In birds and higher mammals, auditory experience during development is critical to discriminate sound patterns in adulthood. However, the neural and molecular nature of this acquired ability remains elusive. In fruit flies, acoustic perception has been thought to be innate. Here we report, surprisingly, that auditory experience of a species-specific courtship song in developing Drosophila shapes adult song perception and resultant sexual behavior. Preferences in the song-response behaviors of both males and females were tuned by social acoustic exposure during development. We examined the molecular and cellular determinants of this social acoustic learning and found that GABA signaling acting on the GABAA receptor Rdl in the pC1 neurons, the integration node for courtship stimuli, regulated auditory tuning and sexual behavior. These findings demonstrate that maturation of auditory perception in flies is unexpectedly plastic and is acquired socially, providing a model to investigate how song learning regulates mating preference in insects.

Large-conductance Ca2+- and voltage-activated K+ channel (BK) open probability is enhanced by depolarization, increasing Ca2+ concentration, or both. These stimuli activate modular voltage and Ca2+ sensors that are allosterically coupled to channel gating. Here, we report a point mutation of a phenylalanine (F380A) in the S6 transmembrane helix that, in the absence of internal Ca2+, profoundly hinders channel opening while showing only minor effects on the voltage sensor active–resting equilibrium. Interpretation of these results using an allosteric model suggests that the F380A mutation greatly increases the free energy difference between open and closed states and uncouples Ca2+ binding from voltage sensor activation and voltage sensor activation from channel opening. However, the presence of a bulky and more hydrophobic amino acid in the F380 position (F380W) increases the intrinsic open–closed equilibrium, weakening the coupling between both sensors with the pore domain. Based on these functional experiments and molecular dynamics simulations, we propose that F380 interacts with another S6 hydrophobic residue (L377) in contiguous subunits. This pair forms a hydrophobic ring important in determining the open–closed equilibrium and, like an integration node, participates in the communication between sensors and between the sensors and pore. Moreover, because of its effects on open probabilities, the F380A mutant can be used for detailed voltage sensor experiments in the presence of permeant cations.

The main goal of this work is to perform a first-time analysis of the thermal cross-coupling in a system composed by some devices in an integration node degree composed by advanced UTBB SOI MOSFETs through numerical simulations, validated with experimental data from the literature. In this analysis, it could be observed that devices located on the channel length direction provoke a reduced thermal coupling and devices with their drain region next to each other suffer of an increased thermal coupling due to the lumped thermal energy. It also could be observed a degradation in some electrical parameters and in the thermal properties of a device under the influence of surrounded devices biased.

AbstractZNF423 encodes 30 C2H2 zinc fingers that bind DNA and a variety of lineage- and signal-dependent transcription factors. ZNF423 genetic variants are proposed to cause neurodevelopmental and ciliopathy-related disorders in humans. Mouse models show midline brain defects, including cerebellar vermis hypoplasia, and defects in adipogenesis. Here I show strong protein sequence constraint among 165 vertebrate orthologs. In contrast, orthologs from invertebrate lineages, spanning larger time intervals, show substantial differences in zinc finger number, arrangement, and identity. A terminal zinc finger cluster common among other lineages was independently lost in vertebrates and insects. Surprisingly, a moderately-constrained non-C2H2 sequence with potential to form a C4-class zinc finger is a previously-unrecognized conserved feature of nearly all identified homologs. These results highlight evolutionary dynamics of a likely signal integration node across species with distinct developmental strategies and body plans. Functions of the newly identified C4-like sequence and lineage-specific fingers remain to be studied.

A competent methodology based on the active power loss reduction for optimal placement and sizing of distributed generators (DGs) in an active distribution network (ADN) with several soft open points (SOPs) is proposed. A series of SOP combinations are explored to generate different network structures and they are utilized in the optimization framework to identify the possible solutions with minimum power loss under normal network conditions. Furthermore, a generalized methodology to optimize the size and the location of a predefined number of DGs with a predefined number of SOPs is presented. A case study on the modified IEEE 33 bus system with three DGs and five SOPs was conducted and hence the overall network power loss and the voltage improvement were examined. The findings reveal that the system loss of the passive network without SOPs and DGs is reduced by 79.5% using three DGs and five SOPs. In addition, this research work introduces a framework using the DG size and the impedance to the DG integration node, to propose a region where the DGs can be optimally integrated into an ADN that includes several SOPs.

Light and brassinosteroid (BR) are master environmental stimulus and endogenous cue for plant growth and development respectively. Great progress has been made in elucidating the molecular mechanisms on the cross-talk between light and BR. However, little is known about how BZR1, the pivotal integration node, is regulated by light and dark. Here, we demonstrated that an intact BR signaling pathway is essential for dark-induced hypocotyl elongation. Consequent expression assay showed that light–dark switch affected BZR1 phosphorylation and accumulation. Moreover, blocking the 26S proteasome pathway promoted the accumulation of both phosphorylated and dephosphorylated BZR1 proteins. Restriction of new protein biosynthesis had multiple effects on BZR1 phosphorylation status and stability, relying on the availability of light and the 26S proteasome pathways. Furthermore, sugar treatment strikingly enhanced the accumulation of total BZR1 under either light or dark conditions, likely by repressing transcript abundance of MAX2, a gene encoding an E3 ligase for BZR1. Finally, light-regulated phosphorylation change of BZR1 requires the existence of endogenous BR as well as functional BIN2 and protein phosphatase 2A (PP2A). Taken together, our results depicted a light-involved complex regulation network of BZR1 stability and phosphorylation status.
