Hyperpolarization turns typically weak NMR and MRI responses into strong signals so that ordinarily impractical measurements become possible. The potential to revolutionize analytical NMR and clinical diagnosis through this approach reflect this area's most compelling outcomes. Methods to optimize the low-cost parahydrogen-based approach signal amplification by reversible exchange with studies on a series of biologically relevant nicotinamides and methyl nicotinates are detailed. These procedures involve specific 2H labeling in both the agent and catalyst and achieve polarization lifetimes of ca. 2 min with 50% polarization in the case of methyl-4,6-d2-nicotinate. Because a 1.5-T hospital scanner has an effective 1H polarization level of just 0.0005% this strategy should result in compressed detection times for chemically discerning measurements that probe disease. To demonstrate this technique’s generality, we exemplify further studies on a range of pyridazine, pyrimidine, pyrazine, and isonicotinamide analogs that feature as building blocks in biochemistry and many disease-treating drugs.

The N–O heterocycles are biologically relevant scaffolds and versatile building blocks in contemporary organic synthesis. In this short review, we effort to showcase the involvement and elevation of various cycloaddition strategies towards the production of N–O heterocycles 1,2-oxazines, 1,2-oxazepanes, and 1,2-oxazetidines. A blueprint of advantages and challenges associated with these synthetic endeavors is provided.

In recent times, many biologically relevant building blocks such as amino acids, peptides, saccharides, nucleotides and nucleosides, etc. have been prepared by mechanochemical synthesis. However, mechanosynthesis of lipids by ball milling techniques has remained essentially unexplored. In this work, a multistep synthetic route to access mono- and diacylglycerol derivatives by mechanochemistry has been realized, including the synthesis of diacylglycerol-coumarin conjugates.

AbstractA series of astronomical observations obtained over the period 1986 to 2018 supports the idea that life is a cosmic rather than a purely terrestrial or planetary phenomenon. These include (1) the detection of biologically relevant molecules in interstellar clouds and in comets, (2) mid-infrared spectra of interstellar grains and the dust from comets, (3) a diverse set of data from comets including the Rosetta mission showing consistency with biology and (4) the frequency of Earth-like or habitable planets in the Galaxy. We argue that the conjunction of all the available data suggests the operation of cometary biology and interstellar panspermia rather than the much weaker hypothesis of comets being only the source of the chemical building blocks of life. We conclude with specific predictions on the properties expected of extra-terrestrial life if it is discovered on Enceladus, Europa or beyond. A radically different biochemistry elsewhere can be considered as a falsification of the theory of interstellar panspermia.

We discovered a new retro-Mannich reaction of in situ prepared pyrazolopyridines to give pyrazolopyrimidines that have hitherto been underrepresented in the heterocyclic chemistry literature. The isolation of a linear hydrolysis product supports a mechanistic hypothesis for this rearrangement process. In order to establish a broader access and explore potential biological applications for these medicinal chemistry building blocks, we investigated the scope of the reaction and generated small amine- as well as amide-based libraries through reductive aminations and amide couplings, respectively.

An accurate prediction of the molecular acidity by employing ab initio or density functional approaches for typical molecular systems is still challenging. Recently, we proposed to utilize two quantum descriptors, molecular electrostatic potential (MEP) and the sum of valence natural atomic orbital (NAO) energies on the nucleus of both the acidic atom and leaving proton, to quantitatively evaluate the pKa values. This new approach has been validated by a number of organic and inorganic systems and justified within the framework of density functional reactivity theory (DFRT). In this work, we apply the approach to building blocks of biological systems, namely, 20 natural α-amino acids and 5 DNA/RNA bases, together with a few other biologically relevant species. Our results show that there exists a strong linear correlation between MEP on the nucleus of the N atom and the sum of N 2p NAO energies, with the correlation coefficient R2 = 0.99. Also, we observe that both MEP on the nitrogen nucleus and the sum of N 2p NAO energies correlate well with experimental pKa values, with the correlation coefficient equal to 0.91. Using this established model, we predicted the trend of pKa changes of amino acids in proteins with different dielectric constants. We also applied the model to predict pKa values for dipeptides. Implications of these linear relationships to understand functions and reactivity of biological systems are discussed as well.

Phthalimides have found their main application in organic synthesis as protecting groups for primary amines during the multistep synthesis of biologically relevant targets. On the other hand, phthalimide functionalization is rather challenging and it is traditionally associated with the use of over-stoichiometric amounts of environmentally hazardous reagents.
