Митотическая нестабильность кольцевых хромосом может приводить к появлению клеточных клонов с различной генетической структурой. В качестве модели нестабильности кольцевых хромосом в митозе мы использовали фибробласты от пациентов с r(8), r(13), r(18) и r(22) и полученные из них индуцированные плюрипотентные стволовые клетки (ИПСК). Линии ИПСК с r(22) имели относительно стабильный кариотип на протяжении десятков (до 60) пассажей и сохраняли неизменную структуру кольцевой хромосомы. Кариотип линий ИПСК с r(8) и r(18) на ранних пассажах стабильный, планируется его изучение на поздних пассажах. Наибольшее разнообразие кариотипа выявлено в линиях ИПСК с r(13), в которых наблюдали различные перестройки и выраженную клеточную гетерогенность. Определение факторов, влияющих на митотическую стабильность кольцевых хромосом, может иметь значение для консультирования пациентов. Mitotic instability of ring chromosomes can lead to the appearance of cell clones with different genetic structure. IPSCs from fibroblasts of patients with r(8), r(13), r(18), and r(22) were used as a model of ring chromosomes mitotic behavior. Karyotypes of iPSC lines with r(8) and r(18) have so far been evaluated only in the early passages, lines with r(22) have maintained a relatively stable karyotype up to 60 passages. The occurrence of rearrangements and cellular heterogeneity was found characteristic for r(13) iPSCs. The determination of factors affecting the ring chromosomes mitotic stability would be beneficial for the patient’s prognosis.

Background: Herbicides are very beneficial in the crop yield with the aid of controlling weeds within the agriculture, but several herbicides are chronic in soil. Objective: In this study, nanoparticles and the packages of synthesized novel silica nanoparticles were studied for the preconcentration of herbicides. Methods: These nanoparticles prepared by the Stöber mechanism were purified and functionalized. Nanoparticles thus prepared successfully were used as supporting material for the preconcentration of residues of herbicides in the water. Results: Preconcentration was achieved by preparing the silica-based solid-phase-extraction cartridges. Nanoparticles used for this purpose were within the range of 50-250 nm. An SPE cartridge was prepared by packing 200 mg of silica nanoparticle in the empty cartridge of diameter 5.5 cm and length 0.6 cm in between PTFE frits. Aqueous solutions of 0.1 μg/ml of herbicides were prepared separately, and 10 ml of the solution was passed through the cartridge at the rate of 0.2 ml/min. After passing 10 ml volume of the aqueous solution, residues adsorbed on the cartridge were eluted using 2 ml of acetonitrile. The eluate was injected to determine the herbicide residue adsorbed on the SPE cartridge. Conclusion: In the study, it was found that greater than 90% of the herbicide residues were trapped on silica nanoparticle-based SPE cartridge. An analytical method was developed for the simultaneous determination of these herbicides. The residues were quantified by LC-MS/MS with ESI mode.

The procedure for the determination of carbamate pesticides in soil was optimized. The following factors affecting the final results were investigated: extracting solvent, extraction procedure, extract purification procedure, and soil type. Triple extraction with acetone and purification of the extract on a two-stage purification column containing an activated carbon-silica gel 1+1 mixture were found optimal. The extracts after treatment were analyzed by RP-HPLC with UV detection. The method developed allows carbamate pesticides in soil to be determined at concentrations in excess of 30 μg kg-1.

It has been found that phosphoric acid decreases the first excitation maximum of Ce(III) at 256 nm, increases the second excitation maximum at 297 nm and shifts the fluorescence maximum from 350 to 346 nm. Under optimum conditions, with λexc = 297 nm and λem = 346 nm, Ce(III) can be determined fluorimetrically with a detection limit of 1.2 ng ml-1 in 12M-H3PO4 medium. No interference was observed from a 20-200 fold excess of HCl, H2SO4, Na, K, NH4+, Al and the rare earth elements. HNO3 interferes and Ce(IV) and Fe(III) interfere strongly. It follows from the stereofluorograms of Ce and Tb that the spectra of the two elements are practically independent. The detection limit for Tb(III) in 0.02-2.5M-H2SO4 medium for λexc = 222 nm and λem = 494 nm is 33 ng ml-1. No interference was observed from a 5-20 fold excess of Al3+ and the other rare earth elements. The determination is slightly less sensitive in H3PO4 or HCl medium. The relative standard deviation of the measurement for 10 ng ml-1 Ce(III) or 50 ng ml-1 Tb(III) is about 3%.

Abstract A procedure for the enzymatic determination of lecithin and sphingomyelin in aqueous solution is described. The phospholipids are first dissolved in chloroform:methanol (2:1 by vol), the solvent is evaporated, and the residue is redissolved in an aqueous zwitterionic detergent solution. The enzymatic reaction sequences of both assays involve hydrolysis of the phospholipids to produce choline, which is then oxidized to betaine, thus generating hydrogen peroxide. The hydrogen peroxide is subsequently utilized in the enzymatic coupling of 4-aminoantipyrine and sodium 2-hydroxy-3,5-dichlorobenzenesulfonate, an intensely red color being formed. The presence of a non-reacting phospholipid enhances the hydrolysis of the reacting phospholipid. Thus we added lecithin to the sphingomyelin standards and sphingomyelin to the lecithin standards. This precise procedure may be applicable to determination of lecithin and sphingomyelin in amniotic fluid.
