AbstractObjectivesEvaluate the influence of the genetic variant rs9939609 of the FTO gene on anthropometric characteristics and whether parental obesity is related to children and adolescents being overweight.MethodsA total of 2,364 children and adolescents between 6 and 17 years old were genotyped and the lipid profile, plasma glucose level, and anthropometric characteristics were measured to assess adiposity.ResultsThe AA genotype (risk) was associated with higher body mass index (BMI Z-score; p = 0.006), waist circumference (WC; p = 0.001), and triglycerides (p = 0.033). The association of the participants’ adiposity characteristics with the parents’ BMI and FTO genotypes showed an association of the BMI Z-score when either the mother or father was overweight or obese (p = 0.028 and p = 0.029). In the overweight or obese father/eutrophic mother, we also observe an association of FTO rs9939609 with WC (p = 0.039). The effect of these variables on the risk of obesity was also tested: overweight or obese mother (OR = 1.82, p = 0.041), overweight and obese parents (OR = 3.09, p < 0.0001), and FTO rs9939609 AA genotype (OR = 2.08, p = 0.0004) were associated. With regard to altered WC and high body fat percentage (BF%), either overweight or obese parents (OR = 2.39, p < 0.0001; OR = 1.92, p < 0.002) showed an association. The FTO rs9939609 AA genotype (OR = 1.99, p = 0.0002) was associated with altered WC.ConclusionsThe results show that parental weight also contributes to obesity and may interact with the FTO genetic make-up.

Motivation/Background: In 2001, non-communicable diseases (NCDs) accounted for 60 percent of 56 million deaths worldwide and 47% of global burden of disease. In all countries, evidence suggested underlying determinants of NCDs were largely same. These include increased consumption of rapidly processed energy-dense nutrient-poor foods high in fat, sugar, salt and reduced levels of physical activity. The objective of this study was to determine prevalence, pattern and determinants of fast foods consumption among school age children in Jos metropolis. Method: In this study, we used descriptive cross-sectional study and included 347 mothers or care givers with children age 6 to 12 years in our sample size. We applied structured interviewer-administered questionnaire during data collection and analyzed the data with SPSS version 21.0. Results: Prevalence of fast food consumption among school age children in Jos metropolis in the last one week is 91.1%. Knowledge of health risk associated with fast food consumption was 86.5%. Flour based fast food were most commonly consumed. We established an association between meal skipping, fast food advertisement and fast food consumption. Conclusions: We recommend the need to focus public enlightenment on health risk associated with fast food consumption among school age children. Likewise, children should be encouraged to carry home-made food to school and legislation on fast food advertisement directed at children should be enacted.

Objective: A common "obesity-risk" variant rs9939609 in the FTO gene was recently found to affect appetite, and the gene is sensitive to regulation of amino acids. We examined the interaction between FTO genotype and protein intake on the long-term changes in appetite in a randomized controlled trial. Research Design and Methods: We genotyped FTO rs9939609 in 737 overweight adults in the 2-year Preventing Overweight Using Novel Dietary Strategies (Pounds Lost) trial and assessed four appetite related traits including cravings, fullness, hunger, and prospective consumption. Results: We found that dietary protein significantly modified genetic effects on changes in food cravings at 6 months, after adjustment for age, sex, ethnicity, baseline body mass index, weight change, baseline value for food cravings (P for interaction=0.027). The A allele was associated with a greater decrease in food cravings among the participants with high protein diet intake (P=0.027), but not in those low protein diet group (P= 0.384). Weight regain from 6 months to 24 months attenuated the gene-protein interactions. Protein intakes did not modify the FTO genotype effects on other appetite measures. Conclusions: Our data suggest that individuals with the FTO rs9939609 A allele might obtain more benefits in reduction of food cravings by choosing a hypocaloric and higher protein weight-loss diet.
