SUMMARY Higher temperatures allow lower sulphuric acid concentrations and shorter heating times to be used in the Kober colour reaction for oestrogens. A one-stage reaction which is completed in 5 min. at 120° is described for oestrone and oestradiol, and a two-stage reaction which requires two periods of heating for 5 min. at 120° is described for oestriol. The conditions were applied to the Ittrich fluorescence procedure. A spectrophotofluorimetric correction was developed in which fluorescence was measured at wavelengths for excitation and emitted light near the optima for the oestrogens and at another combination at which the oestrogens produced virtually no fluorescence whereas that of impurities was not diminished. Extraction, centrifugation and fluorimetry were performed in specially designed cells. The sensitivity is 0·05–0·1 ng./sample with a linear response up to 300 ng. and a precision better than 4% in the range 1·0–100 ng.

SUMMARY 3α-, 3β-, 11β-, 16β-, 17β- and 20β-hydroxysteroid dehydrogenases have been localized histochemically in the Leydig cells of prepubertal and adult human testes; 3α-, 16β- and 17β-hydroxysteroid dehydrogenases were present in the seminiferous tubules also. A similar pattern was found in cryptorchid testes. In addition 3β-sulphoxy steroids, including DHA sulphate, gave a good colour reaction in human Leydig cells. Testes from oestrogen-treated subjects had no histochemically demonstrable hydroxysteroid dehydrogenases and this applied also to infarcted testes. Testes from a case of Klinefelter's syndrome were found to lack 17β- and 20β-hydroxysteroid dehydrogenases in the Leydig cells. The biochemical significance of these results is discussed.

After diazotation 2-amino-3-hydroxythiazolium chlorides react with activated aromatic compounds to build azo dyes. In absence of coupling components 2-amino-3-hydroxy-4-methylthiazolium chloride gives a colour reaction with nitrite ions, which is suitable for photometric detection of nitrite in aqueous solution.
