A new colorimetric assay for the detection of sulfide anions with high sensitivity and selectivity is reported, utilizing Au-Hg alloy nanorods (Au-HgNRs) as probe. Au-HgNRs were prepared by modifying gold nanorods (AuNRs) with reducing agent and mercury ions. In an aqueous solution with sulfide anions, the formation of mercuric sulfide due to redox reaction between the amalgams and sulfide anions greatly changed the surface chemistry and morphology of the Au-HgNRs, leading to a red shift of the localized surface plasmon resonance (LSPR) absorption peak, accompanied by a change in colorimetric response. A good linear relationship was obtained between the LSPR peak wavelength shift and concentration of sulfide anion in the range of 1 × 10−5−1 × 10−4 mol/L. The selectivity of this method has been investigated by other anions. The colorimetric sensing system successfully detected sulfide in wastewater from leather industry.

A simple and sensitive assay for Ag+ was developed with unmodified gold nanoparticles (AuNPs) by using dynamic light scattering techniques. Ag+ could induce the oligonucleotide (5′-ATC ACT ATA TCA TAT ACT CAT-3′) to change from a single-stranded structure to a double-stranded structure and desorb from the surface of AuNPs, which triggered the aggregation of AuNPs in the salt solution. The average hydrodynamic diameter of aggregated AuNPs could be detected by using dynamic light scattering techniques. Under the optimum conditions, the average hydrodynamic diameter of AuNPs is proportional to the concentration of Ag+ within the range of 13.3–100.0 nmol/L, with a detection limit of 3.2 nmol/L. The method is easy to operate and has low sample consumption, high sensitivity and selectivity.

In this study, trace arsenic(iii) (AsIII) in aqueous solution was detected by applying a classical aptamer-based gold nanoparticles colorimetric sensing strategy. An arsenic aptamer was used as a sensing probe and gold nanoparticles as a colorimetric indicator. In the absence of AsIII, the gold nanoparticles were stabilised by the arsenic aptamer and remained dispersed at high NaCl concentrations, displaying a red solution. Contrarily, in the presence of AsIII, the gold nanoparticles were prone to aggregation, owing to the formation of aptamer–AsIII complex between the arsenic aptamer and AsIII, and thus exhibited a blue solution. By monitoring the colour change, a simple and fast colorimetric assay for AsIII was established with a detection range of 1.26–200 ppb and a detection limit of 1.26 ppb. Because this colorimetric assay only involves common reagents and can be assessed visually, it holds great potential for arsenic(iii) monitoring in environment-related and other applications.

We report a label-free, fast, fluorescence turn on assay for Hg2+detecton by using mercury-specific DNA (MSD), Sybr Green I (SG) and gold nanoparticles (AuNPs). SG efficiently discriminates MSD and MSD/Hg2+complex. The addition of gold nanoparticle decreases the background fluorescence signal further for MSD. The fluorescence intensity of MSD/Hg2+complex keeps constant after addition of AuNPs. This property improves the signal-to-background ratio and decreases the detection limitation further. In addition, the method shows improved selectivity compared with that in the absence of AuNPs. This strategy could be applied to the detection of potassium ions and showed good generality.

Digital microfluidic (DMF) has been a unique tool for manipulating micro-droplets with high flexibility and accuracy. To extend the application of DMF for automatic and in-site detection, it is promising to introduce colorimetric sensing based on gold nanoparticles (AuNPs), which have advantages including high sensitivity, label-free, biocompatibility, and easy surface modification. However, there is still a lack of studies for investigating the movement and stability of AuNPs for in-site detection on the electrowetting-based digital microfluidics. Herein, to demonstrate the ability of DMF for colorimetric sensing with AuNPs, we investigated the electrowetting property of the AuNPs droplets on the hydrophobic interface of the DMF chip and examined the stability of the AuNPs on DMF as well as the influence of evaporation to the colorimetric sensing. As a result, we found that the electrowetting of AuNPs fits to a modified Young–Lippmann equation, which suggests that a higher voltage is required to actuate AuNPs droplets compared with actuating water droplets. Moreover, the stability of AuNPs was maintained during the processing of electrowetting. We also proved that the evaporation of droplets has a limited influence on the detections that last several minutes. Finally, a model experiment for the detection of Hg2+ was carried out with similar results to the detections in bulk solution. The proposed method can be further extended to a wide range of AuNPs-based detection for label-free, automatic, and low-cost detection of small molecules, biomarkers, and metal ions.

In this study, we have used new approach for detection of the methyl parathion residues i.e., by inhibiting half of the peroxidase-like activity of gold nanoparticles. We have investigated the peroxidase like activity of gold nanoparticles by colorimetric assay and optimized the pH, temperature, incubation time and different concentrations of H2O2 by using TMB as substrate as well as organophosphates effect on their enzyme mimetic activity. Kinetic study of gold nanoparticles has shown better catalytic activity than horseradish peroxidase at pH 3.5. The peroxidase substrate TMB (3,3,5,5-tetramethylbenzidine) can be oxidized by H2O2 by the enzymatic action of the gold nanoparticles resulting in a blue-coloured product, oxidized TMB. The principle involved behind the inhibition of enzymatic activity of nanozyme is due to hindrance of electron transfer mechanism TMB-H2O2-Au NPs system by the methyl parathion. The peroxidase activity is selectively reduced with increasing methyl parathion concentration. This sensing method has lowest limit of detection of 78.95 nM. This study can be used for development of sensitive and cost effective technique for sensing of harmful pesticides.

In this work, stable gold nanoparticles (AuNPs) were prepared with good compatibility and favorable catalytic activity. They were developed as a peroxidase mimic for colorimetric detection of cysteine with a weakened blue solution, which supplied high sensitivity and selectivity.
