and eventually translated and reflected until the molecule arrives in a position characterized by the highest absolute values of the eigenvalues observed on the Cartesian coordinates. The rotation alone can provide eight alternate positions relative to the reflexes of each coordinate.

A water-soluble fluorescent probe for Zn2+based on a Schiff base derivative was prepared. It displays a rapid response toward Zn2+with selectivity and sensitivity and has been successfully applied to imaging in living cells.

Background: Morin has many pharmacological functions including antioxidant, anticancer, anti-inflammatory, and antibacterial effects. It is commonly used in the treatment of antiviral infection, gastropathy, coronary heart disease and hepatitis B in clinic. However, researches have shown that morin is likely to show prooxidative effects on the cells when the amount of treatment is at high dose, leading to the decrease of intracellular ATP levels and the increase of necrosis process. Therefore, it is necessary to determine the concentration of morin in biologic samples. Method: Novel water-soluble and green nitrogen and sulfur co-doped carbon dots (NSCDs) were prepared by a microwave heating process with citric acid and L-cysteine. The fluorescence spectra were collected at an excitation wavelength of 350 nm when solutions of NSCDs were mixed with various concentrations of morin. Results: The as-prepared NSCDs were characterized by transmission electron microscopy, X-ray diffraction and X-ray photoelectron spectroscopy. The fluorescence intensity of NSCDs decreased significantly with the increase of morin concentration. The fluorescence intensity of NSCDs displayed a linear response to morin in the concentration 0.10-30 μM with a low detection limit of 56 nM. The proposed fluorescent probe was applied to analysis of morin in human body fluids with recoveries of 98.0-102%. Conclusion: NSCDs were prepared by a microwave heating process. The present analytical method is sensitive to morin. The quenching process between NSCDs and morin is attributed to the static quenching. In addition, the cellular toxicity on HeLa cells indicated that the as-prepared NSCDs fluorescent probe does not show obvious cytotoxicity in cell imaging. Our proposed method possibly opens up a rapid and nontoxic way for preparing heteroatom doped carbon dots with a broad application prospect.

AbstractCyanophycin (multi-l-arginyl-poly-l-aspartic acid; also known as cyanophycin grana peptide [CGP]) is a biopolymer that could be used in various fields, for example, as a potential precursor for the synthesis of polyaspartic acid or for the production of CGP-derived dipeptides. To extend the applications of this polymer, it is therefore of interest to synthesize CGP with different compositions. A recent re-evaluation of the CGP synthesis in C. glutamicum has shown that C. glutamicum is a potentially interesting microorganism for CGP synthesis with a high content of alternative amino acids. This study shows that the amount of alternative amino acids can be increased by using mutants of C. glutamicum with altered amino acid biosynthesis. With the DM1729 mutant, the lysine content in the polymer could be increased up to 33.5 mol%. Furthermore, an ornithine content of up to 12.6 mol% was achieved with ORN2(Pgdh4). How much water-soluble or insoluble CGP is synthesized is strongly related to the used cyanophycin synthetase. CphADh synthesizes soluble CGP exclusively. However, soluble CGP could also be isolated from cells expressing CphA6308Δ1 or CphA6308Δ1_C595S in addition to insoluble CGP in all examined strains. The point mutation in CphA6308Δ1_C595S partially resulted in a higher lysine content. In addition, the CGP content could be increased to 36% of the cell dry weight under optimizing growth conditions in C. glutamicum ATCC13032. All known alternative major amino acids for CGP synthesis (lysine, ornithine, citrulline, and glutamic acid) could be incorporated into CGP in C. glutamicum.
